2 VIII Zusammenfassung Für die gaschromatographische Analyse von Aldosen wird häufig die Derivatisierung zu Aldonitrilacetaten gewählt. Anhand der Monosaccharide Rhamnose, Arabinose, Xylose, Mannose, Glucose und Galactose wurde mittels HPLC gezeigt, dass nach der Oximbildung mit Hydroxylamin und anschliessender Acetylierung mit 1-Methylimidazol als Lösungsmittel und Katalysator nicht als einziges Produkt jeweils die Aldonitrilacetat-Derivate entstehen, sondern dass auch Nebenprodukte in Form von Oximester- und Furanosederivaten gebildet werden. Auch bei der Derivatisierung mit ande ren Katalysatoren wie 4-(Dimefriylamino)pyndin oder 1-Dimethylamino-2-propanol und Pyridin als Lösungsmittel wurde die Bildung dieser Oximester- und Furanosederivate beobachtet. Mit Lobar-Chromatographie wurden diese Nebenprodukte isoliert und mit 1H- und 13C-NMR-Spektroskopie charakterisiert. Bei der GC-Analyse wandeln sich die Oximesterderivate bei genügend hohen Injektortemperaturen mit einer Umwandlungsrate von 90 % reproduzierbar in die entsprechenden Aldonitrilacetatderivate um. Die Furanosederivate werden jeweils in reproduzierbarem Verhältnis gebildet und ihr Anteil beträgt bei allen untersuchten Monosacchariden zwischen 5 und 9 %. Die Resultate der vorliegenden Arbeit zeigen, dass trotz der Bildung dieser Neben produkte die Derivatisierung zu Aldonitrilacetaten zur Quantifizierung von Aldosen angewendet werden darf. Es wurden Bausteinanalysen der unlöslichen Zellwand-Polysaccharide von Kartoffeln und Karotten nach chemischer Hydrolyse und im Falle der Karotten auch nach enzymatischer Hydrolyse durchgeführt. Aus Kartoffeln der Sorte Sirtema wurde das Zellwandmaterial als nahezu stärkefreies Präparat mit einer Ausbeute von 0.5 % isoliert. Nach einer Saeman-Hydrolyse (Vorhydrolyse mit 12 M H2SO4 bei Raumtemperatur, Haupthydrolyse mit 1 M H2SO4 bei 100 C) wurden die Monosaccharide zu Aldonitrilacetaten derivatisiert und gaschromatographisch ein Gehalt von 70 % Neutralzuckem ermittelt. Ausserdem wurden der Pektin- (25 %), Protein- (1.6 %), Lignin- (0.5 %) und Aschegehalt (5 %) ermittelt.

3 IX Das Zellwandmaterial aus Karotten der Sorte Nantaise wurde als alkoholunlöslicher Rückstand (AIR) mit einer Ausbeute von 2.6 % isoliert. Der nach der Saeman-Hydrolyse ermittelte Neutralzuckergehalt ergab 49 %. Daneben wurden der Pektin- (31 %), Protein- (5 %), Lignin- (4 %) und Aschegehalt (8 %) des Zellwandmaterials bestimmt. Die für pflanzliches Zellwandprotein charakteristische Aminosäure Hydroxyprolin wur de in den Proteinen der Kartoffel- und Karottenzellwände zu 4.5 bzw. 2.5 % gefunden. Neben der Saeman-Hydrolyse wurde das Karotten-Zellwandmaterial auch enzymatisch mit den Handelspräparaten Pecönex Ultra SP-L, Celluclast, ß-Glucosidase und Driselase hydrolysiert. dieser Präparate erhalten. Der wirkungsvollste Abbau wurde mit einer Kombination Dabei konnten 92 % des Zellwandmaterials gelöst und im Vergleich zur Saeman-Hydrolyse eine Monomerenbildung von 80 % erzielt werden. Eine Analyse des nach der enzymatischen Hydrolyse anfallenden Rückstands ergab einen Neutralzuckeranteil von ca. einem Drittel, welcher sich zu 70 % aus D-Glucose zusammensetzte und damit auf einen unvollständigen enzymatischen Celluloseabbau hinweist. Mit der enzymatischen Hydrolyse allein wurde keine vollständige Monomerenbildung erreicht. Erst eine Kombination der enzymatischen Hydrolyse mit dem Haupt hydrolyseschritt der Saeman-Prozedur ergab eine mit der Saeman-Hydrolyse ver gleichbare Monosaccharidausbeute (95 % Monosaccharide im Vergleich zur Saeman- Hydrolyse) und -Zusammensetzung. Der Einfluss der Partikelgrösse auf die Ausbeute an Neutralzuckem nach enzymatischer bzw. Saeman-Hydrolyse wurde anhand von gemahlenem und ungemah lenem Zellwandmaterial abgeklärt. Dabei zeigte sich, dass mit der gemahlenen Probe der enzymatische Abbau, insbesondere im Falle der Cellulose, verbessert wurde.

