4 Neues aus der Geschäftsstelle Editorial Ein Rauch verweht, ein Wasser verrinnt, eine Zeit vergeht, eine neue beginnt. (Joachim Ringelnatz, eigentlicher Name: Hans Gustav Bötticher, deutscher Schriftsteller, Maler und Komödiant, ) Liebe Leser des APV Plus-Magazins, zum Jahresende ist es wieder Zeit, auf das vergangene Jahr zu blicken und die Pläne für das kommende Jahr zu schmieden. Viele haben aufgrund der politischen und gesellschaftlichen Situation auf das nun fast vergangene Jahr mit großen Sorgen geblickt. Dazu gab es auch berechtigten Anlass, aber die meisten von Ihnen werden die 12 Monate so wie ich sicher, gesund und gut überstanden haben. Die USA sind unter ihrem neuen Präsidenten nicht in einen verheerenden Atomkrieg gezogen, die Europäische Union ist nicht zerborsten und die Rechtspopulisten haben in den Mitgliedsländern zwar Stimmen hinzugewonnen, konnten aber letztlich nicht die Macht an sich reißen. Wie in Joachim Ringelnatz Zeilen habe ich den Eindruck, dass im letzten Jahr eine mir zuvor lieb gewordene Zeit vergangen ist und eine neue Zeitrechnung begonnen hat. Diese gilt es nun pro-aktiv zu gestalten. Die APV blickt erneut auf ein sehr gutes Jahr zurück. Die European Conference on Pharmaceutics in Krakau war ein großer Erfolg und auch die anderen Veranstaltungen in diesem Jahr konnten zum guten Gesamtergebnis beitragen. Nun fokussieren wir auf das Jahr 2018 und dabei besonders auf das kommende PBP World Meeting in Granada. Hier werden auch die Wahlen für den kommenden Vorstand der APV im Rahmen der Mitgliederversammlung durchgeführt. Einige Vorstandsmitglieder werden aus privaten oder aus beruflichen Gründen für die kommende Legislaturperiode nicht mehr zur Verfügung stehen. Das ist schade, weil wir hervorragend zusammen gearbeitet haben und zu guten Freunden geworden sind. Eine erfolgreiche Zeit vergeht, eine neue Zeit beginnt mit anderen Köpfen. Sie erhalten als Mitglied der APV die Chance, mit Ihrem Stimmzettel die Zukunft der APV mitzugestalten. Machen Sie von Ihrem Stimmrecht Gebrauch und kommen Sie nach Granada. Mein herzlicher Dank geht an dieser Stelle an die vielen engagierten Kräfte, die Fachgruppen- und Vorstandsmitglieder, die im Ehrenamt das Programm der APV im letzten Jahr mitgestaltet haben, sowie an alle Mitarbeiterinnen und Mitarbeiter der Geschäftsstelle in Mainz. 5 Neues aus der Geschäftsstelle Die Geschäftsstelle stellt sich vor Name Melanie Gerhardt Geburtsort Mainz Ausbildung/Berufserfahrung Fachabitur Ausbildung zur Kauffrau für Bürokommunikation Außendienstservice und Vertriebscontrolling in einem mittelständischen Pharmaunternehmen Wie ich zur APV gekommen bin Nach meiner Elternzeit wollte ich vorerst auf geringfügiger Basis als kaufmännische Angestellte wieder in das Berufsleben einsteigen, um mir weiterhin Zeit für meine Tochter nehmen zu können, solange sie noch klein ist. Ein passendes Stellenangebot bei der APV fand ich im Internet. Meine Hauptaufgaben bei der APV Anmeldungen für Veranstaltungen bearbeiten Administrative Aufgaben Vorstandsbetreuung Was ich am liebsten in meiner Freizeit mache am liebsten verbringe ich Zeit mit meiner Familie und wir spielen mit unserer Tochter. Wenn es mal sportlich werden soll, gehe ich gerne ins Jumping, um mich auszupowern. Entspannung finde ich beim Treffen mit Freunden und beim Trinken eines leckeren Latte Macchiato. Ich freue mich bei APV am meisten auf... den Wiedereinstieg ins Berufsleben nach meiner Elternzeit, neue Kolleginnen und Kollegen kennenzulernen und auf die Arbeit im Bereich Teilnehmerverwaltung. Wenn ich einen Wunsch frei hätte würde ich mir wünschen, dass alles im Leben etwas lockerer zugeht und man nicht alles so ernst nimmt. Sich einfach mehr Zeit für Entspannung nimmt, sich mehr auf die wichtigen Dinge im Leben konzentriert und man sich immer wieder bewusst macht, wie froh man sein kann, wenn man gesund ist. Unsere wichtigste Aufgabe ist es, die entsprechenden Erkenntnisse über Entwicklung, Herstellung, Vertrieb und Gebrauch von Arzneimitteln zu vertiefen und dieses Wissen denjenigen zu vermitteln, die beruflich mit diesen Aufgaben befasst sind. Darüber hinaus sind unsere Ziele, die pharmazeutische Verfahrenstechnik, die Qualitätssicherung der Arzneimittelherstellung, sowie die Arzneiformen und ihre Applikationstechniken weiter zu entwickeln, zu verbessern und auf diese Weise die Therapiemöglichkeiten zu erweitern. Die APV leistet somit einen wichtigen Beitrag zur Arzneimittelsicherheit und zur Pharma-Ökonomie. Entscheidend: die APV Fachgruppen Die APV setzt auf die Vernetzung und den interdisziplinären Wissensaustausch in Fachgruppen sowie deren Kommunikation untereinander. Die Fachgruppen entwickeln die APV-Fortbildungskonzepte, die gezielt auf die Bedürfnisse der Teilnehmer aus Industrie, Wissenschaft und Behörde abgestimmt sind. Regelmäßige Treffen der Fachgruppenmitglieder ermöglichen den methodischen Austausch über verschiedene Teilbereiche eines Gebietes. Ein bedeutender Faktor dabei ist der Wissenstransfer über die Grenzen des eigenen Fachgebiets hinaus. Auf den fachgruppenübergreifenden Tagungen bilden die Fachgruppenleiter das wissenschaftliche Entscheidungsgremium für die Bearbeitung neuer Projekte und Konzepte. 08 APV Plus - 04/2017

