4 4 Produktbeschreibung Der PathoTrack MNV ELISA von GVG Diagnostics ist ein indirekter Enzyme Linked Immunosorbent Assay (ELISA) für den schnellen und zuverlässigen Nachweis von Antikörpern gegen das murine Norovirus (MNV) in Serumproben von Mäusen. Der Test ist hochspezifisch, hochsensitiv und schnell! Der PathoTrack MNV ELISA ist mit jeweils mehr als 500 MNV-positven und -negativen Feldseren validiert (Daten auf Anfrage). Sensitivität und Selektivität sind höher als 99 Prozent. Das murine Norovirus ist der Erreger mit der höchsten bekannten Prävalenz innerhalb von Maushaltungen. MNV ist ein unbehülltes, einzelsträngiges RNA-Virus aus der Familie der. Das Virus ist hochansteckend und wird fäkal-oral übertragen. Aufgrund der enorm hohen Infektionsrate werden die Diagnostik und das Monitoring von MNV-Infektionen von der FELASA (Federation of European Laboratory Animal Science Association) vierteljährlich empfohlen. 6 6 Testprinzip Die PathoTrack MNV ELISA-Mikrotiterplatte (96 wells) ist mit einem MNV-spezifischen Antigen beschichtet. A. In einem ersten Schritt binden spezifische Antikörper aus der Serumprobe an das Antigen, ungebundenes Material wird durch Waschen entfernt. B. Anschließend wird ein Anti-Maus-Peroxidase-Konjugat zugegeben. Dieses erkennt Immunglobulin G- und Immunglobulin M-Antikörper (IgG, IgM) aus der Mausprobe und bindet an sie. Ungebundene Konjugatantikörper werden durch Waschen entfernt. C. Im Folgenden wird das Substrat (TMB) zugegeben, welches durch das Peroxidase-Konjugat zu einem blauen Farbstoff umgesetzt wird. D. Die Reaktion wird mit Schwefelsäure abgestoppt. Dabei schlägt die Farbe der Lösung von blau nach gelb um. Die Absorption der Lösung wird bei 450 nm in einem Photometer bestimmt und verhält sich proportional zur Anzahl der MNV-spezifischen Antikörper in der Probe. Testprinzip PathoTrack MNV ELISA

9 Nutzungshinweise Vermeiden Sie eine Kontamination der einzelnen Komponenten. Schütten Sie das benötigte Volumen der Lösungen direkt in die Vorlegewannen. Verwerfen Sie nicht genutzte Lösungen. Vermischen Sie keine Bestandteile des PathoTrack MNV ELISA Kits mit Komponenten anderer ELISA Kits. Der PathoTrack MNV ELISA ist für die Nutzung von destilliertem Wasser aus Ionenaustauscheranlagen (VE-H 2 O) geeignet. Insofern in Ihrem Labor vorhanden, wird die Nutzung von hochreinem bidestillierten Wasser oder Reinstwasser (Milli-Q o.ä.) empfohlen. Verwenden Sie kein Leitungswasser! Benutzen Sie keine Bestandteile des Kits nach Ablauf der Haltbarkeit. Das Verfallsdatum finden Sie auf dem Packungsetikett und den Reagenzien. Voraussetzung für die Präzision der Messergebnisse ist die Verwendung sauberer Glasmaterialien, das sorgfältige Pipettieren und die Einhaltung des Protokolls unter Beachtung der Nutzungshinweise. 10 10 Vor dem Start Bringen Sie alle Lösungen und Reagenzien unmittelbar vor Benutzung auf Raumtemperatur (18 25 C). Eine zu hohe oder zu niedrige Temperatur der Lösungen kann die Testresultate beeinflussen. Verdünnen Sie den W Wash Buffer (10 x) 1:10 mit destilliertem Wasser. Für die Herstellung von z. B. 200 ml verdünntem W Wash Buffer (1 x), geben Sie 20 ml W Wash Buffer (10 x) zu 180 ml destilliertem Wasser. Verdünnen Sie die Serumproben 1:50 in S Sample Buffer. Für die Herstellung von 150 µl Serumverdün nung pipettieren Sie 3 µl Serum in 147 µl S Sample Buffer. Für die Verdünnung des Serums können Sie 0,5 ml oder 1,5 ml Einwegreaktionsgefäße sowie unbeschichtete Vorverdünnungsplatten verwenden. Mischen Sie gut. Vor dem Start PathoTrack MNV ELISA

