3 2. Summary Vacherin Mont d'or cheese was extracted with water, and the extract was fractionated by Ultrafiltration with a membrane exciuding Compounds with a molecular weight under 1'000. The filtrate was then lyophilized. Aliquots of the cheese extract were chromatographed on a Sephadex derivative containing N-(2-pyridylmethyl)glycine-groups in copper form. This resin permits a specific group Separation of peptides by ligand exchange chromatography. A fraction containing mainly a mixture of peptides was subdivided into five groups and further separated by ion exchange chromatography on Aminex A6 and Durrum DC 4 resin, respectively. A large number of peptide containing sub-fractions were obtained. Nine of the most interesting sub-fractions were then characterized by manual gasphase isothiocyanate degradation and HPLC of the amino acid derivatives. With this method seven peptides could be characterized, namely H-Leu- Pro-OH, H-Val-Pro-OH, H-Phe-Pro-OH, H-Lys-Pro-OH, H-Gly-Pro-Val-Arg- OH, H-Tyr-Pro-OH, and H-Arg-Pro-OH. A partial elucidation of the structure was possible for peptides containing Asp, Pro, Val, Leu, and Glu, Leu, and Ala, Pro. The taste qualities of the identified peptides were evaluated in accordance with their Q-values. Four dipeptides were sensorially tested with synthetic analogues and had bitter taste.

4 3 The oecurrence of the characterized peptides in cheese has not been reported heretofore. It can be assumed that the peptides contribute to the overall flavor of Vacherin Mont d'or cheese in a positive way.

5 3. Zusammenfassung Vacherin Mont d'or Käse wurde mit Wasser extrahiert. Der Extrakt wurde ultrafiltriert durch eine Membran mit einer Ausschlussgrenze für Moleküle über 1'000 Dalton. Das Filtrat wurde lyophilisiert. Aliquote Anteile des Extraktes wurden durch Ligandenaustauschchromatographie an einem N-(2-Pyridylmethyl)glycin-Derivat von Sephadex in Kupferform fraktioniert. Dieses Sephadex-Derivat ist geeingnet zur Gruppenabtrennung von Peptiden. Die Peptidfraktion der Ligandenaustauschchromatographie wurde in 5 Fraktionen aufgeteilt, diese Fraktionen wurden durch lonenaustauschchromatographie an Aminex A6 bzw. Durrum DC 4 weiter aufgetrennt. Es resultierten zahlreiche Peptide enthaltende Subtraktionen. Bei neun Subfraktionen wurden die Peptide durch manuellen Isothiocyanat-Abbau in der Gasphase und anschliessende HPLC-Trennung der Aminosäure- Derivate charakterisiert. Es konnte das Vorkommen der folgenden sieben Peptide nachgewiesen werden: H-Leu-Pro-OH, H-Val-Pro-OH, H-Phe-Pro-OH, H-Lys-Pro-OH, H-Gly-Pro-Val-Arg-OH, H-Tyr-Pro-OH, and H-Arg-Pro-OH. Das Vorkommen weiterer Peptide, aufgebaut aus den Aminosäuren Asp, Pro, Val, Leu, und Glu, Leu, und Ala, Pro, konnte nur partiell abgeklärt werden.

6 5 Die sensorischen Eigenschaften der Peptide in Abhängigkeit von ihren Q-Werten wurden untersucht. Das Vorkommen der oben aufgeführten Peptide in Käse ist bisher nicht beschrieben worden. Es darf angenommen werden, dass diese Peptide zum Geschmack des Vacherin Mont d'or Käses beitragen.