Das Spektrum der Veranstaltungen reicht dabei von Expertentreffen über Seminare bis hin zu wissenschaftlichen Kongressen und Ausstellungen. Das APV-Angebot ist in zehn Fachbereiche gegliedert, bei denen garantiert für jeden etwas dabei ist. Die Seminarthemen und -inhalte werden von unseren Experten in den Fachgruppen ausgewählt und konzipiert. Die Seminare und das Rahmenprogramm bieten vielfältige Möglichkeiten für Networking. 22 Messen und Kongresse APV Conference Report Medicines for older adults: Getting Prepared for the scientific and regulatory evolution November 7th & 8th, 2017 in Graz (Austria) Sven Stegemann, Technische Universität Graz, Austria Increasing life expectancy is a fundamental achievement of modern societies, supported by effective drug therapy to treat and manage acute and chronic diseases. Due to the increasing prevalence for chronic diseases, older patients are the major user group of pharmaceutical drug products. They often suffer from multiple diseases, develop disabilities and have to manage complex medication schedules. There is increasing evidence that the pharmaceutical development and drug product design process has to be adapted to the specific needs of the older patient population. Older patients, often only defined by their chronological age, are a heterogeneous population with many different individual trajectories. Within these population, patients with multimorbidity or at very high age ( 80 years) require special attention and have needs beyond the standard care. From a geriatricians standpoint, prescribing is a fine balance between the risk and the relative benefit, which needs to be seen in the context of the co-morbidities, other medicines and the patient response to a treatment as well as the patients personal health goals. In hospital settings, the existing drug products have to be modified or compounded in order to achieve the right dose or to enable drug administration (e.g. through feeding tubes). Similar issues are frequently reported from primary care as medication handling issues and unintended errors lead to poor therapeutic outcomes. To overcome these issues, patient centric drug product design is often being claimed, however, clinical or scientific evidence remain very limited. Recent investigations into the management of different packaging designs by older patients provided some good insight into design elements improving usability and user satisfaction. Implementing patient centric drug product design into an industrial development process has to be seen from an organizational and legislative perspective, since drug development as such is a lengthy and complex process. Moreover, the final drug product has to fulfill many different requirements to assure efficacy, patient safety and accessibility. To achieve the objectives, predictive tools are required allowing the evaluation of different formulations based on relevant in vitro models like dissolution test media that are biorelevant for a specific patient population. With the required development of age-appropriate formulation for pediatric use, experience exist with collaborative efforts to gain the scientific and patient based evidence to improve products for a special patient population. Taking into account that older patients are the major user group of pharmaceutical products, the pharmaceutical industry should be encouraged to evaluate the implementation of patient centric product design based on incremental product enhancement within the development timelines. Moving to patient centric drug products might also include to make use of advancing manufacturing technologies allowing for small volume or even personalized manufacturing. Especially the emerging digital health technologies will create new ways of treating and monitoring patients as well as move from a reactive to a proactive healthcare delivery. Using product-user interface analysis in the development of computerized medical recording software have been applied successfully to improve the value and performance for healthcare professionals. The same applies for the communication to laypeople like patients to provide important information in a clear and simple way. Human Factor Design has been established for the development of device based drug products following a logic process involving the targeted patient. To assure the best risk to benefit profile of a drug product, the Reflection Paper on pharmaceutical development of medicines for use in the older population have been published as a draft for public consultation. The reflections review the scientific information and summarizers the identified major aspects that should be considered during the development to address the needs of older patients. This APV conference brought together a broad group of stakeholders and experts from the medical, pharmaceutical, Human Factor Design and technology field concluding that patient centric drug product design has to continue to support the scientific and professional collaboration to improve medicines for older patients. Based on the feedback, APV will take the leadership in continuing the interdisciplinary collaboration and exchange to build the required scientific evidence towards patient centric drug product design from the concept through to the market. 22 APV Plus - 04/2016

23 Messen und Kongresse 20. POWTECH in Nürnberg brachte die Welt der Verfahrenstechnik zusammen Die Welt zu Gast: 39 Prozent internationale Aussteller Save the Date: POWTECH 2019 vom 9. bis 11. April Die 20. POWTECH in Nürnberg ist nach drei erfolg- und ereignisreichen Tagen zu Ende gegangen. Die bisher internationalste POWTECH brachte in sechs Messehallen 835 Aussteller aus 29 Ländern zusammen. 