11 Arbeiten Sie zügig. Die Verwendung von Mehrkanalpipetten wird empfohlen. Setzen Sie den PathoTrack MNV ELISA keiner direkten Sonneneinstrahlung oder starken Lichtquellen aus. Pipettieren Sie jeweils 100 µl der gebrauchsfertigen PC Positive Control und NC Negative Control sowie der verdünnten Serumproben in die wells der Mikrotiterplatte. 2. Decken Sie die Platte ab und inkubieren Sie diese 45 Minuten bei Raumtemperatur. Entfernen Sie die Lösung durch Absaugen oder Ausklopfen. 3. Waschen Sie die Platte 3 x mit je 300 µl W Wash Buffer (1 x). Klopfen Sie die Platte vorsichtig auf einem Einwegtuch trocken. 4. Pipettieren Sie 100 µl 1 Conjugate Solution in jedes well, decken Sie die Platte ab. 5. Inkubieren Sie die Platte 30 Minuten bei Raumtemperatur. Entfernen Sie die Lösung durch Absaugen oder Ausklopfen. 6. Waschen Sie 3 x mit je 300 µl W Wash Buffer (1 x). Klopfen Sie die Platte vorsichtig auf einem Einwegtuch trocken. 13 7. Pipettieren Sie 100 µl 2 Substrate Solution in jedes well und decken Sie die Platte ab. 8. Inkubieren Sie die Platte 15 Minuten bei Raumtemperatur. 9. Stoppen Sie die Reaktion durch die Zugabe von 100 µl 3 Stop Solution. 10. Messen Sie die Absorption (OD) bei einer Wellenlänge von 450 nm innerhalb von 20 Minuten in einem Spektrophotometer. 14 14 Auswertung Validierung des Tests Um die Testergebnisse auswerten zu können, müssen folgende Kriterien erfüllt werden: Der Absorptionswert der 1,0 sein. Der Absorptionswert der 0,3 sein. Positive Control muss Negative Control muss Sollten diese Kriterien nicht erfüllt sein, muss der Test wiederholt werden. 15 Interpretation der Ergebnisse Für die Auswertung des PathoTrack MNV ELISAs werden die Mess werte OD Probe zunächst auf die Kontrollen PC und NC normiert. Benutzen Sie dafür folgende Formel: OD Probe OD NC OD % = x 100 % OD PC OD NC Proben mit OD % > 10 % sind als positiv zu bewerten. Antikörper gegen MNV wurden nachgewiesen. Proben mit OD % 10 % sind als negativ zu bewerten. Antikörper gegen MNV wurden nicht nachgewiesen. Tipp: Eine automatische Normierung der Messwerte ist in den gängigen Spektrophotometern möglich. 22 22 Product description PathoTrack MNV ELISA from GVG Diagnostics is an indirect enzyme-linked immunosorbent assay (ELISA) for the rapid, reliable detection of antibodies to murine norovirus (MNV) in serum samples from mice. The test is highly specific, highly sensitive and fast! PathoTrack MNV ELISA has been validated with over 500 MNV-positive and 500 MNV-negative field sera (data on request). Its sensitivity and selectivity exceed 99 %. Murine norovirus is the pathogen with the highest known prevalence within stocks of mice. MNV is a non-enveloped, single-stranded RNA virus from the Caliciviridae family. The virus is highly infectious and is transmitted by the faecal-oral route. Due to the extremely high infection rate, the quarterly diagnosis and monitoring of MNV infections is recommended by FELASA (the Federation of European Laboratory Animal Science Associations). 24 24 Test principle The PathoTrack MNV ELISA microplate (96 wells) is coated with an MNV-specific antigen. A. In the first step, specific antibodies from the serum sample bind to the antigen. Unbound material is removed by washing. B. Then an anti-mouse peroxidase conjugate is added, which recognizes immunoglobulin G and immunoglobulin M antibodies (IgG, IgM) from the mouse sample and binds to them. Unbound conjugate antibodies are removed by washing. C. The substrate (TMB) is then added, which is converted by the peroxidase conjugate into a blue dye. D. The reaction is stopped with sulphuric acid. The colour of the solution changes from blue to yellow. The absorbance of the solution is measured in a photometer at 450 nm. 27 General precautions Avoid contamination of the individual components. Pour the required volume of the solutions directly into the serving trays. Discard unused solutions. Do not mix any components of PathoTrack MNV ELISA kit with components from other ELISA kits. PathoTrack MNV ELISA is suitable for the use of water from ion exchange filters (deionized water). Using highly pure double-distilled water or ultrapure water (Milli-Q or equivalent) is recommended if available in your laboratory. Do not use tap water. Do not use any kit components after their expiry date. The expiry date can be found on the label and reagents. Clean glassware, careful pipetting, adherence to the protocol and following the user instructions are essential for precise results. PathoTrack MNV ELISA General precautions 27

