3 Summary The ribosome is a universally conserved ribonucleoprotein complex that translates the genetic message encoded on mrnas into proteins. The tasks of message decoding and peptide bond formation are carried out by two subunits of unequal size the large and the small ribosomal subunit. Even though universally conserved, the ribosome has undergone compositional and structural changes throughoutevolution.thecryo electronmicroscopy(cryo EM)structureofthe50S largeribosomalsubunitfromamethanogenicarchaeondescribedinthefirstpartof this thesis visualizes examples of such structural change in the domain Archaea. Additionally,thestructurerevealsthehighconservationofthebindingofaprotein factor involved in biosynthesis of ribosomes and translation control, termed initiationfactor6,tothelargeribosomalsubunit. Eukaryotic ribosomes are the most complex of all ribosomes. Their biogenesis theproductionofribosomesbythecell requiresalargenumberofnon ribosomal proteins that function in production, folding, and assembly of ribosomal components, as well as the nuclear export, maturation, and quality control of the pre ribosomalparticles. Arx1, Rei1, and Jjj1, a group of eukaryotic 60S ribosome biogenesis factors, has beensuggestedtobindtothepre 60Sparticlenearthepolypeptidetunnelexit.Arx1 facilitates the export of the pre 60S particle from the nucleus into the cytoplasm. After completion of pre 60S particle export, Arx1 has to be released from the pre 60S subunit. Rei1 and Jjj1 have been implicated in this process, which additionally involvestheatpasessa. In the second part of this thesis, complexes of Arx1, Rei1, and Jjj1 with the 60S ribosomal subunit were reconstituted from highly purified 60S subunits and recombinantly expressed proteins. Based on the finding that Rei1 significantly stabilizes the 60S Arx1 interaction, the structures of the 60S Arx1 Rei1, 60S Arx1 Rei1 Jjj1,and60S Rei1complexesweredeterminedbysingle particlecryo EM. Thestructureofthe60S Arx1 Rei1complexcouldberefinedtoapproximately8Å resolution,allowingtheunambiguousassignmentoftheorientationofboundarx1 on the ribosomal subunit, and a detailed interpretation of the molecular contacts. The structure shows that Arx1 is sandwiched between the polypeptide tunnel exit regionandaeukaryotic specificrrnaexpansionsegment.thisbindingsitesuggests that Arx1 serves to protect this functionally important part of the ribosome during ribosome biogenesis. Rei1 and Jjj1 were visualized in immediate proximity of Arx1 andrei1,inagreementwiththeideathatbothrei1andjjj1functionintherelease ofarx1fromthenascentsubunit. Taken together, the work in this thesis has provided novel insights into the biogenesis,evolution,andregulationofthelargeribosomalsubunit. XI

Obschon das Ribosom universell konserviert ist, war es während der Evolution Veränderungen seiner Struktur und Zusammensetzung unterworfen. Der erste Teil dieser Dissertation beschreibt die kryo elektronenmikroskopische (Kryo EM) Struktur der grossen50suntereinheitauseinemmethanogenenarchaebakterium,welchesolche evolutionärenveränderungenderribosomenstrukturbeispielhaftzeigt.diestruktur zeigt zudem die hoch konservierte Bindung des Initiationsfaktors 6, der an der BiosynthesedesRibosomsundanderTranslationsregulationbeteiligtist,andie50S Untereinheit. EukaryotischeRibosomensinddiekomplexestenallerRibosomen.IhreBiogenese die Produktion von Ribosomen durch die Zelle erfordert eine grosse Anzahl an nicht ribosomalen Proteinfaktoren. Diese sind an der Produktion, der Faltung und dem Zusammenbau der ribosomalen Komponenten sowie dem nukleozytoplasmatischen Export, der finalen Reifung, und der Qualitätskontrolle der ribosomalenpartikelbeteiligt. Arx1, Rei1, und Jjj1 bilden eine Gruppe von eukaryotischen 60S Biogenese Faktoren, von der vermutet wird, dass sie in der Nähe des Polypeptid Tunnelausgangsandiepre 60SUntereinheitbinden.Arx1unterstütztdenExportdes pre 60S PartikelsausdemZellkerninsZytoplasma.NacherfolgtemExportmussArx1 vonderribosomalenuntereinheitabgelöstwerden.eswirdvermutet,dassrei1und Jjj1indiesemProzess,derzudemdieSsa ATPasebenötigt,funktionieren. ImzweitenTeildieserDissertationwurdenKomplexevonArx1,Rei1undJjj1mit der 60S Untereinheit aus aufgereinigten 60S Untereinheiten und rekombinant exprimierten Proteinen rekonstituiert. Auf Basis der Beobachtung, dass Rei1 die Interaktion zwischen Arx1 und der 60S Untereinheit stark stabilisiert, wurden die Strukturen der 60S Arx1 Rei1, 60S Arx1 Rei1 Jjj1, und 60S Rei1 Komplexe mittels Einzelpartikel Kryo EMbestimmt. Die Struktur des 60S Arx1 Rei1 Komplexes konnte bis zu einer Auflösung von beinahe8åverfeinertwerden,wasdieeindeutigebestimmungderorientierungdes gebundenen Arx1 auf der ribosomalen Untereinheit sowie eine detaillierte Interpretation der molekularen Kontakte zuliess. Die Struktur zeigt, dass Arx1 zwischen der Region des Polypeptid Tunnelausgangs und einem für Eukaryoten spezifischenrrna Expansionssegmenteingeklemmtist.MöglicherweiseschütztArx1 diesenfunktionellwichtigenteildesribosomswährendderribosomen Biogenese.