39 Prozent der Aussteller reisten aus dem Ausland an, eine neue Bestmarke. Damit untermauerte die POWTECH ihre Position als weltweit führende Messe für Pulver-, Schüttgut-, und Partikeltechnologien und als Pflichttermin für Maschinen- und Anlagenbauer, um ihre Neuheiten und Innovationen zu präsentieren. Die POWTECH erwies sich als wichtigster Treffpunkt für die Schüttgut-Community vielfältigster Branchen mit Schwerpunkt Food, Pharma und Chemie bis hin zu Bau, Keramik oder Recycling. Ein internationales Publikum von über Besuchern profitierte neben dem breiten Messeangebot von einem hochkarätigen Fachprogramm. Die POWTECH 2017 hat beeindruckt, bewegt und die besten Köpfe der Branche zusammengebracht, fasst Beate Fischer, Veranstaltungsleiterin POWTECH bei der NürnbergMesse, zusammen. Nur hier können Verfahrenstechniker ein unvergleichbar breites Angebot aller mechanischen Verfahren begutachten, nur hier haben sie in zahllosen Vorträgen und Seminaren Zugriff auf Expertenwissen aus erster Hand. Die POWTECH sucht auch nach 20 Messeausgaben in Nürnberg ihresgleichen. Gute Gespräche, viele Exponate Auf einer Nettofläche von Quadratmetern präsentierten die Aussteller mechanische Verfahrenstechnik in ganzer Vielfalt. An vielen Ständen bestimmten komplette Anlagen und Maschinen zum Fördern, Trennen, Mahlen, Mischen, Rühren oder Agglomerieren das Bild, viele davon im Livebetrieb. Darüber hinaus zeigten die Aussteller Lösungen zur Steuerung und Automatisierung, sowie Komponenten und prozessbegleitende Technologien. Dazu zählen unter anderem Mess- und Analysegeräte, Filter-, Luftreinhaltungs- und Explosionsschutz-Equipment. Die Aussteller lobten die sehr hohe Qualität der Gespräche an ihren Ständen und das hohe Interesse der Fachbesucher. International wie noch nie Unter den internationalen Ausstellern kam die größte Gruppe mit 59 Unternehmen aus Italien, gefolgt von der Schweiz mit 34 Ausstellern und Großbritannien mit 28 Ausstellern. Ein deutlicher Zuwachs war zudem aus China zu verzeichnen (insgesamt 18 Aussteller). Die POWTECH 2017 lockte Fachbesucher nach Nürnberg (Herbstveranstaltung 2014: Fachbesucher). Rund 40 Prozent von ihnen kamen aus dem Ausland angereist, allen voran aus den Ländern Österreich, Italien, der Schweiz, Tschechien und Polen. Neben technischen Innovationen aus aller Welt bot ihnen die Messe viele Networking-Anlässe und ein umfangreiches Fachprogramm mit Fortbildungscharakter. Pulver-Profis im branchenübergreifenden Dialog Praxisnahe Vorträge rund um die mechanische Verfahrenstechnik gab es in zwei Fachforen auf der Messe zu erleben. Im Außengelände stellte der Ind Ex Verband im Rahmen von spektakulären Live-Explosionen die Wirksamkeit von Ex-Schutz-Equipment unter Beweis. Der VDMA Fachverband Allgemeine Lufttechnik präsentierte in Halle 2 die Sonderschau Staubfreie Zukunft mit zahlreichen Exponaten. Die zweite Sonderschau stand unter dem Motto Pharma- Feststoffe und vereinte rund 20 Maschinen und Anlagen für die relevanten Prozessschritte vom Mahlen bis zur Partikelanalyse. APV Plus - 04/

International experts from academia and industry presented in their seminars innovative and new applications of phospholipids. Categorised in five topics: Analytics and Characterisation Techniques, In vitro/physicochemical Properties, Drug Delivery, Technological Approaches and Skin Application, 69 scientists presented their posters on topics in phospholipid research. The first day started with a general introduction to phospholipids. PD Dr. Peter van Hoogevest (Phospholipid Research Center, DE) and Dr. Ralf-Olaf Quinkert (Lipoid GmbH, DE) highlighted properties, manufacturing and analytics of phospholipids. An example for the versatile deployment of phospholipids presented Prof. Dr. Alfred Fahr (University of Jena, DE) vividly. His explanations focused on special lipid bilayers, comprising phosphatidylserine, arranged in cylindrical continuous sheets, called cochleates. The audience enthusiastically listened to these explanations and to the subsequent session on examples of the industrial use of phospholipids presented by Dr. Josbert Metselaar (University of Twente, NL), Dr. Daryl Drummond (Merrimack Pharmaceuticals, US) and Dr. Jürgen Schmitt (B. Braun Melsungen AG, DE). Next to the speakers, exhibitors of different companies facilitated an active exchange between academia and industry as well. Representatives of Particle Solutions Innovations B.V., Avestin Europe GmbH, Malvern Instruments GmbH, Microfluidics and Polymun Scientific presented their skills. Research projects with a special focus on the parenteral administration of phospholipids were highlighted by the seminars of Prof. Dr. Jean-Christophe Leroux (ETH Zürich, CH) Detoxification using parenteral lipid formulations and Prof. Dr. Karsten Mäder (University of Halle, DE) Phosphatidylserine enriched phospholipids as ant-inflammatory agents. The first day concluded with a solemn announcement of the Thudichum-Award winners The Phospholipid Research Center donated the Thudichum Award for the second time. 25 Messen und Kongresse and biochemist Johann Ludwig Wilhelm Thudichum ( ), who isolated and characterised for the first time numerous compounds of the brain including phospholipids and related species. He recognised the physiological importance of phospholipids. This year, the members of the scientific committee awarded Dr. Tom van Rooij (Erasmus MC, Rotterdam, NL) for his outstanding publications in pharmaceutical phospholipid research. Dr. Tom van Rooij examined in the group of Prof. Dr. Klazina Kooiman ultrasound contrast agents formulated in lipid-coated microbubbles for imaging and therapeutic applications. For the first time the Thudichum Life Award has been awarded. Dr. Herbert Rebmann (Honorary President of the Phospholipid Research Center) congratulated Prof. Dr. Daan Crommelin for his lifelong outstanding achievements in the field of phospholipid research. Prof. Dr. Crommelin is a pioneer in the field of stabilisation and characterisation of phospholipids and liposomes. Congratulations to Prof. Dr. Daan Crommelin and Dr. Tom van Rooij for their excellent scientific research! After a first day of discussion, establishing new contacts but also happy reunions among the participants, we had a wonderful and delicious boat trip with the Königin Silvia on the Neckar river. The second day of the symposium started with a session on Oral Administration of Phospholipids. Prof. Dr. Alexander Treusch (University of Southern Denmark, DK) summarised the latest results on the