28 28 Before starting Before use bring all solutions and reagents immediately to room temperature (18 25 C). The test results may be impaired if the solutions are too warm or too cold. Dilute the W Wash Buffer (10 x) 1:10 with distilled water. To produce for example 200 ml diluted W Wash Buffer (1 x), add 20 ml W Wash Buffer (10 x) to 180 ml distilled water. Dilute the serum samples 1:50 in S Sample Buffer. To produce 150 µl serum dilution, pipette 3 µl serum into 147 µl S Sample Buffer. Use 0.5 ml or 1.5 ml disposable reaction vessels or uncoated pre-dilution plates to dilute the serum. Mix well. Before starting PathoTrack MNV ELISA

29 Work quickly. The use of multichannel pipettes is recommended. Do not expose PathoTrack MNV ELISA to direct sunlight or strong light sources. PC Positive Control and NC Negative Control are ready to use and should not be diluted. PathoTrack MNV ELISA Before starting 29

30 30 PathoTrack ELISA protocol 1. Pipette 100 µl each of the ready-to-use PC Positive Control and NC Negative Control as well as the diluted serum samples into the wells of the microplate. 2. Cover the microplate and incubate it for 45 minutes at room temperature. Remove the solution by aspiration or tapping. 3. Wash the plate with 300 µ1 W Wash Buffer (1 x) three times. Gently knock the plate dry on a disposable cloth. 4. Pipette 100 µl 1 Conjugate Solution into each well and cover up the plate. 5. Incubate the plate at room temperature for 30 minutes. Remove the solution by aspiration or tapping. 6. Wash with 300 µl W Wash Buffer (1 x) three times. Gently knock the plate dry on a disposable cloth. PathoTrack ELISA protocol PathoTrack MNV ELISA

31 7. Pipette 100 µl 2 Substrate Solution into each well and cover the plate. 8. Incubate the plate for 15 minutes at room temperature. 9. Stop the reaction by adding 100 µi 3 Stop Solution. 10. 33 Calculation and interpretation of the results To evaluate PathoTrack MNV ELISA, the OD sample measurements must first be normalized to the controls PC and NC. Use this formula: OD Sample OD NC OD % = x 100 % OD PC OD NC Samples in which OD % > 10 % are considered positive. Antibodies to MNV have been detected. Samples in which OD % 10 % are considered negative. Antibodies to MNV have not been detected. Tip: Obtained OD-values can be automatically normalized in most spectrophotometers.
