6 Intended Use Intended Use Note: Only use the device for applications that fall within the range of the intended use. Otherwise, the protective and safety equipment of the device could fail. Description The CM 2.1S conductivity monitor measures the electrical resistance of a buffer solution if it contains salts and, by doing so, indicates the solution's conductivity. This makes it possible to trace programed buffer gradients during HPLC analysis. The monitor is composed of an analysis unit and a flow cell and can be operated with a ph sensor. The flow cell and ph sensor must be ordered separately. If the device will be used without a ph sensor, protect the connection on the front side of the device by inserting the blind plug supplied in the scope of delivery. 8 Intended Use Features The conductivity monitor operates contact-free and is maintenance-free, as liquids are guided through the flow cell alone and not into the analysis unit. The contact-free sensor is made of the materials titanium and PEEK. Flow cells Flow cells are available for the flow rate ranges from 0 10 ml/min and from ml/min. The flow cell is not included in the scope of delivery and must be ordered separately. It is integrated into the flow path via 1/16" capillary, that are fixed to the flow cell. ph sensor A flow cell for the ph sensor can be connected to the monitor by means of a mounting bracket. The ph sensor and the mounting bracket are not included in the scope of delivery and can be ordered separately. 8 KNAUER CM 2.1S User Manual V6710, Version 1.0

9 Intended Use Eluents Even small quantities of other substances, such as additives, modifiers, or salts can influence the durability of the materials. If there is any doubt, contact the Technical Support of the manufacturer. 12 Safety for Users Safety for Users Professional group The user manual is addressed to persons that are qualified as chemical laboratory technicians or have completed comparable vocational training. The following knowledge is required: Fundamental knowledge of liquid chromatography Knowledge regarding substances that are suitable only to a limited extent for use in liquid chromatography Knowledge regarding the health risks of chemicals Participation during an installation of a device or a training by the company KNAUER or an authorized company. If you do not belong to this or a comparable professional group, you may not perform the work described in this user manual under any circumstances. In this case, please contact your superior. 13 Safety for Users Use of inline filters Once they have been used, never re-use capillaries in other areas of the HPLC system. Only use a given PEEK fitting for one specific port and never re-use it for other ports. Always install new PEEK fittings on each separate port. Follow KNAUER or manufacturer's instructions on caring for the columns More safety-relevant information is listed in alphabetical order in the following table: Topic Eluent lines Flammability Leaks Power cable Power strip Power supply Self-ignition point Solvent tray Explanations Install capillaries and hoses in such a way that liquids cannot get into the interior in case of a leak. Organic eluents are highly flammable. Since capillaries can detach from their screw fittings and allow eluent to escape, it is prohibited to have any open flames near the analytical system. Regularly check if any system components are leaking. Defective power cables are not to be used to connect the device and the mains power. If several devices are connected to one power strip, always consider the maximum power consumption of each device. Only connect devices to voltage sources, whose voltage equals the device's voltage. Only use eluents that have a self-ignition point higher than 150 C under normal ambient conditions. Risk of electrical shock or short circuit if liquids get into the device's interior. 14 Safety for Users Topic Toxicity Explanations Organic solvents are toxic above a certain concentration. Ensure that work areas are always well-ventilated! Wear protective gloves and safety glasses when working on the device! Where is use of the device prohibited? Never use the system in potentially explosive atmospheres without appropriate protective equipment. For further information, contact the Technical Support of KNAUER. Take the device out of operation At any time, take the device completely out of operation by either switching off the power switch or by pulling the power plug. Decontamination Contamination of devices with toxic, infectious or radioactive substances poses a hazard for all persons during operation, repair, sale, and disposal of a device. DANGER! Danger if getting in contact with toxic, infectious or radio-active substances. Before disposing off or sending away contaminated devices, commission an expert with the decontamination. All contaminated devices must be properly decontaminated by a specialist company or the operating company before they can be recommissioned, repaired, sold, or disposed of. 17 Installation Installation Contacting the Technical Support Various options are available for contacting the technical support: Phone Fax Requests may be submitted in English