1 Abteilung Zelluläre Chemie Direktorin: Prof. Dr. rer. nat. Rita Gerardy-Schahn Forschungsprofil Die wissenschaftlichen Themen meiner Gruppe werden durch das Interesse an der Biosynthese und Funktion zellulärer Glycoconjugate gebündelt. Als Glycocalyx (über Proteine und Lipide gebunden) bilden die Zucker den äußeren Saum der tierischen Zelle und im Wesentlichen das Vokabular, mit dessen Hilfe die Zelle in Kommunikation mit ihrer Umgebung tritt. Veränderungen im Glycosylierungsmuster können schnell erreicht werden und begleiten die Steuerung zahlreicher (mit hoher Wahrscheinlichkeit aller) biologischen Prozesse. Einige Beispiele im Vertebraten sind: die Ausbildung ontogenetischer Muster, Adhärenz- und Wanderung von Entzündungs- und Nervenzellen, Entwicklung und Vermehrung von Tumorzellen. Im letzteren Falle werden Glycotope auch genutzt, um Tarnkappen auszubilden, die die Tumorzellen vor dem Immunsystem schützen. Schließlich ist die Empfindlichkeit eines Organismus für Pathogene im Wesentlichen über die anwesenden Zuckerdeterminanten bestimmt. Wir konzentrieren uns auf die Darstellung von Glycosylierungswegen im Verlauf zellulärer Entwicklungsprogramme, um so biologische Schalter zu definieren, über deren Beeinflussung das Differenzierungsprogramm von Zellen unterbrochen, in seiner Richtung verändert, oder, wie im Falle regenerativer Vorgänge gefordert, umgekehrt werden kann. In einem zweiten Schwerpunkt, der Glycoparasitologie, steht die Darstellung von Unterschieden in den Glycosylierungswegen von Wirt und eukaryontischen Pathogenen (Aspergilus fumigatus; Leishmania major) im Mittelpunkt. Mit der Aufdeckung von Unterschieden sollen Zielstrukturen für den therapeutischen Angriff definiert werden. Ein dritter Schwerpunkt widmet sich dem Studium der Evolution von Bakteriophagen. Studienobjekte sind dabei Phagen, die bekapselte, humanpathogene Bakterien befallen. Neben dem Erkenntnisgewinn zur Entwicklung von Wirtsspezifitäten, besteht das langfristige Ziel dieser Arbeiten in der Erarbeitung neuer, Phagen-basierter Therapiekonzepte zur Bekämpfung bakterieller Infektionen. Einen Spitzenplatz hinsichtlich Kreativität im Bereich der Glycobiochemie übernimmt die Pflanzenwelt und mit Dr. Hans Bakker ist, wie der nachfolgende Bericht zeigt, die PflanzenGlycoBioTechnologie auch in unser Labor gekommen. Forschungsprojekte Green complements red: The Modelling of plants towards production of mammalian type glycoproteins Clinical studies carried out with recombinant glycoproteins (e.g. EPOGEN/EPREX: Erythropoetin alpha from Amgen or AVONEX: Interferone beta 1-a from BioGen) have impressively demonstrated that minor changes in the protein glycosylation patterns are of significant influence on biological functions. 2 Because out of 360 recombinant proteins that are currently under investigation for medical application >200 are glycoproteins, the search for efficient, safe and reliable glycoprotein production systems is a major challenge in pharmaceutical research. Moreover, because each therapeutic protein has its specific, for therapeutic use optimal glycoprofile, the development of tailored production systems is essential to improve effectiveness and thereby reduce medication costs. Traditionally, Chinese hamster ovary or similar culture cells have been used for the production of therapeutic glycoproteins. While the obvious advantage of these systems resides in the production of mammalian type glycoproteins, there are numerous disadvantages associated with the mammalian cell production systems: Mammalian cell cultures are as a rule coast intensive, exhibit limited productivity, produce particularly in the situation of overproduction of recombinant proteins microheterogeneity in the glycosylation patterns and, not least, bear the risk that recombinant proteins are contaminated by viruses or prions. One of the future production platforms for therapeutic proteins is thought to be plant based. Especially when relatively large quantities of proteins are required, plants are the most economic hosts and recombinant proteins isolated from plants bear no risk to transfer pathogens. However, to make glycoprotein drugs in plants, plant specific glycosylation pathways must be switched to mammalian (particularly human) type. The problem is exemplified for therapeutic antibodies in Figure 1. A recombinant antibody isolated from a murine or other mammalian cell expression system, carries N-glycans in the Fc-domain, which, as schematically shown, are bi-antennary, carry core fucose in 1,6 linkage and terminate with galactose at both antennae. If made in plant cell cultures, the basic structure of the N-glycans is, at a first glance, fairly similar, however, differences (core fucose added in 1,3 linkage; xylose added to the bisecting mannose and lack of terminal galactose) that look small in the scheme offer the most challenging problem in therapeutic use as they provide immunogenicity and allergenicity. To make plants a suited system for the production of glycoprotein drugs, the systems must be engineered to loos plant type and gain mammalian type glycosylation pathways. 66 Forschungsbericht 2006

3 In cooperation with the group of Prof. Dirk Bosch at Plant Research International in Wageningen, Netherlands, I have been involved for several years now in the development of strategies to engineer plant cells towards mammalian type glycosylation. The first goal of our studies, thereby, consisted in the provision of proof that mammalian glycosyltransferases can be functionally introduced into the plant cell system. After the installation of the galactosylation pathway in tobacco plants had been successfully achieved (Bakker et al., PNAS 98: , 2001; Elbers et al., Plant Physiol. 26: , 2001) we were motivated to further refine the plant cell system to obtain mammalian identical structures. To rationalise the currently applied engineering strategies, it is important to understand, that the biosynthesis of glycan structures in all eukaryotic cells proceeds while the protein is transported from ER to Golgi to the plasma membrane. Biosynthetic steps are strictly consecutive because the glycosyltransferases are vectorially organised along ER and Golgi membranes. Logically, the transport of the maturing glycoprotein along the cascade of glycosyltransferases is crucial for the product that is synthesized and remodelling of enzyme positions changes the outcome of the biosynthetic products. Starting from this consideration our engineering strategy has been designed. It is known from earlier studies that xylose addition to the bisecting mannose in plants occurs in the medial Golgi at the same time as the addition of fucose (see Fig. 2A). Moreover, from pioneering experiments we knew that, if galactose was transferred to the maturing glycan core, the newly generated glycan was no longer an acceptor for xylosylation and fucosylation. Consequently we designed a plant cell in which the transgene, the mammalian galactosyltransferase, was forced to compete for the space of the endogenous xylosyltransferase. This was achieved by expression of a hybrid enzyme (called XylGalT), consisting of the N-terminal domain of Arabidopsis thaliana xylosyltransferase (contains subcellular transport signals) and the catalytic domain of human beta-1,4-galactosyltransferase Forschungsbericht

4 (GalT). The introduced changes in the spacial organisation of the glycosyltransferases in tobacco plants caused a sharp reduction of N-glycans with xylose and fucose residues. In line with our hypothesis, the early action of the galactosyltransferase prevented the action of the plant specific xylosyl- and fucosyltransferase (figure 2). In the recently published article (Bakker et al., PNAS 103: , 2006) we, therefore, demonstrates that a single measure was followed by three important consequences towards the installation of mammalian type glycosylation in plants.
