3 Abstract C-type cytochromes are ubiquitous electron transfer proteins with covalently bound heme as a cofactor. The mechanism by which the two thioether bonds between the cysteines of the CXXCH motif and the vinyl groups of the heme are formed is still unclear. In Escherichia coli, the operon ccmabcdefgh is known to be required for cytochrome c maturation. One of these proteins, CcmE, binds heme covalently at a strictly conserved histidine and subsequently delivers it to apo-cytochrome c. It was therefore denominated heme chaperone. Purification of soluble CcmE allowed biochemical characterization of this periplasmic protein. A major progress was the determination of the structure of soluble apo-ccme by NMR. It belongs to the OB (oligonucleotide/oligosaccharide binding) fold, a β-barrel arrangement characteristic for transient interactions with cargo molecules. As heme is neither an oligonucleotide nor an oligosaccharide, we suggested calling this fold more generally a cargo motif. Alanine scanning mutagenesis of the CcmE protein did not identify any residues involved in an acid-base catalyzed mechanism. The strongest reduction of holo-cytochrome c formation was found upon mutation of three hydrophobic residues close to the solvent-exposed heme binding histidine 130. This led to the proposal of a mechanism in which heme first interacts with a hydrophobic platform at the surface of CcmE, and then the covalent link is formed in a second step. This hypothesis has been substantiated recently by Daltrop et al. (2002). Using this information, heme was modeled to the surface of CcmE. Analysis of a heme-containing peptide resulting from a tryptic digest of holo-ccme showed that no protons are lost. This means that the heme binding reaction is an addition rather than a substitution. The required double bond is most probably provided by one of the vinyl groups of heme. To test this idea, histidine 130 was exchanged to a cysteine, thus mimicking the thioether bond in a c-type cytochrome. In fact, the CcmE H130C mutant showed residual heme binding; in its isolated heme peptide, heme was also bound covalently to the mutated cysteine residue. It has previously been shown that the heme chaperone CcmE directly interacts with CcmC and CcmF. In other proteins belonging to the OB fold, interactions with other subunits are performed via an α-helix at the C-terminal end of the compact core. However, in CcmE only a short α-helical turn could be identified, and the 29 residues directly after histidine 130 1

4 Abstract showed enhanced flexibility. This C-terminal domain was investigated by creating different truncated mutants. Weak heme binding was still observed when the complete C-terminal domain after H130 was removed, and for detectable levels of c-type cytochromes only one additional residue, D131, was required. Wild-type levels of holo-ccme were found for CcmE 1-138, and CcmE was fully active regarding cytochrome c formation. This indicated that only the short region directly after H130 is necessary for the function of CcmE, but not the α-helical turn and the following amino acids. Therefore, the last 20 of the C-terminal residues are in contrast to other proteins of this fold probably not necessary for interactions with other subunits of the Ccm complex. The N-terminal membrane anchor is also not required for heme binding of CcmE, but cytochrome c formation with soluble CcmE devoid of this domain is drastically reduced. 2

