2 ABSTRACT Learning acquisition of new information, and memory dynamic storage of this information are among the most fundamental cognitive functions of the brain. Unraveling the workings of these processes is essential not only for a better understanding of how we experience our environment, but also to generate new means of treating devastating neuropsychiatric diseases such as dementias. As we learn and create memories, the brain undergoes through a lot of changes at cellular and molecular level. The most prominent change that marks the storage of memory over a long time scale is enhanced synaptic communication between brain cells. This by itself is brought by a highly orchestrated series of molecular signaling. At first, transient protein modifications, such as phosphorylation and dephosphorylation are sufficient to alter the localization and activity of synaptic proteins, and synaptic communication. But, long-lasting memory require gene expression and synthesis of new gene products, i.e. RNAs and proteins, that function as structural or functional components of synapses, or as regulatory molecules. Gene expression is regulated at several levels and through various mechanisms. One of these mechanisms, which is a subject of this thesis, is epigenetics alteration in gene expression due to chromatin modifications, without changes in DNA sequences. The goal of this thesis is to study the extent to which epigenetic changes, specifically histone post-translational modifications and micro-rnas are involved in memory formation, and how they are regulated by a common signaling molecule, namely Protein Phosphatase 1. In the first study, we find that learning induces a number of histone modifications in the mouse Hippocampus, which subside with time. In contrast, the same modifications become more prominent in Prefrontal Cortex as the memory matures. We also find that inhibiting Protein Phosphatase 1 accentuates these changes. The results reinforce our previous knowledge on the involvement of epigenetic changes in memory, and additionally suggest that epigenetics can coordinate the transfer of a memory trace from the hippocampus to the cortex. In the second study, we identified micro- RNAs that are differentially regulated after learning, including the mirna cluster mir- 183/96/182. Artificially overexpressing this cluster in the mouse hippocampus improves memory and prevents expression of genes that suppress synaptic plasticity. In addition, the biogenesis of these micro-rnas is significantly enhanced upon inhibition of Protein 9

3 Abstract Phosphatase 1. In sum, the work described here reveals a layer of epigenetic regulation that accompanies memory formation and persistence. It also provides a novel evidence for the regulation of neuronal micro-rna biogenesis by a Phosphatase. Finally, it highlights the importance of mir-183/96/182 cluster in memory formation, providing a foundation to explore the potential of these micro-rnas in memory modification in the future. 10

4 Zusammenfassung ZUSAMMENFASSUNG Lernen und Gedächtnis, der Erwerb neuer Informationen und ihre dynamische Speicherung, gehören zu den grundlegenden kognitiven Funktionen des Gehirns. Zu klären wie diese Prozesse funktionieren, ist wichtig, um besser zu verstehen, wie wir unsere Umwelt erleben, aber auch, um neue Behandlungsmethoden für neuropsychiatrische Erkrankungen wie Demenz zu finden. Während wir lernen und Erinnerungen generieren, geschehen im Gehirn viele Veränderungen auf zellulärer und molekularer Ebene. Die auffälligste Veränderung, die der Speicherung von Erinnerungen über lange Zeit zugrunde liegt, ist eine verbesserte synaptische Kommunikation zwischen Gehirnzellen, die wiederum durch eine hoch orchestrierte Serie molekularer Signalwege entsteht. Zunächst reichen vorübergehende Proteinveränderungen aus, wie etwa Phosphorylierung und Dephosphorylierung, um Ort und Aktivität synaptischer Proteine sowie synaptische Kommunikation zu verändern. Die langfristige Speicherung erfordert jedoch Genexpression und die Synthese neuer Genprodukte, also RNAs und Proteine, die als strukturelle oder funktionale Komponenten von Synapsen oder als regulatorische Moleküle fungieren. Die Genexpression wird auf mehreren Ebenen und durch verschiedene Mechanismen reguliert. Einer dieser Mechanismen und Gegenstand dieser Arbeit ist Epigenetik, die Veränderung der Genexpression durch Modifikationen des Chromatins, ohne die DNA-Sequenz zu verändern. Ziel dieser Arbeit ist zu untersuchen, in welchem Ausmass epigenetische Veränderungen, vor allem post-translationale Histonmodifikationen und micro-rnas, bei der Bildung des Gedächtnisses eine Rolle spielen und wie sie durch das wichtige und allgegenwärtige Signalmolekül Proteinphosphatase 1 reguliert werden. In der ersten Studie wurde gezeigt, dass Lernen Histonmodifikationen im Hippocampus der Maus hervorruft, die mit der Zeit wieder nachlassen. Im Gegensatz dazu werden die gleichen Veränderungen im präfrontalen Kortex mit der Zeit stärker. Es wurde auch gefunden, dass eine Hemmung von Proteinphosphatase 1 diese Veränderungen akzentuiert. Diese Ergebnisse untermauern bisheriges Wissen zum Einfluss epigenischer Veränderungen auf das Gedächtnis und lassen darüber hinaus darauf schliessen, dass Epigenetik den Transfer von Gedächtnisspuren vom Hippocampus zum Cortex koordinieren kann. In der zweiten Studie wurden micro-rnas identifiziert, die nach dem Lernen unterschiedlich reguliert werden, darunter das micro- RNA-Cluster mir-183/96/182. Dieses Cluster im Hippocampus der Maus künstlich zu 11

5 Zusammenfassung überexprimieren, verbessert das Gedächtnis und verhindert die Expression von Genen, die synaptische Plastizität unterdrücken. Ausserdem wird die Biogenese dieser micro-rnas signifikant durch die Hemmung von Proteinphosphatase 1 verstärkt. Zusammenfassend zeigt diese Arbeit eine Ebene epigenetischer Regulation, welche die Bildung und Persistenz des Gedächtnisses begleitet. Sie liefert zudem einen neuen Nachweis, dass Phosphatase die Biogenese neuronaler micro-rna reguliert. Zuletzt betont diese Arbeit die Bedeutung von mir-183/96/182 bei der Gedächtnisbildung und legt damit die Basis, um zukünftig das Potenzial dieser micro-rnas bei der Veränderung des Gedächtnisses zu untersuchen. 12