application of Archaeal Lipids in the Oral Delivery of Therapeutic Peptides, whereas Prof. Dr. Anette Müllertz (University of Copenhagen, DK) elucidated the use of monoacyl-phospholipids in oral self-nanoemulsifying drug delivery systems. A new approach to improve the bioavailability of poor water-soluble drugs with a phospholipid-functionalised calcium carbonate based drug delivery system was presented by Prof. Dr. Jörg Huwyler (University of Basel, CH). Phospholipids are versatile excipients. The working group of Prof. Dr. Yvonne Perrie (University of Strathclyde, GB) demonstrated, inter alia, the versatility of phospholipids as part of the development of lipid-based delivery systems for oral vaccination. More focused on the mechanism of action of phospholipids in the human body was the following session. PD Dr. Andreas Koeberle (University of Jena, DE) presented the inhibitory effect of polyunsaturated phosphatidylcholine on Akt (also known as protein kinase B), as a potential treatment target in cancer therapy. Dr. Jai Prakash (University of Twente, NL) demonstrated a new approach in the field of cancer therapy using special liposomes, comprising carboxylated phospholipids, for a targeting to tumor-associated M2 macrophages. The already mentioned seminars highlighted current innovative phospholipid research and development topics in Europe and North America. What about phospholipid research in South America e.g. in Brazil? The last session of the Phospholipid Symposium 2017 ended with a special seminar on phospholipid research in Brazil. Prof. Dr. Frédéric Frézard (Universidade Federal de Minas Gerais, BR) introduced in main research fields, highlighted impressively current ongoing projects and pointed out promising perspectives. Traditionally, the symposium was concluded by the Young Session chaired by Prof. Dr. Christel Müller-Goymann (University of Braunschweig, DE). The students of six pre-nominated posters were invited to give a short presentation about their research. The following three posters were selected and awarded with a 500 poster prize: Joyce Azzi (Lebanese University, LB): Effect of liposome fatty acyl chains saturation on the photostability of encapsulated biomolecules. Ana Ferreira-Silva (Universidade de Lisboa, PT): Antagonist G-labeled long circulating liposomes actively targeted to lung tumor cells. Phillipp Uhl (University Hospital Heidelberg, DE): Cell penetrating liposomes enable oral delivery of vancomycin. The success of the 5th International Symposium on Phospholipids in Pharmaceutical Research 2017 was based on the many creative minds and inspiring scientists who demonstrated the versatile applications of phospholipids in pharmaceutical research. 26 Messen und Kongresse Highlights of APV/IPEC Europe Excipient Conference 2017 Phil Taylor, IPEC Europe, Belgium The APV/IPEC Europe Excipient Conference 2017 was held on September in Berlin, Germany, providing the usual high-quality insights into regulatory developments in the excipient field and their application. The well-attended event got underway with three parallel workshops on issues that are of particular importance at present in the excipients arena. Christa Farber of Staatl. Gewerbeaufsichstsamt Hannover and Eberhard Kwiatkowski of PharmAdvantageIT covered the topic of data integrity, examining how this concept can be applied to the excipient manufacture and supply chain. Astrid Stockrahm of DFE Pharma tackled quality agreements, showing how IPEC s forthcoming QA guide and template can ease the relationship between excipient supplier and pharmaceutical manufacturer. Rounding out the trio was a workshop on stability testing of excipients by Tanja Natterer of Aug. Hedinger GmbH & Co, which showed how the IPEC Excipient Stability Guide can help in the design of an effective testing programme. Thomas Storm of Novartis kicked off the conference proceedings on the second day, with a presentation on the challenges of multi-compendial compliance for excipients, reviewing the status of pharmacopoeial harmonisation as well as requirement beyond pharmacopoeial compliance. He said that compliance requires continuing efforts in development and routine quality assurance and unfortunately also redundant testing as the large majority of excipient monographs are not harmonised and there are incompatible standards or specifications between regions. The harmonisation effort is a successful process but slow, and industry would like to see more of it, and faster, he said, noting that functionality-related characteristics are becoming more of a focus. The topic of extractables and leachables from excipient storage containers, and their potential as a source of impurities, was tackled by Steven Watt of A&M StabTest GmbH, who noted that while this has been considered in finished drug products there does not appear to be any direct guideline on this as it applies to excipients. He described leachable/extraction studies and workflows used in finished products and described how these concepts could be applied to excipients in storage containers. Jean-Claude Soule of Eli Lilly covered quality audits and how they can be used to ensure excipient supply chain security, reviewing supply chain vulnerabilities, regulatory expectations and guidelines for quality management system audits laid down in ISO 19011:2002. He described the various warning signals that can be looked for during an audit advised on ways to handle a refused audit, concluding that a combination of auditing, testing and pedigree is the best way to reduce risk. He noted that the quality agreement between an excipient supplier and user must describe all the companies involved from raw materials to finished excipient. Speaking on behalf of the IPEC Federation, Frank Milek of Aug. Hedinger GmbH & Co examined the challenges posed by the complexity of the excipient supply chain, focusing on a string of events in which adulterated excipients have resulted in fatalities since the 1990s, the regulatory response to those tragedies, and showing how tools such as the recently-revised IPEC GDP guide can be deployed to ensure the safe supply of excipients. He also described how the IPEC Federation can facilitate that vision. The organization s position as the umbrella association for IPECs in the Americas, Europe, Japan, China and India put it in an important position to achieve this goal, he said, through 26 APV Plus - 04/2017