5 Zusammenfassung C-Typ Cytochrome sind weit verbreitete Elektronentransport-Proteine, in welchen Häm durch zwei Thioetherbindungen kovalent gebunden ist. Der Mechanismus, wie die beiden Cysteine aus dem CXXCH-Motiv mit den Vinylgruppen verknüpft werden, ist weitgehend unbekannt. Escherichia coli benötigt die acht Gene des Operons ccmabcdefgh für diesen als Cytochrom c-reifung bezeichneten Prozess. Eines dieser Genprodukte, CcmE, bindet die Hämgruppe in einem ersten Schritt kovalent an ein streng konserviertes Histidin und liefert sie anschliessend an Apo-Cytochrom c weiter. Aus diesem Grund wurde CcmE als Hämchaperon bezeichnet. Reinigung von löslichem CcmE ermöglichte die biochemische Charakterisierung dieses periplasmatischen Proteins. Einen Meilenstein stellte die Lösung der Struktur von löslichem Apo-CcmE mittels NMR-Spektroskopie dar. CcmE konnte dem OB-Faltungsmuster (für Oligonukleotid/Oligosaccharid-bindend) zugeordnet werden, einer β-fass-struktur, die meist bei Proteinen zu finden ist, welche transiente Wechselwirkungen mit den transportierten Molekülen eingehen. Da Häm weder ein Oligonukleotid noch ein Oligosaccharid ist, schlugen wir für das Faltungsmuster den allgemeineren Namen Cargo-Motiv vor. Das Ersetzen von konservierten Resten in CcmE durch Alanin ergab keine Aminosäuren, welche auf einen Säure-Base-katalysierten Mechanismus hinwiesen. Stattdessen wurde bei Mutation von drei hydrophoben Resten eine starke Reduktion der Cytochrom c-bildung beobachtet. Diese befinden sich alle in der Umgebung des Häm bindenden Histidins 130, welches an der Oberfläche des Proteins liegt. Als Folge wurde ein Reaktionsmechanismus vorgeschlagen, bei dem Häm zuerst mit einer hydrophoben Plattform auf dem Proteinmantel interagiert und erst anschliessend die kovalente Häm-Histidin-Bindung ausgebildet wird. Diese Hypothese wurde kürzlich durch eine Publikation von Daltrop et al. (2002) erhärtet. Mit Hilfe dieser Information wurde Häm an die Oberfläche von CcmE modelliert. Ein tryptischer Verdau von Holo-CcmE ergibt ein Häm-Peptid, das im Vergleich zu freiem Häm plus dem Peptid keine Protonen verloren hat. Das bedeutet, dass es sich beim Prozess der Hämbindung um eine Additionsreaktion handelt und nicht um eine Substitution. Die dazu benötigte Doppelbindung stammt wahrscheinlich von einer der Vinylgruppen des Häms. Um dies zu überprüfen, wurde Histidin 130 zu einem Cystein mutiert, um eine Situation analog zu 3

6 Zusammenfassung einem c-typ Cytochrom mit einer Thioetherbindung zu erzeugen. Tatsächlich wies die CcmE H130C-Mutante schwache Hämbindung auf, und das daraus isolierte Hämpeptid ergab, dass das Eisenporphyrin ebenfalls kovalent an das Cystein gebunden war. Wie bereits früher gezeigt, interagiert das Hämchaperon CcmE mit CcmC und CcmF. In anderen Proteinen, die das gleiche Faltungsmuster wie CcmE aufweisen, findet diese Wechselwirkung durch eine α-helix am C-terminalen Ende des Proteins statt. Für CcmE konnte in diesem Teilstück jedoch nur eine kurze α-helikale Struktur identifiziert werden, und die 29 Aminosäuren unmittelbar nach Histidin 130 wiesen eine erhöhte Flexibilität auf. Diese C-terminale Domäne wurde mit Hilfe von mehreren verkürzten Mutanten untersucht. Selbst wenn alle Reste nach H130 entfernt wurden, konnte noch Hämbindung nachgewiesen werden, und für detektierbare Mengen an Cytochrom c reichte eine einzige zusätzliche Aminosäure, D131, aus. CcmE produzierte Wildtyp-Mengen an Holo-CcmE, und CcmE war hinsichtlich der Cytochrom c-bildung unvermindert aktiv. Dies bedeutet, dass lediglich eine kurze Region gleich anschliessend an H130 für die Cytochrom c-reifung benötigt wird, aber nicht die kurze α-helikale Struktur und die folgenden Reste. Somit sind die letzten 20 Aminosäuren im Gegensatz zu anderen Proteinen wahrscheinlich nicht für die Interaktion mit anderen Untereinheiten des Ccm-Komplexes erforderlich. Der N-terminale Membrananker ist im Unterschied zur C-terminalen Domäne nicht nötig für die Hämbindung, Cytochrom c-reifung ist aber in löslichem CcmE ohne diese Verankerung drastisch reduziert. 4