or German. Setup When setting up the device, please take care to select a suitable setup location that meets the specifications. The specifications and step-by-step instructions can be found in the following section. Location requirements Caution! Defect of the device due to overheating possible! Protect the device against exposure to direct sunlight. Make sure the room is well-ventilated. Keep clear at least 15 cm at rear and 5 10 cm at each side for air circulation. For the device to be used as intended, all of the requirements for the location must be met (refer to the specifications for ambient conditions in the chapter Technical Data): Sunlight: Protect the device from exposure to direct sunlight. A/C system: Set up the device at a location not exposed to air drafts. Vibration: Do not place the device in the vicinity of machines that cause floor vibrations. 18 Installation Unpacking Prerequisite No damage to the packaging was detected. Tools Utility knife Caution! Damage to projecting parts possible while carrying, setting up and installing a device. Grip the device around the middle of its side panels. In case necessary, ask a second person for help. Process 1. Set up the delivery in such a way that writing on the label is rightside-up. Using the utility knife, cut the adhesive tape and open the packaging. 2. Remove the foam insert. Remove the accessories and the user manual. 3. Open the accessories kit and take out all accessories. Check the scope of delivery. In case any parts are missing, contact the technical support. 4. Clasp the device at its side panels and lift it out of the packaging. 5. Remove the foam inserts from the device. 6. Check the device for signs of damage that occurred during transport. In case you notice any damage, contact the technical support. 7. Set up the device in its location. 8. Remove the protective foil. Transport lock The monitor is delivered without a transport lock. 19 Installation Integration in an HPLC system Integrate the conductivity monitor in the HPLC system. If the conductivity monitor will be used in an HPLC system together with a UV detector and a ph sensor, the monitor should be connected downstream from the UV detector and upstream from the ph cell in order to accommodate the varying pressure stability of the cells. 20 Installation Space requirements Side clearance to other devices: At least 5 cm, if there is another device on one side. At least 10 cm, if there are devices set up on both sides. Flow cell The flow cell is delivered with permanently connected capillaries. These capillaries cannot be removed from the flow cell. If the capillaries become clogged and repeated flushing with water does not succeed in cleaning them, the flow cell must be replaced. A plug for attaching the flow cell to the monitor is located on the rear side of the flow cell. 21 Installation Attaching the flow cell Prerequisite The device has been switched off. Tools Allen screwdriver Process 1. Plug the flow cell onto the socket 1 on the front side. 2. Connecting the capillaries Fig. 7 Screws for flow cell Prerequisite The device has been switched off. Process The capillaries are part of the flow cell and cannot be removed. To connect these capillaries to other capillaries for integration into an HPLC flow path, use the supplied couplings. INLET: At this point, attach the capillary that runs from the column or an upstream detector. OUTLET: At this point, connect the capillary that runs to the waste, another detector, or a fraction collector. KNAUER CM 2.1S User Manual V6710, Version

22 Installation Next steps Install the ph sensor at the connection on the front side of the monitor, or plug this connection using the blind plug supplied in the scope of delivery. ph sensor The ph sensor is used for measuring of the ph value of saline solutions. For this purpose a ph electrode and a ph flow cell are required. When flushing the monitor or using non-saline solutions, the ph electrode has to be removed from the ph flow cell and replaced with a dummy electrode. 23 Installation Installing the ph flow cell and ph electrode Prerequisite The mounting bracket is mounted and the ph flow cell is fixed to the mounting bracket. Process 1. Screw the locking ring 2off the ph flow cell and pull it over the cable of the ph electrode Remove the protective cap from the ph electrode. 3. Plug the ph electrode into the ph flow cell 3and tighten the locking ring 2on the flow cell. 4. Insert the plug of the ph electrode into socket on the front side of the monitor. Figure Fig. 9 Installation of ph sensor Next steps Integrate the ph flow cell into the HPLC flow path by installing it downstream of the conductivity monitor. Screwing on the capillaries Prerequisite The ph sensor has been mounted to the monitor. Process Connect the capillaries in the flow direction. Use the bushings and ferrules with flat bottom supplied in the scope of delivery of the mounting bracket. Do not use any tools to tighten the bushings. OUTLET: At this connector, connect the capillary that runs to the waste, another detector, or a fraction collector. 