4 X Summary The derivatization to aldonitrile acetates is a commonly employed method for the GC analysis of aldoses. With the monosaccharides rhamnose, arabinose, xylose, mannose, glucose and galactose it was shown by HPLC, that the formation of oximes with hydroxylamine followed by acetylation using 1-methylimidazole as solvent and catalyst yields not only the aldonitrile acetate derivatives but also by-products in the form of oxime esters and furanose derivatives. The formation of these oxime esters and furanose derivatives has also been observed during derivatization with other catalysts such as 4-(dimethylamino)pyridine or 1-dimethylamino-2-propanol and Pyridine as solvent. The by-products were isolated by Lobar-chromatography and characterized by 1H- and 13C- NMR spectroscopy. During GC analysis, when sufficient high injector temperatures are employed, the oxime ester Compounds are completely transformed into the corresponding aldonitrile acetate derivatives. The furanose derivatives are formed in reproducible ratios and they account for 5 to 9 % of each monosaccharide tested. The results of this work show that despite the formation of these by-products the aldonitrile acetate derivatization procedure may be used to quantify aldoses. Monosaccharide analysis of the insoluble Polysaccharides of the cell walls of potatoes and carrots was carried out afler Chemical hydrolysis. The carrot cell walls were also subjected to enzymatic hydrolysis. An almost starch-free cell wall material was isolated in 0.5 % yield from potatoes, cv. Sirtema. After Saeman hydrolysis (pre-hydrolysis with 12 M H2SO4 at room temperature and main hydrolysis using 1 M H2SO4 at 100 C) the monosaccharides obtained were derivatized to aldonitrile acetates. The neutral sugar content of the cell wall material was found to be 70 % as determined by gas chromatography. Pectin (25 %), protein (1.6 %), lignin (0.5 %) and ashes (5 %) were determined as well.

5 XI The cell wall material of carrots, cv. Nantaise, was isolated as the alcohol-insoluble residue (AIR); a 2.5 % yield was obtained. The neutral sugar content determined after Saeman hydrolysis was found to be 49 %. Pectin (31 %), protein (5 %), lignin (4 %) and ashes (8 %) were determined as well. The potato and carrot cell walls were found to contain respectively 4.5 % and 2.5 % hydroxyproline, which is the characteristic amino acid of the plant cell wall material. In addition to Saeman hydrolysis, the cell wall material of carrots was subjected to enzymatic hydrolysis using commercial enzymes such as Pectinex Ultra SP-L, Celluclast, ß-glucosidase and Driselase. The use of a combination of all enzyme preparations led to the most extensive degradation, the amount of dissolved cell wall material being 92 %. In this case, the monomer formation was 80 % of that obtained by Saeman hydrolysis. About one third of the residue obtained after enzymatic hydrolysis was found to be neutral monosaccharides consisting of 70 % D-glucose, which indicates that the enzymatic degradation of cellulose was incomplete. A complete monomer formation could not be achieved by using enzymatic hydrolysis only. However, a combination of the enzymatic hydrolysis and the main hydrolysis step of the Saeman procedura resulted in a monosaccharide yield (95 % mono saccharides compared to Saeman hydrolysis) and monosaccharide composition comparable to that obtained by Saeman hydrolysis. The influence of parb'cle size on the neutral sugar yield was examined using milfed and non-milled cell wall material. This showed that the enzymatic degradation, especially on the cellulose, was favored on the milled material.