27 Messen und Kongresse measures such as the development and adoption of harmonised guidance and risk-based global regulatory standards, and serving as a reliable and credible source if information relating to excipients. A regulatory perspective on the implementation of ICH Q3D on elemental impurities which applies to marketed products including new mutual recognition applications of already approved products from December - was delivered by Bruno Spieldenner of the European Directorate on the Quality of Medicines and Healthcare (EDQM). He discussed the implementation of ICH Q3D in the context of the European Pharmacopoeia (Ph. Eur.) general text, chapters, methods and monographs, and reviewed approaches for specific elemental impurities tests, calling for additional expertise and support especially from manufacturers to revise and maintain those tests. He also took some time to review the implementation of the new requirements in the certification procedure, referring to a policy document adopted by EDQM in August. Nikola Matic of Kline & Co broadened out the discussion with a presentation on the global excipients market, and particular those used in oral solid dosage forms which remains the most important sector for excipient suppliers in terms of size and while fragmented had an estimated value of $4bn in 2016, with a volume consumption of around 641,000 tonnes. The market is expected to grow at a healthy 5.6% rate to 2021, driven by an expanding overall pharma and particularly the generic drug market, the increasing complexity and low bioavailability of new active pharmaceutical ingredients (APIs) that need functional excipients, and the development of improved excipient grades for new applications. Johann Philipp Hebestreit of BASF began the technical part of the conference programme with a presentation on preventing alcohol-induced dose dumping (ADD), the problem of unintended and excessive release of a pharmacologically-active ingredient as a result of alcohol consumption, which is known to be an issue with some sustainedrelease formulations. With the use of case studies, he showed how formulations can be at risk of ADD, and described how it can be avoided through the use of excipients such as BASF s polymer-based Kollidon SR. Laura de Miguel of Omya International took on another technical topic, looking at filler-binder excipients for direct compression and particularly co-processed excipients new or novel combinations of established excipients that in this setting can improve characteristics such as compactability, flowability and friability. With regulatory pathways to approval of novel excipients challenging, co-processed excipients help improve finished product characteristics, delivering a marketing advantage and potentially extending a product s life cycle. Excipients are also an important enabler of transdermal drug delivery systems (TDDS), and Sebastian Braun of Tesa Labtec GmbH broke down the various components of a TDDS, and showed how excipients fit into the structure of these products. He also looked at the documentation requirements involved if a drug master file (DMF) is available or not, and discussed the impact of the EU guideline on formalised excipient risk assessment in the context of TDDS development. The technical programme also included a presentation by Kofi Asare-Addo from the University of Huddersfield School of Applied Sciences on advanced imaging in pharmaceutical materials, covering technologies such as infinite focus microscopy and how they can be applied to the design of robust formulations for drug delivery. Finally, Mats Hertel of Christian-Albrechts-Universitat Pharmazeutisches Institut looked at the use of excipients as drug carriers for pulmonary drug delivery, including the importance of matching the carrier to the device and optimal blending practices. APV/FIP Joint-Workshop in Nuremberg Prof. Dr. Sandra Klein, University of Greifswald, Germany Dissolution testing is an important tool from early development to life cycle management of a drug product. With the aim to give an update on the application of dissolution testing in assessing critical formulation attributes with regard to bioavailability of the respective drug product, on September 26-27, 2017 on the initiative of Dr. Horst- Dieter Friedel who is co-chair of the FIP Focus Group on Dissolution/In Vitro Drug Release APV and FIP co-organized a Joint-Workshop on Oral Biopharmaceutic Tools in Nuremberg. The workshop was planned by Prof. Dr. Jennifer Dressman, the chair of the APV Focus Group on Biopharmaceutics and Pharmacokinetics and Prof. Dr. Sandra Klein, representing APV, and, Prof. Dr. Christos Reppas and Dr. Horst-Dieter Friedel from the FIP Focus Group on Dissolution/In Vitro Drug Release and had a total of 57 attendees. Day 1 of the workshop was dedicated to the presentation of new approaches in biorelevant/biopredictive in vitro models. After a general introduction by Dr. Horst-Dieter Friedel, Bayer AG, Berlin, the scientific lectures started with a presentation given by Dr. Cord Andreas from the University of Frankfurt. He introduced the biopharmaceutics risk assessment roadmap (BioRAM) concept which is intended to optimize drug