24 Installation Connections All of the control connections are located on the rear side of the device Legend 1 LAN connection 2 Analog ports 3 RS-232 port 4 Mains connection 5 Ground Fig. 10 Monitor: Connections on rear side Connecting the Devices to a Computer via LAN This chapter describes how to set up a local area network (LAN) and how a network administrator can integrate this LAN into a company network. The description applies to the operating system Windows and all conventional routers. Note: To set up a LAN, we recommend to use a router. The following steps are necessary: Process 1. On the computer, go to the Control Panel and check the LAN properties. 2. Connect the router to the devices and the computer. 3. On the computer, configure the router to set up the network. 4. Install the chromatography software from the data storage device. 5. Switch on the device and run the chromatography software. 24 KNAUER CM 2.1S User Manual V6710, Version 1.0

25 Installation Note: The port ist set to at the factory. The IP port numbers in the device configuration of the chromatography software must be identical to those in the device, otherwise the connection fails. Configuring the LAN Settings The LAN uses only one server (which is normally the router) from that the devices automatically receive their IP address. Prerequisite In Windows power saving, hibernation, standby, automatic Windows update and screen saver must be deactived. In case you use a USB-to-COM box, the option "Allow the computer to turn off this device to save power" in the Device Manager must be deactivated for all USB hosts. Only for Windows 7: For the network adapter, the option "Allow the computer to turn off this device to save power" in the Device Manager must be deactivated. Procedure 1. In Windows 7 chlick on Start Control Panel Network and Sharing Center. 2. Double-click on LAN Connection. 3. Click on the button Properties. 4. Select Internet Protocol version 4 (TCP/IPv4). 5. Click on the button Properties. 6. Check the settings in the tab General. The correct settings for the DHCP client are: a) Obtain an IP address automatically b) Obtain DNS server address automatically 7. Click on the button OK. Connecting the Cables To avoid interference, we recommend to operate the HPLC system separate from the company network. This is why we recommend using a router. A router has several LAN ports and one WAN port that can be used to integrate the LAN into a wide area network (WAN), e. g. a company net- KNAUER CM 2.1S User Manual V6710, Version

26 Installation work or the Internet. On the other hand, the LAN ports serve to set up a network from devices and a computer. Note: A patch cable is included in the accessories kit. A router is not included in the scope of delivery. To connect the router to a WAN, an additional patch cable is required, which is also not supplied within the scope of delivery. Use the patch cable to connect the router and the computer. Repeat this step to connect all devices. 2. 27 Installation Configuring the Router The router is preset at the factory. You can find a label at the bottom side of the router, on which IP address, user name, and passwort are printed. These information help to open the router configuration. Procedure 1. To open the router configuration, start your Internet browser and enter the IP address (not for all routers). 2. Enter user name and password. 3. Configure the router as DHCP server. 4. In the router configuration, check the IP-address range and make changes if necessary. Result Once the router has assigned IP addresses to all devices, the chromatography software can be used to remotely control the system. Integrating the LAN into a Company Network A network administrator can integrate the LAN into a company network. In this case the WAN port of the router is used. Prerequisite There is a patch cable for the connection. Procedure 1. Check that the IP-address range of the router and of the company network do not overlap. 2. In case of an overlapping, change the IP-address range of the router. 3. Use the patch cable to connect the router WAN port to the company network. 4. Restart all device, including the computer. Controlling Several Systems Separately in a LAN Devices connected to a LAN communicate through ports, which are part of the IP address. If more than one HPLC system is connected to the same LAN and you plan on controlling them separately, you can use different ports to avoid interference. Therefore, the port number for each device must be changed and this same number must be entered into the device configuration of the chromatography software. We recommend to use the same port number for all devices in the same system. Note: KNAUER CM 2.1S User Manual V6710, Version

