3 Kurzinformation RIDASCREEN CIS (Art. Nr.: R4302) ist ein Enzymimmunoassay zur Bestimmung von Kuhmilch in Schafs- und Ziegenmilch bzw. in Schafs- und Ziegenkäse. Alle Reagenzien für die Durchführung des Enzymimmunoassays - inkl. Standards - sind im Testkit enthalten. Ein Testkit ist ausreichend für 48 Bestimmungen (einschl. Standardbestimmungen). Zur Auswertung benötigt man ein Mikrotiterplatten-Photometer. 4 1. Verwendungszweck RIDASCREEN CIS ist ein Enzymimmunoassay zur Bestimmung von Kuhmilch in Schafs- und Ziegenmilch bzw. in Schafs- und Ziegenkäse. 2. Allgemeines Da Kuhmilch deutlich billiger ist als Schafs- und Ziegenmilch, wird Schafs- und Ziegenmilch häufig mit Kuhmilch verfälscht. Milchverfälschungen können u.a. mit chromatographischen und elektrophoretischen Verfahren nachgewiesen werden. Diese Methoden erfordern einen erheblichen zeitlichen und arbeitstechnischen Aufwand und sind daher für den Einsatz in der Routineanalyse nur bedingt geeignet. 3. Testprinzip Grundlage ist die Antigen-Antikörper-Reaktion. Der Nachweis von Kuhmilchzusatz erfolgt über die Bestimmung von bovinem IgG (Immunglobulin G ist eine Antikörperklasse). Diese Antikörper sind ein natürlicher Bestandteil der Kuhmilch. Die Vertiefungen der Mikrotiterplatte sind mit spezifischen Antikörpern gegen bovines IgG beschichtet. Im Falle einer Verfälschung des Produktes mit Kuhmilch, bindet darin enthaltenes IgG an den Fänger-Antikörper. Nicht gebundene Reaktionspartner werden anschließend in einem Waschschritt wieder entfernt. Mit einem Peroxidase-gekoppelten Antikörper gegen bovines IgG wird gebundenes Antigen (in diesem Fall bovines IgG) nachgewiesen. Nicht gebundenes Enzymkonjugat wird anschließend in einem Waschschritt wieder entfernt. Der Nachweis erfolgt durch Zugabe von Substrat und Chromogen. Gebundenes Enzymkonjugat wandelt das Chromogen in ein blaues Endprodukt um. Die Zugabe des Stopp- Reagenzes führt zu einem Farbumschlag von blau nach gelb. Die Messung erfolgt photometrisch bei 450 nm. 4 RIDASCREEN CIS

5 4. Packungsinhalt Mit den Reagenzien einer Packung können 48 Bestimmungen durchgeführt werden (einschließlich Standardbestimmungen). Jedes Testkit enthält: 1 x Mikrotiterplatte mit 48 Kavitäten (6 Streifen à 8 Einzelkavitäten) beschichtet mit Antikörpern gegen bovines IgG 3 x Milchstandards *), je 1,3 ml 1A: 10 % Kuhmilchanteil 2A: 1 % Kuhmilchanteil 3A: 0,1 % Kuhmilchanteil 3 x Käsestandards *), je 1,3 ml 1B: 10 % Kuhmilchanteil 2B: 1 % Kuhmilchanteil 3B: 0,1 % Kuhmilchanteil 1 x Konjugat (1,3 ml)... roter Verschluss Peroxidase-konjugierte Antikörper, Konzentrat 1 x Substrat (7 ml)... grüner Verschluss enthält Harnstoffperoxid 1 x Chromogen (7 ml)... blauer Verschluss enthält Tetramethylbenzidin 1 x Stopp-Reagenz (14 ml)... gelber Verschluss enthält 1 N Schwefelsäure 1 x Puffer (13 ml)... weißer Verschluss Konjugatverdünnungspuffer *) In den Angaben sind die Verdünnungsfaktoren (100 für Milch, 10 für Käse) bereits enthalten, so dass bei der Auswertung der Anteil an Kuhmilch direkt abgelesen werden kann. 5. 6 6. Vorsichtsmaßnahmen Dieser Test ist nur von geschultem Laborpersonal durchzuführen. Die Gebrauchsanweisung zur Durchführung des Tests ist strikt einzuhalten. Das Stopp-Reagenz enthält 1 N Schwefelsäure (R36/38, S2-26). 7. Reagenzien und ihre Lagerung Die Reagenzien bei 2-8 C lagern. Komponenten des Testkits auf keinen Fall einfrieren. Nicht benötigte Kavitäten zusammen mit dem Trockenmittel im Folienbeutel gut verschlossen aufbewahren und weiterhin bei 2-8 C lagern. Die farblose Chromogenlösung ist lichtempfindlich, deshalb direkte Lichteinwirkung vermeiden. Nach Ablauf des Verfallsdatums (siehe Testkit-Außenetikett unter Expiration) kann keine Qualitätsgarantie mehr übernommen werden. Ein Austausch von Einzelreagenzien zwischen Kits verschiedener Chargennummern ist nicht zulässig. 