product development and performance by using therapy-driven target drug delivery profiles as a framework to achieve the desired therapeutic outcome. The BioRAM considers the therapeutic target with the drug substance characteristics and enables collection of critical knowledge for development of a dosage form that can perform consistently for meeting the patient's needs. The presentation highlighted, how the key factors are identified and how in vitro, in vivo, and in silico modeling and simulation techniques are used to elucidate the optimal drug delivery rate and pattern. The next speaker was Dr. Uwe Münster from Bayer AG, Germany who gave an overview of the goals and activities of the OrBiTo project, an IMI initiative dedicated to better understand oral drug absorption and to use this knowledge for developing new biopharmaceutics in vitro and in silico tools that perspectively should help to reduce the number of preclinical and clinical studies required for new drug formulations, but also could be applied in a regulatory framework, e.g. in the registration process of generic products. He presented results from work performed in the different workpackages of the project and also discussed future objectives. Prof. Dr. Peter Langguth from the University of Mainz then presented on how to establish an in vitro in vivo relationship (IVIVR) or -correlation (IVIVC). He introduced various strategies for designing predictive in vitro test methods and showed their implementation by showing results from different case studies. He also nicely presented the chances and pitfalls with different in vitro models and clearly pointed out that a mechanistic understanding of the in vivo behavior of the dosage form is essential for obtaining an IVIVC for a given drug formulation. In the final part of his presentation he presented results from a survey performed in the OrBiTo project that was performed to get an overview of how IVIVC is established in industry and what are the biggest hurdles in trying to establish biopredictive in vitro models. He concluded that the chances of a successful IVIVC increase with the application of in vivo bio-predictive dissolution methodologies and that alternative approaches like physiologically-based pharmacokinetic (PBPK) modeling would allow successful IVIVC development for drugs with more complicated absorption/disposition behavior. The next presentation was given by Prof. Dr. Christos Reppas from the University of Athens, Greece who first discussed the different interpretations of what is a biorelevant dissolution method and then in detail introduced the different physiological and dosage form related parameters to be addressed in a biorelevant dissolution experiment. He explained in detail the different levels of simulation of luminal composition and how much complexity is required for adequately screening a certain formulation. He connected this question with the developability classification system (DCS) and highlighted also the limits when the aim is to predict drug solubilities in human aspirates based on results obtained in solubility experiments with biorelevant media. 28 APV Plus - 04/2017

29 Messen und Kongresse The last presentation of the first day was given by Dr. Kerstin Frank from Boehringer Ingelheim, Germany. She talked about dissolution methodologies used in preclinical testing and how these can be guided by API properties, results from other early stage preclinical tests and dosage form characteristics. Major criteria discussed were the selection of different apparatus or media types. She also presented two case studies where several of the discussed dissolution models were applied to prototype formulations containing BCS class II compounds and concluded that in early stage drug development, a close interdisciplinary cooperation is needed and that it is further important to identify critical drug/formulation attributes as early as possible to be able to develop the right in vitro test methods. The second day of the workshop started with a presentation from David Turner from Certara, UK. He gave a comprehensive introduction in the use of PBPK models and detailed information of how different PBPK models are designed, how they have advanced over the last years by implementing physiological data of different populations, intraluminal fluid dynamics, transporter expression along the GI tract etc. and finally, how they can be applied to obtain a physiologically-based IVIVC. Dr. Cord Andreas then gave a talk on the prediction of in vivo performance from modified release (MR) dosage forms using biorelevant dissolution. With results from a case study on the examination of drug release of MR formulation containing a BCS class II compound with compendial and biorelevant test methods, he could clearly show the importance of properly addressing the changing environment in the different sections of the GI tract when the aim is to obtain an IVIVC for such a formulation. In another case example, he nicely showed how in vitro and in silico approaches can be combined to obtain a meaningful IVIVC for a highly soluble drug. Dr. David Sperry from Eli Lilly, USA then presented an industrial view on the prediction of in vivo performance using biorelevant dissolution. He gave an insight into the application of biorelevant media and models in the pharmaceutical industry. Cases examples included the application of two-step dissolution models for poorly soluble weak bases, the application of the Focused Beam Reflectance Measurement (FBRM) to track particles in the dissolution vessel for better understanding the dissolution process and the main determinants of dissolution rate after disintegration of a solid dosage form. He also highlighted the importance of the composition of the dissolution medium when the aim is to establish