28 Installation The port ist set to at the factory. The IP port numbers in the device configuration of the chromatography software must be identical to those in the device, otherwise the connection fails. Procedure 1. Change the port number of the device. 2. Enter the port number in the chromatography software. Result The connection is established. Note: Refer to the troubleshooting chapter, if you experience connection problems. Note If the LAN cable has been connected and there is no fault with the connection, there is a green light at the LED on the LAN port on the rear side of the device. During data transfer, there is a yellow flashing light at the second LED. Software The conductivity monitor is embedded in the software packages OpenLAB and ClarityChrom (from version 5). Analog output If the analog output is used, the device sends out the measuring values as varying voltage. In analyzing the data, note the following conversion factors: ph value ph 0 = 50 mv ph 7 = 500 mv ph 14 = 950 mv Conductivity: The measured conductivity (ms/cm) is converted directly to voltage (mv), thus 1 ms/cm equates to 1 mv. 28 KNAUER CM 2.1S User Manual V6710, Version 1.0

29 Installation Power connection The devices are equipped with a universal switched-mode power supply rated for V AC. The switched-mode power supply automatically selects the correct supply voltage. Electric shock hazard caused by an improperly grounded power connection. Ground the power connection according to the pertinent regulations! Use a three-conductor line cord! Switch off the module and pull the power plug to completely isolate it from the supply voltage. Process 1. Set the ON/OFF switch 1 to the OFF position. 2. After connecting the power supply to the device, plug in the power supply. 30 Operation Operation The device is exclusively controlled by means of the membrane keyboard. It is not possible to control it using the software, but the measurement can be monitored via software Initial startup Before the initial startup, check the following items: A connection to the computer using the desired interface has been established. The software has been installed. The flow cell has been attached properly. Switch-on Prerequisite The device has been installed at a suitable site. The device is connected to the power supply. The initial startup has been completed. Process 1. Set the ON/OFF switch 1 on the power supply to the ON position. Figure 1 Fig. 14 Power supply: Switch in the ON position Result The display lights up and a green LED on the power supply indicates that the device is supplied with power. 31 Operation Basic operation Figure Fig. 15 Status display Fig. 16 Arrow keys Explanation The status display shows the conductivity (line 1), the present gradient composition (line 2, left), and the ph value (line 2, right). Press both arrow keys simultaneously to activate the keypad. Hold down one arrow key to scroll through the menus with the other key. Use either of the buttons to adjust the values or to switch from one setting to the next. Menus Status display Description The measuring signal can be offset within a set range between % and %. It is possible to set the correction factor and reference temperature for the temperature sensor located in the flow cell: The reference value for the correction factor is 1.7 %. The reference value for the reference is 25 C. With the menus Factor 1-4 it is possible to define up to 4 calibration points. For each point, enter the conductivity or the correction factor in order to generate a correct calibration curve. KNAUER CM 2.1S User Manual V6710, Version

32 Operation Status display Description Two menus are used for calibrating the ph sensor. #0 is preset to ph07. #1 can be adjusted to a value between ph02...ph12 and corresponds to the ph value of the second calibration solution that is used for calibration. Select this menu to view the serial number. Select this menu to view the serial number of the flow cell. This function is not available for all flow cells. Select this menu to view the operating hours of the conductivity monitor. This menu is used to select the interface for the PC connection: LAN DHCP 9600 Baud (RS 232) With this menu it is possible to display of the current progress of the programmed buffer gradients in the status display. To do so, it is first necessary to enter the conductivity of the initial concentration of the programmed gradient. In this menu, enter the conductivity of the final concentration of the programmed buffer gradient. 32 KNAUER CM 2.1S User Manual V6710, Version 1.0

33 Operation Select an interface for control of the device To view the measuring results of the conductivity monitor via software, the interface used must be selected on the device. PC: LAN DHCP PC: 9600 Baud (RS-232) Prerequisite A physical connection has been established. The monitor has been switched on. Process 1. Press both arrow keys. 2. Hold down the left arrow key and use the right arrow key to scroll until the correct display appears. 3. Release the arrow keys and then press the left or right arrow key to select an option. 35 Operation Setting the temperature compensation Ions that have dissolved in water have varying conductivity depending on the temperature. The conductivity of NaCl solutions increases on average by around 2 % per C. The default temperature setting in the device is 25 C. In order to account for the deviation in conductivity at a different ambient temperature, this must be corrected in the TEMP-COR menu. For this, use a NaCl standard. The reference value for the correction factor is 1.7%. Prerequisite The monitor has been switched on. Process 1. Press both arrow keys. 2. 