3 Abstract in this thesis solid-state NMR investigations on a fragment of the HET-s prion protein are presented, Prion proteins are associated with a numberof infeetive neurodegenerative diseases such as BSE in cattle, Serapie in sheep and goat, and CID in humans, Prion proteins can exist in two different conformations: a non-infective and an infective one called thc prion state. The prion form aggregates to amyloid, which consists out of protein fibrils, Amyloid plaques are not only a symptom in prion diseases, but are also found in other neurodegenerative diseases such as Alzheimer's and Parkinson's diseases, The HET-s protein of the filamentous fungus Podospora anserina is a prion protein. Interestingly its infective form is probably not involved in a disease, but has a beneficial function for its hast. Thcrefore, investigations on this protein are not only important to better understand the formation and propagation of amyloid fibrils, but will also help to deepen our understanding of prion biology. Since amyloids are non soluble and non crystallinethe standard methods ofprotein structure deterrnination (i.e. liquid-state NMR and X-ray crystallography) cannot be applied to these systems, For solid-state NMR investigations, however, neither soluble proteins nor protein crystals are needed, Hence solid-state NMR is vcry weil suited to determine the structure of amyloid fibrils. In this work ac-terminal fragrnent of HET-s, namely from residuc 218 to 289, was investigated, This fragment will be callcd HET-s( ) in the following. HET-s(218~289) was shown to be the protease resistant core of the HET-s amyloid fibril. Furthermore, HET-s( ) alone can form amyloid fibrils and can be used to trigger prion infection. The first step of the structure determination with solid-state NMR was the resonance assignment of uniformly ISN and 13C labeled HET-s( ) amyloid fibrils. For this assignment a series 01' 13C _ l3c and lsn _ 13 C 2D and 3D spectra was recorded. With these spectra it was possible to obtain the side-chain as weil as the backbone assignment of 50 of the 72 aminoacid residues of HET-s( ), These amino acids are highly ordered and relatively rigid. Furthennore, the chemical-shift analysis showed that these residues are mostly in a ß-sheet 11

4 12 Abstract confonnation. Using a combination of liquid- and solid-state methods it was possible to identify additional amino acids. The sequential assignment of these residues, however, was not possible. These residues are located in dynamic parts of the fibril and the chemical-shift analysis revealed that they are in a random-coil conformation. Therefore, the HET-s( ) fibrils are dynarnieally and structurally very heterogeneous. Uniformly as weil as alternatingly Be labeled samples of HET-s( ) were used to record spectra containing information about internuelcar distances within the fibril. The proton-driven spin diffusion spectra used for that purpose were characterized by severe signal overlap and unambiguousness in the signal assignment. Therefore, a computer program was used for the automatie interpretation of these speetra. Because it was not possible to distinguish between inter- and intra-monomer signals in these speetra, ea1culations were done making assumptions about the intermolecular arrangement of the monomers within the fibril. The resulting model of the HET-s( ) monomer forms a parallel ß-sandwich structure build out of four ß-strands. This model is in very good agreement with ail data coming from other methods.