an IVIVC and discussed both the chances and limits of in-house models and commercially available in vitro and in silico PBPK software for predicting in vivo release performance of solid oral dosage forms. At the end of his presentation, he presented an interesting work flow applied for bioavailability risk assessment and underscored the importance of the detailed information that is needed on product performance in such a procedure. The session on novel oral biopharmaceutics tools was completed by a presentation of Dr. Sheila Annie Peters from Merck KGaA, Germany. She presented industrial case examples on PBPK modeling. A focus of her presentation was how to verify PBPK models to establish confidence and how to perform a sensitivity analysis and an uncertainty analysis to demonstrate robustness of the model. In agreement with the statements made in previous talks, she pointed out that the proper verification of a model requires a detailed mechanistic understanding of dosage form performance and that currently there is still a big gap in knowledge in the complex interplay of parameters that are involved the essential processes determining the plasma profile of a drug after its oral administration. The second session of day 2 of the workshop was dedicated to quality control (QC) applications. Dr. Anita Nair form Merck KGaA, Germany was the first speaker of this session and presented Quality by Design (QbD) applications of drug critical process parameters. She explained the elements of QbD starting with the quality target product profile for a product, through identifying critical quality attributes, critical material and process attributes to developing a control strategy by defining specifications for materials, product and unit processes that would ensure quality of the product. She highlighted what role properly specified dissolution test methods play in this context when they are regarded as critical quality attributes. Dr. Lorenz Liesum, Novartis, Switzerland then reported of how Process Analytical Technology (PAT) tools and surrogate model development can be implemented in the pharmaceutical industry and how this fits with the FDA vision of control strategy. He clearly pointed out critical factors with regard to drug and excipient properties and manufacturing process that can be of either well-known or random origin and can impact dissolution of the drug product. Moreover, he presented an interesting case example in which after applying a specific validated mathematical model dissolution of a given product was nicely predicted by using critical process attributes. 30 Messen und Kongresse also clearly pointed out the preconditions for using such models in a prospective manner and showed up the limits for their application. Prof. Dr. Christos Reppas closed the session on QC applications with a talk on linking QC methodology with biowaivers and bridging studies. A particular focus of this presentation was set on dissolution test conditions to be applied for a BCS-based biowaiver and how to translate the BCS biowaiver test method into a QC method. The last session of the workshop was dedicated to regulatory opportunities and the objective was to provide global perspectives on the role of dissolution testing on product design, control and approval. Dr. Arzu Selen, FDA, USA was the first speaker and presented the FDA perspective on how in vitro and in vivo drug product performance can be connected. She discussed the role that dissolution testing has played in drug product development and QC and highlighted the progress, dissolution methodologies have undergone to become more and more biopredictive. She further discussed the potential impact of predictive methods and presented possible approaches for clinically relevant specifications for drug dissolution/release and spent also some time on discussing the BioRAM strategy as an integrated systems approach to drug development. She concluded that advanced dissolution testing can impact candidate selection, drug product design and development as well as drug product life cycle management and also pointed out that a proper mechanistic understanding is essential for developing such models and also pointed out that novel dissolution testing approaches, novel systems approaches and utilization of IVIVC and IVIVR are highly encouraged. Dr. J. Michael Morris, formerly HPRA, Ireland shared the EU perspective on the use of dissolution tests. He also stated that dissolution testing is a useful development tool to provide information for formulation development and manufacture process optimization and that a properly designed dissolution method can be a surrogate for in vivo bioequivalence. The focus of his presentation was set at on pharmacopoeial requirements and information provided in ICH Q6A and Q4B guidelines and he also discussed the requirements for dissolution test methods for novel dosage forms. The final presentation was given by Dr. Tomokazu Tajiri, Astellas, Japan who presented the Japanese perspectives on the role of dissolution testing in development. He discussed the harmonization of dissolution methodologies in the Japanese (JP), the United States (USP) and the European Pharmacopoeia (EP), but also pointed out that there are still some significant differences between JP and USP & EP. He also highlighted that for dissolution test design, particularly for experiments simulating gastric conditions, the high prevalance of achlorhydria in Japan is currently an important parameter to consider when designing dissolution methods. The fact of the prevalence of achlorhydria in the Japanese population is also reflected in the recommendation to not use enzymes in dissolution media since at higher gastric ph, the conversion of pepsinogen into pepsin will not be possible. A marked difference can