8. Anzeichen für Reagenzienverfall bläuliche Färbung der Chromogenlösung vor Zugabe in die Kavitäten Extinktion kleiner 0,8 (E 450 nm < 0,8) für Standard 1A und 1B 9. Probenvorbereitung Die Proben kühl und lichtgeschützt lagern Milch Milchproben 1:100 (1+99) mit dest. Wasser verdünnen (z.b. 0,1 ml Milch + 9,9 ml dest. Wasser) jeweils 100 µl pro Kavität im Test einsetzen 9.2. Käse 1 g Käse mit 10 ml dest. 7 10. Testdurchführung Testvorbereitungen Alle Reagenzien vor Gebrauch auf Raumtemperatur (20-25 C) bringen. Das Antikörper-Enzymkonjugat (Flasche mit rotem Verschluss) liegt als Konzentrat vor. Da die rekonstituierte Konjugatlösung nur begrenzte Haltbarkeit aufweist, immer nur soviel Konjugat-Konzentrat mit Puffer verdünnen, wie unmittelbar benötigt wird. Das Konjugat-Konzentrat vor Entnahme vorsichtig mischen. Um die gebrauchsfertige Konjugatlösung herzustellen, muss das Konzentrat 1:11 (1+10) mit Puffer verdünnt werden (z.b. 100 µl Konzentrat + 1 ml Puffer, ausreichend für 1 Mikrotiterstreifen). Die Milchstandardlösungen (in 1 % Schafsmilch, dest. Wasser, 0,1 % Azid) liegen in folgenden Konzentrationen vor: Standard 1A: 0,1 % Kuhmilchanteil in verdünnter Schafsmilch Standard 2A: 0,01 % Kuhmilchanteil in verdünnter Schafsmilch Standard 3A: 0,001 % Kuhmilchanteil in verdünnter Schafsmilch Unter Berücksichtigung der Proben-Verdünnung von 1:100 entsprechen die Standards daher 10 %, 1 % und 0,1 % Kuhmilchanteil in der Probe. Die Käsestandardlösungen (in 1 % Schafsmilch, dest. Wasser, 0,1 % Azid) liegen in folgenden Konzentrationen vor: Standard 1B: 340 ng/g bovines IgG, entspricht ca. 1,0 % Kuhmilchanteil in Schafs- bzw. Ziegenkäse. Standard 2B: 34 ng/g bovines IgG, entspricht ca. 0,1 % Kuhmilchanteil in Schafsbzw. Ziegenkäse. Standard 3B: 3,4 ng/g bovines IgG, entspricht ca. 0,01 % Kuhmilchanteil in Schafs- bzw. Ziegenkäse. Unter Berücksichtigung der Proben-Verdünnung von 1:10 entsprechen die Standards daher 10 %, 1 % und 0,1 % Kuhmilchanteil in der Probe. 8 10.2. Testdurchführung Sorgfältiges Waschen ist sehr wichtig. Ein Eintrocknen der Kavitäten zwischen den Arbeitsschritten vermeiden. 1. So viele Kavitäten in den Halterahmen einsetzen, wie für alle Standards in Doppelbestimmung und Proben in Einfachbestimmung benötigt werden. Die Positionen der Standards und der Proben protokollieren. 2. Je 100 µl der Standardlösung (Milch und / oder Käse) bzw. der vorbereiteten Proben als Doppel- bzw. Einfachbestimmung in die entsprechenden Kavitäten pipettieren und 30 min bei Raumtemperatur (20-25 C) inkubieren. 3. Die Kavitäten durch Ausschlagen der Flüssigkeit leeren und die Restflüssigkeit durch kräftiges Ausklopfen (dreimal hintereinander) auf saugfähigen Labortüchern entfernen. Die Kavitäten mit jeweils 250 µl destilliertem Wasser waschen. Diesen Vorgang zweimal wiederholen. 4. Je 100 µl verdünnte Enzymkonjugat-Lösung in die entsprechenden Kavitäten pipettieren, vorsichtig manuell mischen und 30 min bei Raumtemperatur (20-25 C) inkubieren. 5. Die Kavitäten durch Ausschlagen der Flüssigkeit leeren und die Restflüssigkeit durch kräftiges Ausklopfen (dreimal hintereinander) auf saugfähigen Labortüchern entfernen. Die Kavitäten mit jeweils 250 µl destilliertem Wasser waschen. Diesen Vorgang zweimal wiederholen. 6. Je 50 µl Substrat und je 50 µl Chromogen in die entsprechenden Kavitäten pipettieren. Vorsichtig manuell mischen und 30 min bei Raumtemperatur (20-25 C) im Dunkeln inkubieren. 7. Je 100 µl Stopp-Reagenz in jede Kavität pipettieren und vorsichtig manuell mischen. Die Extinktion bei 450 nm innerhalb von 30 min nach Zugabe des Stopp-Reagenzes messen. 11. Auswertung Milch: Den Extinktionsmittelwert (450nm) jedes Standards (1A, 2A, 3A) berechnen. Liegt die Extinktion (450nm) der Milchprobe unterhalb des Standards 3A, so enthält die Milch keine oder weniger als 0,1 % Kuhmilch. Liegt die Extinktion (450nm) der Milchprobe zwischen Standard 2A und 3A, so enthält die Milch weniger als 1 % Kuhmilch. Liegt die Extinktion (450nm) der Milchprobe oberhalb des Standards 2A, so enthält die Milch mehr als 1 % Kuhmilch. 8 RIDASCREEN CIS