5 Zusammenfassung Die folgende Arbeit befasst sich mit Festkörper-NMR Untersuchungen an einem Fragment des Prion Proteins HET-s. Prion Proteine werden für eine Reihe von infektiösen, neurodegenerativen Krankheiten verantwortlich gemacht, unter ihnen BSE in Rindern, Serapie in Schafen und Ziegen und CJD in Menschen. Prion Proteine können zwei Formen annehmen: Eine infektiöse und eine nichtinfektiöse Form. Die infektiöse Form dieser Proteine aggregiert zu so genanntem Amyloid, welches überwiegend aus Protein-Fibrillen zu bestehen scheint. Amyloid Ablagerungen findet man nicht nur in Verbindung mit Prionenerkrankungen, sondern auch in Zusammenhang mit anderen neurodegenerativen Krankheiten wie Alzheimer oder Parkinson. Das HET-s Protein aus dem filamentösen Pilz Podospora anserina ist ein Prion Protein. Interessanterweise scheint die infektiöse Form dieses Proteins keine Krankheit hervorzurufen, sondern hat eine positive Funktion für den Pilz. Untersuchungen an diesem Protein können deshalb nicht nur helfen die Bildung und Ausbreitung von Amyloid Fibrillen zu verstehen, sondern werden auch zu einem tieferen Verständnis der Prion Biologie beitragen. Aufgrund ihrer nichtlöslichen und nichtkristallinen Form lässt sich die dreidimensionale Struktur von Amyloiden nicht mit den klassischen Methoden der Proteinstrukturaufklarung, namentlich der Flüssigkeits-NMR und Röntgenkristallographie, aufklären. Für die Untersuchung eines Proteins mit Festkörper NMR Spektroskopie ist es hingegen weder nötig, dass das Protein löslich, noch dass es als Einkristall vorliegt. Deshalb ist Festkörper-NMR Spektroskopie sehr gut geeignet um die Struktur von Amyloid Fibrillen aufzuklären. In der hier vorliegenden Arbeit wurde ein C-terminales Fragment von Aminosäure 218 bis 289 von HET-s (HET-s( ) ) untersucht. Bei diesem Fragment handelt es sich um den proteaseresistenten Teil der von HET-s gebildeten Amyloid Fibrille, und es wurde gezeigt, dass dieses Fragment allein schon ausreicht um Fibrillen zu bilden und eine Prion Infektion hervorzurufen. Der erste Schritt in der Strukturbestimmung von HET-s( ) in seiner amyloiden Form bestand aus der 15N und 13C Festkörper-NMR Resonanzzuordnung. Diese wurde an einem 13

6 14 Zusammenfassung vollständig 15N und 13e markierten Probe von HET-s( ) durchgeführt, wobei eine Reihe von 13e _l3e und 15N _ 13e 2D und 3D Korrelationsspektren aufgenommen wurde um sowohl die Resonanzen innerhalb einer Aminosäure zuzuordnen als auch die Position dieser Aminosäuren innerhalb der Proteinsequenz zu bestimmen. 50 der 72 Aminosäuren von HET-s( ) konnten mit Hilfe dieser Experiment zugeordnet werden. Diese Aminosäuren zeichnen sich durch relative hohe Rigidität und durch eine beinahe kristalline Ordnung aus. Zudem konnte man aus den gemessenen Resonanzen schließen, dass sich diese Aminosäuren überwiegend in der ausgestreckten Konformation von ß-Faltblättern befinden. Mit einer Kombination aus Festkörper- und Flüssigkeits-NMR Methoden konnten zusätzliche Aminosäure von HET-s( ) identifiziert werden, deren sequentielle Zuordnung allerdings nicht möglich war. Diese Aminosäuren befinden sich in dynamischen Teilen von HET-s( ) und besitzen ihren chemischen Verschiebungen zur Folge auch keine geordnete Sekundärstruktur. HET-s( ) ist demnach dynamisch und strukturell sehr heterogen. An sowohl vollständig als auch alternierend 13e markierten Proben von HET-s( ) wurden im Anschluss daran Spektren aufgenommen, die Information über Atomabstände innerhalb der Fibrille enthielten. Die Spektren zeichneten sich durch starke Signalüberlappung und Nichteindeutigkeit der Signale aus. Deshalb wurde ein Computerprogramm zur automatische Auswertung dieser Spektren eingesetzt. Da es nicht möglich war inter- von intramolekularen Signalen zu unterscheiden, wurden unter Annahme unterschiedlicher intermolekularer Anordnung die Faltung des HET-s( ) Monomers innerhalb der Fibrille bestimmt. Das resultierende Strukturmodell von HET-s( ) besteht aus einem parallelen ß-Sandwich. Dieses Modell stimmt hervorragend mit allen publizierten Daten von HET-s( ) übcrein.