also be observed in the number of dissolution test experiments to be performed at various test conditions that are required for registering a drug product. In addition to this, to date, the biowaiver concept is not applied in Japan and also the regulatory authorities do not accept IVIVC approaches. In summary, the workshop highlighted the importance of oral biopharmaceutics tools in the development of safe and effective oral dosage forms. The presentations underlined the progress in vitro dissolution testing and in silico modeling has made during the past decades and provided the participants with a deep insight into the development of physiologically-based in vitro and in silico models and the importance of interdisciplinary approaches. It was also of great importance to hear the regulatory viewpoints in Europe, Japan and the United States and that there is definitely a chance for a paradigm shift. The participants made use of their opportunity to discuss with all the speakers and hopefully got a lot of new ideas to be implemented in terms of improve the predictivity and robustness of current in vitro and in silico models. 30 APV Plus - 04/2017

31 Messen und Kongresse 10 th Conference of the European Paediatric Formulation Initiative Smita Salunke, PhD, UCL School of Pharmacy, EuPFI, London, United Kingdom Marking the 10th anniversary of the Paediatric Regulation and foundation of the European Paediatric Formulation Initiative (EuPFI) consortium, EuPFI convoked its 9th annual conference with the International Association of Pharmaceutical Technology (APV) on 20th and 21st September 2017 at the Biological and Chemical Research Centre, University of Warsaw, Poland. The participation of almost 180 delegates from Industry, Academia, Regulatory and other organizations from 26 countries with diverse backgrounds and involvement in both formulation development and regulatory submissions provided a productive setting to reflect back on the past decade of paediatric formulations, to celebrate accomplishments, to welcome new researchers and enrich networking opportunities and to consider how novel and emerging technologies will yield further insights into what the future may bring. The two pre-conference workshops provided the much needed focus and hands on involvement to both experienced professionals and students. 1) A taster session on designing taste- masked oral paediatric formulation prototypes and how to plan their palatability assessment moderated by Joanne Bennett, Catherine Tuleu, Jenny Walsh, Anne-Marie Pensé-Lhéritier, and Smita Salunke, 2) Biopharmaceutics evaluation of food effects in paediatric medicines development for products co- administered with food moderated by Hannah Batchelor, Sandra Klein and Henry Yunhui Wu We went on a journey of the last 10 years. Started with a moving film from the Goodman foundation, Dr. Catherine Litalien reminded the delegates of the very reason why they were all there and that working together we can make a difference. Dr Sandrine Chaippini (EMA) took us through what the EU regulation has achieved, for example more mindfulness around excipients especially colours. Dr. Nasir Hussain highlighted the room for improvement with the issues around acceptability not tackled appropriately, despite the regulatory ask. The lack of progress for older drugs and that despite the testimony of 1 PUMA success story (INFRACORT) was highlighted by Dr. Martin Whitaker. We also heard other success stories from Dr. Jörg Breitkreutz and Dr. Dirk Leutner round the improvement of compounding at EU level (PaedForm). The Japanese delegates Tomonori Shiokawa and Hidemasa Nagai proved in their effort to encourage cooperation in between EU and Japan. Moving slowly into the future, Dr Mohamed Albed Alhnan highlighted the applicability of 3D printing: the future of patient centric manufactured medicine and clinical phase appropriate products. The Interesting and engaging talks in the focus sessions were emanated from each EuPFI workstream (Excipients, Taste assessment and Taste Masking, Administration devices, Age appropriateness of formulations, and Biopharmaceutics). The speakers included: Rajneesh Taneja, Dejan-Krešimir Bučar, Jeremy.A. Bartlett, Andrew Farrington, Jim Ridings, Gary inwards, Jens Van Den Abeele, Henry Yunhui, Wu, Lee Immins, Trevor Riley and Sébastien Morin. The innovation show case session was presented by Carmen Popescu from Roquette and Corinna Fischer from Balda Medical GmbH & Co. KG. The soap box session had excellent contributions from Justyna Katarzyna Czarnocka, Stephanie Keser, Lisa Freerks, Bastian Hahn, Mariagiovanna Scarpa, Claire Lewis, Tânia Fernandes, Deborah Pasko and Erik Wollmer. Once again this was a very successful meeting and we hope that by bringing together those with insights and expertise from a range of relevant perspectives, and by sharing good practice, this conference has made a contribution to stimulating research into addressing the challenges in paediatric formulation development and will lead to further progress in this area. We would like to thank all of you as attendees for your active contributions to the discussions in this meeting. A special issue with selected presentations from the conference, editorial and abstracts will be published in International Journal of Pharmaceutics in the coming months. The speakers at the conference highlighted the importance of both appreciating how far we have come in the last 10 years but also how much further we still have to go to really combat the problem of formulating better medicines for children and to be more global, encompassing developing countries and their needs. So, we are counting on you to continue sharing your thinking about the many possible scenarios that could be developed, and how to best prepare for the next 10 years.
