3 -1-1 Zusammenfassung Die vorliegende Arbeit befasst sich im ersten Teil mit einer neuen Methode zur enantio- und diastereoselektiven Herstellung a-verzweigter ß-Amino- Ausgehend von 3-Aminopropionsäure, (S)-3-Aminobuttersäure und (S)-3- Amino-5-methylhexansäure wurden Tetrahydro-pyrimidinone des Typs I hergestellt. Beim Aminopropionsäure-Derivat wurde auf dieser Stufe eine Racematspaltung mittels HPLC im präparativen Massstab durchgeführt. Die Umsetzung der Verbindungen des Typs enantiomerenreinen Dihydro-pyrimidinen des Typs II. I mit Meerwein-Salz führte zu HN N^Ofl2 N N OR2 3^R' ^CT^^R1 0 i ii Die Lithium-Enaminate aus II wurden mit verschiedenen Elektrophilen (Halogenide, Aldehyde, Imine und Enoate) umgesetzt, wobei der elektrophile Angriff vollständig diastereoselektiv trans zur terf-butylgruppe am Aminalzentrum der Dihydro-pyrimidine erfolgte. Die Aldol- und Michael- Additionen lieferten bevorzugt die Verbindungen des Jifce-Angriffes, wobei die relative Konfiguration der Produkte durch Kristallstrukturanalysen bestimmt wurde. Nach Abspaltung der Schutzgruppen wurden die Alkylierungsprodukte unter milden Bedingungen zu den entsprechenden Amino-methylestern hydrolysiert. Die Aldolprodukte wurden direkt zu den ß-Amino-ß'-hydroxysäuren umgesetzt. In einem zweiten Teil der Arbeit wurde das chirale Dendrimer zweiter Generation m hergestellt und dessen Einschlussverhalten gegenüber leichtflüchtigen Verbindungen untersucht.

Durch eine Mannicfc-artige Reaktion konnte mit einem aktivierten Imin diastereoselektiv eine Aminomethylgruppe in den ß- Aminosäureester eingebaut werden. Dadurch wurde die Verbindung IV erhalten. IA Da die direkte Transformation der Estergruppe in IV in eine Amidgruppe nicht gelang, wurde die Methylestergruppe verseift. Die weiteren Stufen zum duralen Triamin konnten in Rahmen der vorliegenden mehr durchgeführt werden. Arbeit nicht

5 -3-2 Summary This thesis deals in the first part with a new methodology for the preparation of enantio- and diastereoisomerically pure a-branched ß-amino acids. Starting from 3-aminopropionic acid, (S)-3-aminobutyric acid and (S)-3- amino-5-methylhexanoic add, tetrahydro-pyrimidinones of type I were prepared. The two enantiomers of the aminopropionic add derivative were separated by preparative HPLC. Treatment of the Compounds of type I with Meerwein's salt lead to enantiopure dihydro-pyrimidines of type II. fx HN^N^OR2 O^B' fx N^N^OR2 N^^R1 I II The lithium enaminates derived from II were allowed to read with several eledrophiles (halides, aldehydes, imines and enoates). The electrophilic attack occured diastereoselectively from the face trans to the terf-butyl group at the aminal center of the dihydro-pyrimidines. In the aldol- and Michaeladditions the products of like attack were predominantly formed. The relative configurations were determind by X-ray structure analysis. Cleavage of the protecting groups from the products of alkylation and hydrolysis under mild conditions afforded the corresponding amino methyl esters. The products of the aldol additions were directly converted to the ß- amino-ß'-hydroxy adds. In the second part of the thesis the second generation dendrimer HI was synthesized and the formation of guest/host complexes volatile Compounds was studied. with various

With a Mannich-type reaction a second nitrogen group could be introduced diastereoselectively in the ß-amino acid. Using this methodology Compound IV was prepared. IA The direct transformation of the ester group of Compound IV into an amide group was not successful. Therefore, the ester group was saponified. The final Steps of the synthesis of the chiral triamine could not be accomplished within this thesis.
