2 Insitu Biocell Viability Analyser Anwendung Application Exposed Particle Surface Area Measurement Die Qualität des Produktes wird durch den Herstellungsprozess bestimmt. Maßgeblich ist dabei, dass die Zellen unter den optimierten Bedingungen fermentiert werden. Häufig erfolgt die Überprüfung dieser Bedingungen jedoch über Werte, die nur indirekt einen Rückschluss auf einzelne Parameter erlauben. Der Sequip BCVA Sensor hingegen richtet den Blick direkt auf die Zellen. Dabei bietet der BCVA Sensor die Möglichkeit zur in situ Messung der Parameter Zellzahl, Vitalität, Zellwachstum, Morphologie sowie der optischen Dichte, durch die fraktionelle Darstellung einzelner Zellgrößen, im Fermenter präzise in Echtzeit. Damit ist eine direkte Kontrolle und Steuerung der Zelleigenschaften möglich. The quality of a product is decided by the production process. It is necessary that cells are fermented under optimal conditions. But often the conditions are tested by values, which are give only indirect results about single parameters. The Sequip BCVA Sensor examines the cells directly. In doing so BCVA Sensor offers the opportunity of precise real time in situ measurement of the parameters cell count, vitality, cell growth, morphology and optical density, through display of fraction of single cell sizes, in the fermenter. Therefore a direct control and governing of cell properties is possible. 4 Insitu Bio Cell Viability Analyser Wirkungsweise Die Reflexion des Lichtes wird durch die Zellen (und alle anderen Partikel) im Beobachtungsraum auf unterschiedliche Art beeinflusst. Befindet sich eine Zelle im Fokus des Laserstrahls, so verändert sich das Signal. Unter Berücksichtigung der Geschwindigkeit der Fokusrotation können die Größe und Form der Zelle bestimmt werden. Lebende und tote Zellen reflektieren das Licht unterschiedlich. Somit können sie durch differenzierte Signalformen unterschieden werden. Daraus lässt sich die Vitalität berechnen. Das vom Licht durchleuchtete Volumen ist bekannt, so dass die Zellzahl berechnet werden kann. Aufgrund der Multi Capture Signal Analysis (MCSA) erfolgt die Bestimmung der verschiedenen Parameter parallel und zeitgleich. Die Darstellung des zeitlichen Verlaufs der Signale ermöglicht die online Beobachtung und Kontrolle des Prozesses. Mode of operation The reflection of light is affected by the cells (and all other particles) in the range of observance in different ways. When a cell is in the laser beam, the signal changes. Considering the rate of focus rotation the size and shape of the cell can be identified. Living and dead cells reflect the light different. Therefore the specific signal shape can be distinguished. So the vitality can be analysed. The volume scanned by the beam is known, so the cell count can be calculated. Based on the Multi Capture Signal Analysis (MCSA) the identification of the different parameters is parallel and simultaneously. The diagram of the chronological sequence of the signals allows online observation and control of the process.