9 Käse: Den Extinktionsmittelwert (450nm) jedes Standards (1B, 2B, 3B) berechnen. Liegt die Extinktion (450nm) der Käseprobe unterhalb des Standards 3B, so enthält de0r Käse keine oder weniger als 0,1 % Kuhmilch. Liegt die Extinktion (450nm) der Käseprobe zwischen Standard 2B und 3B, so enthält der Käse weniger als 1 % Kuhmilch. Liegt die Extinktion (450nm) der Probe oberhalb des Standards 2B, so enthält der Käse mehr als 1 % Kuhmilch. Anmerkung: Der RIDASCREEN CIS-Test detektiert natürliche Bestandteile (bovines IgG) von Kuhmilch. Der Test bleibt bei der Verwendung von ultrahocherhitzter Kuhmilch (H-Milch) negativ, da die Immunglobuline (IgGs) bei diesem Verfahren denaturieren und damit nicht mehr detektiert werden können. Diese Angaben entsprechen dem heutigen Stand unserer Kenntnisse und sollen über unsere Produkte und deren Anwendungsmöglichkeiten informieren. Sie haben somit nicht die Bedeutung, bestimmte Eigenschaften der Produkte oder deren Eignung für einen konkreten Einsatzzweck zuzusichern. 11 1. Intended use RIDASCREEN CIS is an enzyme immunoassay for the detection of cow s milk in sheep s and goat s milk and in sheep s and goat s cheese. 2. General Sheep s and goat s milk are much more expensive than cow s milk, which is often used to extend sheep s and goat s milk. The falsification of milk can be detected chromatographically or by electrophoresis. These methods are laborious and require sophisticated equipment. Therefore, they are of limited value in routine screening of milk and cheese. 3. Test principle The basis of the test is the antigen-antibody reaction. The presence of cow s milk in a sample is determined by the immunological detection of bovine IgG (immunoglobulin G is an antibody class), which is a natural constituent of cow s milk. The wells of the microtiter strips are coated with specific antibodies against bovine IgG. In the case of adulterated products, the antibodies contained in the cow s milk will bind to the immobilized antibody. Any unbound components are removed in a washing step. By adding a peroxidase conjugated antibody directed against bovine IgG, bound antigen (in this case bovine IgG) is detected. Any unbound enzyme conjugate is then removed in a washing step. Enzyme substrate and chromogen are added to the wells and incubated. Bound enzyme conjugate converts the chromogen into a blue product. The addition of the stop reagent leads to a color change from blue to yellow. The measurement is made photometrically at 450 nm. RIDASCREEN CIS

13 6. Warnings and precautions for the users This test should only be carried out by trained laboratory employees. The instruction for use must be strictly followed. The stop solution contains 1 N sulfuric acid (R36/38, S2-26). 7. Storage instructions Store the kit at 2-8 C (35-46 F). Do not freeze any test kit components. Return any unused microwells to their original foil bag, reseal them together with the desiccant provided and further store at 2-8 C (35-46 F). The colorless chromogen is light sensitive, therefore, avoid exposure to direct light. No quality guarantee is accepted after expiry of the kit (see kit label). Do not interchange individual reagents between kits of different lot numbers. 