General aim:3 STEP Purification Under normal conditions you lose at each purification step ca 20%, 1.Step capture 2. Step purify 3. Step polish Each step should be a different type of separation, has different capacities and different performance. E.g. capacity decreases from step1 to 3, performance increases step 1-3.
General rules Until you don t know better -Use protease inhibitors -Work fast (prepare buffers, SDS PAGE, Blots etc) -Keep cold protein (slower denaturation and less proteolysis) -Do NOT freeze in between; if necessary shock freeze with glycerol. -Start with high amounts of protein -Make small scale tests prior large scale preparations