42 Functionality Tests Functionality Tests Installation Qualification (IQ) Installation report Certification on the functionality of the device. During installation of the device, an installation report (IQ document) is created upon request in coordination with the Technical Support of KNAUER. This installation report needs to be completed in full and signed by both parties. It serves as proof of the properly executed installation and functionality of the device. Operation Qualification (OQ) Extensive functionality test Extensive test of the device s functionality. A successfully executed OQ ensures that the device functions properly. Test Intervals Run the extensive functionality test at the following time intervals: Average use Device test 1 to 5 days/week: Every 6 months More than 5 days/week or 24 hours/day: Every 3 months Operation with buffer solutions or other salt solutions: Every 3 months Execution The test can carried out either by KNAUER's Technical Support or by a technical service authorized by KNAUER. 43 Troubleshooting Troubleshooting If the device stops functioning or there are connection problems, start by checking the following items: Is the status display lit up? Are the LEDs at the LAN connection lit up? When the cable is connected, a green LED lights up. There is a flashing yellow light at the second LED if the data exchange is in progress. Is the LED at the power supply lit? LAN In case no connection between the computer and the devices can be established, go through the following points. Check after each point, if the problem is solved. If you did not manage to locate the problem, call the Technical Support. 1. Check the status of the LAN connection in the Windows taskbar: Connected Connection not established If no connection was established, test the following: Is the router on? Is the patch cable connected correctly to the router and the computer? 2. Check the router settings: Is the router set to DCHP server? 44 Troubleshooting 3. Check all connections. Are the patch cable connected to the LAN ports and not the WAN port? Are all cable connections between devices and router correct? Are the cables plugged in tightly? 4. If the router is integrated into a company network, pull out the patch cable from the WAN port. Can the devices communicate with the computer, even though the router is disconnected from the company network? 5. In case you own a Control Unit, check the settings in the menu Setup > Network. Is LAN-DHCP set for controlling? Did the device receive an IP address? 6. Turn off all devices, router, and computer. Firstly turn on the router, secondly the devices and the computer. Has this been successful? 7. Replace the patch cable to the device with that no connection could be established. Has this been successful? 8. 46 Maintenance and Care Maintenance and Care Proper maintenance of your HPLC device will ensure successful analyses and reproducible results. Maintenance contract The following maintenance work on the device may only be performed by the manufacturer or a company authorized by the manufacturer and is covered by a separate maintenance contract: Opening the device Removing the hood or the side panels Caring for the device Prerequisite The monitor has been switched off. The power plug has been pulled. Caution! Intruding liquids can cause damage to the device! Place solvent bottles next to the device or in a solvent tray. Moisten the cleaning cloth only slightly. 47 Maintenance and Care Flushing the flow cell Increased system pressure and reduced sensitivity may be the result of a clogged flow cell. Thus, generally flush the conductivity monitor with water. Often deposits can be removed by repeated flushing of the flow cell. Flushing solution The following eluents are recommended for flushing: Water Isopropanol Prerequisite The dummy electrode has been inserted into the ph flow cell. Tools Syringe Caution! Contamination of the flow cell caused by oil drops! Do not use compressed air for drying. Process 1. Fill the eluent into a syringe and inject it into the capillary at the inlet to the flow cell. Next, allow it to take effect for 5 minutes. 2. Fill the syringe with water and inject again. 3. Remove the flow cell from the monitor and use nitrogen stream to dry it. 54 Disposal Disposal Hand in old devices or disassembled old components at a certified waste facility, where they will be disposed of properly. AVV marking in Germany According to the German "Abfallverzeichnisverordnung" (AVV) (January, 2001), old devices manufactured by KNAUER are marked as waste electrical and electronic equipment: WEEE registration KNAUER as a company is registered by the WEEE number DE in the German "Elektroaltgeräteregister" (EAR). The number belongs to category 8, which, among others, comprises laboratory equipment. All distributors and importers are responsible for the disposal of old devices, as defined by the WEEE directive. End-users can send their old devices manufactured by KNAUER back to the distributor, the importer, or the company free of charge, but would be charged for the disposal. Eluents and other operating materials All eluents and other operating materials must be collected separately and disposed of properly.