8. 14 10. Test implementation Preliminary comments Bring all reagents to room temperature (20-25 C / F) before use. The antibody enzyme conjugate (enzyme conjugate, bottle with red cap) is provided as a concentrate. Since the diluted enzyme conjugate solution has a limited stability, only the amount which actually is needed should be reconstituted. Before pipetting, the conjugate concentrate should be shaken carefully. For reconstitution, the conjugate concentrate is diluted 1:11 (1+10) in buffer (e.g. 100 µl concentrate + 1 ml buffer, sufficient for 1 microtiter strip). The standards for milk (in 1 % sheep s milk, dist. water, 0.1 % azide) are provided ready for use in the following concentrations: Standard 1A: 0.1 % cow s milk diluted in sheep s milk Standard 2A: 0.01 % cow s milk diluted in sheep s milk Standard 3A: % cow s milk diluted in sheep s milk Considering sample dilution of 1:100, the standards correspond to 10 %, 1 % and 0.1 % cow s milk in a sample. 15 10.2. Test procedure Carefully follow the recommended washing procedure. Do not allow microwells to dry between working steps. 1. Insert a sufficient number of wells into the microwell holder for all standards to be run in duplicate and samples to be run in single determination. Record standard and sample positions. 2. Add 100 µl of each standard solution (milk and / or cheese) to separate duplicate wells and the prepared sample to separate wells. Incubate for 30 min at room temperature (20-25 C / F). 3. Pour the liquid out of the wells and tap the microwell holder upside down vigorously (three times in a row) against absorbent paper to ensure complete removal of liquid from the wells. Fill all the wells with 250 µl distilled water and pour out the liquid again. Repeat two more times. 4. Add 100 µl of the diluted enzyme conjugate to each well, mix gently by shaking the plate manually and incubate for 30 min at room temperature (20-25 C / F). 5. Pour the liquid out of the wells and tap the microwell holder upside down vigorously (three times in a row) against absorbent paper to ensure complete removal of liquid from the wells. Fill all the wells with 250 µl distilled water and pour out the liquid again. Repeat two more times. 6. Add 50 µl of substrate and 50 µl of chromogen to each well. Mix gently by shaking the plate manually and incubate for 30 min at room temperature (20-25 C / F) in the dark. 7. Add 100 µl of the stop reagent to each well. Mix gently by shaking the plate manually and measure the absorbance at 450 nm. Read within 30 minutes after addition of stop solution. 11. Results milk: The mean absorbance at 450 nm is determined for each standard (1A, 2A, 3A). Samples, that give an absorbance at 450 nm less than the standard 3A, are not falsified or contain less than 0.1 % cow s milk. Samples, that give an absorbance at 450 nm less than standard 2A und greater than standard 3A, contain less than 1 % cow s milk. 16 cheese: The mean absorbance at 450 nm is determined for each standard (1B, 2B, 3B). Samples, that give an absorbance at 450 nm less than the standard 3B are not falsified or contain less than 0.1 % cow s milk. Samples, that give an absorbance at 450 nm less than standard 2B and greater than standard 3B, contain less than 1 % cow s milk. Samples, that give an absorbance at 450 nm greater than standard 2B, contain more than 1 % cow s milk). Remark: The RIDASCREEN CIS test detects natural components of cow s milk (bovine IgG). The test will remain negative, if ultra-high-heated cow s milk is used. Immunglobulines (IgGs) denaturate in the process of ultra high heating and therefore they cannot be detected.
