Abstract This thesis describes the development of methodologies for the asymmetric synthesis of Tröger s base-derivatives (TBs). Their unique structural features such as the presence of configurationally stable stereogenic nitrogen atoms and a V-shaped hydrophobic cavity between two aromatic rings have made them attractive candidates for numerous applications such as drug delivery, catalysis and molecular recognition. The delivery of TBs as single enantiomers remains a pertinent challenge. As the reported methods are typically suited for distinct examples, the aim was to develop strategies that are more generally applicable for the enantioselective preparation of TB derivatives. The first methodology explored in this thesis was the enzymatic resolution of TBs. Substrates bearing hydroxyl groups were subjected to kinetic resolutions via acetylation in the presence of a bacterial enzyme (Candida antarctica lipase type B), affording (S,S)-enantiomers of TB-diols (59) in an enantiomerically pure form (Scheme 1). The resolved compounds underwent post-modifications without loss of ees, providing access to enantiomerically pure precursors, which can be transformed to ligands for asymmetric catalysis and molecular recognition. ix

Another investigated strategy was an extension of a previous finding in our group. It had been discovered that racemic TB derivatives can be synthesised via a double aza-michael addition (DAMA) of tetrahydrodiazocine (44a) to electron-poor alkynes. Diastereoselective DAMA was carried out successfully with chiral auxiliaries. This project aimed to develop a catalytic version of the asymmetric DAMA to circumvent the application of stoichiometric amounts of chiral materials. The selective synthesis of one enantiomer of a TB derivative was attempted with various alkynes and chiral catalysts. In the case where 3-butyn-2-one (142) was the Michael acceptor, (DHQD) 2PHAL as the catalyst in MeCN gave ee values of over 60%. To the best of our knowledge, this is the best result for the synthesis of N-stereogenic compounds by an enantioselective catalysis. Double activation of 3- butyn-2-one (142) by adding gold(i) as a Lewis acid, followed by further addition of 3-butyn-2-one (142) after three days led to an improved product yield of 70% (Scheme 2). The unexpectedly high ee values seen in the presence of (DHQD) 2PHAL provided an incentive for the reduction in the high loading of this expensive catalyst (up to 30 mol%). Attention was thus paid to the incorporation of phase-transfer-catalysts (PTCs) which are known to assist in asymmetric aza- Michael additions. Importantly, the simple set up and mild conditions with PTCs make them appropriate for large scale operations. Since the blank reaction between tetrahydrodiazocine (44a) and 3-butyn-2-one (142) was very slow, efforts were first made to find appropriate reaction conditions for improved reactivity of the system before performing asymmetric synthesis. Pleasingly, in the presence of BnEt 3NBr/MeCN, the product yield after 12 hours was significantly enhanced from 19 to 62%. Base-free conditions were beneficial for improving the reaction rate. Further addition of 3-butyn-2-one (142) after the initial 12 hours gave TB-derivative 71h in a near-quantitative yield (Scheme 3). x

Lastly, the research presented in this thesis expanded the synthetic scope of the aza-michael addition of tetrahydrodiazocine (44a). A series of activated alkynes and alkenes were evaluated but only a few proved reactive. In the absence of an activating substituent, the Michael acceptor proved to be unreactive. These products containing modified bridges could serve as scaffolds for further molecules (Scheme 4). xi

Zusammenfassung In der vorliegenden Doktorarbeit werden Entwicklungen der Methoden für asymmetrische Synthese Trögerscher Basen (TBs) thematisiert. Wegen der aussergewöhnlichen Eigenschaften der TBs wie zum Beispiel die Präsenz konfigurativ stabiler, chirogener Stickstoff-Zentren und die hydrophobe, V- förmige Tasche gelten die TBs als attraktive Verbindungen für verschiedene Anwendungen wie zum Beispiel in der Molekülerkennung oder als Liganden für die asymmetrische Katalyse. Trotzdem bleibt die Herstellung von TBs in enantiomerenreiner Form eine grosse Herausforderung. Da die in der Literatur beschriebenen Methoden typischerweise nur für bestimmte TB-Variante funktionieren, wurde in dieser Doktorarbeit versucht, allgemeingültige und effiziente Strategien zur asymmetrischen Herstellung substanzieller Mengen von TBs zu entwickeln. Die erste untersuchte Methode war die enzymatische Racematspaltung der TBs. Kinetische Racematspaltung der Substrate wurde durchgeführt, wodurch die Hydroxylgruppen mit einem Enzym bakteriellen Ursprungs (Candida antarctica lipase type B) als Katalysator acetyliert wurden. So wurden die (S,S)-Enantiomere der DihydroxyTBs (59) enantiomerenrein isoliert (Scheme 1). Die enantiomerenreinen Verbindungen konnten in den Postmodifikationen ohne Verlust der Enantiomerenreinheit modifiziert werden. Der Zugang zu enantiomerenreinen Vorläufern für Liganden oder zur Molekülerkennung wurde eröffnet. xii

Eine andere Strategie rührt von einer früheren Entdeckung unserer Gruppe her. Es war schon bekannt, dass TBs durch doppelte aza-michael Addition (DAMA) von Tetrahydrodiazocin (44a) an elektronarmen Alkynen synthetisiert werden können. Diastereoselektive DAMA war in Gegenwart chiraler Hilfsreagenzen erfolgreich. Ein Ziel dieser Doktorarbeit war es, eine katalytische Variante der asymmetrischen DAMA zu entwickeln, um die Verwendung stoichiometrischer Mengen von Hilfsreagenzen zu vermeiden. Die selektive Synthese eines Enantiomers eines TB-Derivats wurde mit verschiedenen Alkynen in Gegenwart von unterschiedlichen Katalysatoren versucht. Mit 3-Butyn-2- on (142) als Michael Akzeptor und (DHQD) 2PHAL als Katalysator in MeCN, ee von über 60% wurde erreicht. Die Ausbeute wurde mit einem Gold(I) Komplex als Lewis Säure und portionenweiser Zugabe von 142 bis zu 70% erhöht (Scheme 2). Von den überraschend hohen Werten von ee in Gegenwart von (DHQD) 2PHAL wurden wir inspiriert, zu versuchen, das Loading des teuren Katalysators (bis zu 30 mol%) zu reduzieren. Phasentransferkatalysatoren (PTCs) schienen uns äusserst interessant, da Beispiele von erfolgreichen asymmetrischen PTC-katalysierten organischen Reaktionen existieren, auch Aza- Michael Additionen. Solche Reaktionen sind einfach anzusetzen, werden unter milden Bedingungen durchgeführt und sind deshalb für Synthesen im grösseren Massstab geeignet. Da die DAMA von 44a und 142 ohne Katalysator sehr langsam war, wurde zuerst versucht, die Reaktivität zu erhöhen. Die Ausbeute des Produkts wurde merklich erhöht in Gegenwart von BnEt 3NBr/MeCN, von 19 zu 62%. Bei portionenweiser Zugabe von 142 wurde das Produkt 71h in fastquantitativer Ausbeute isoliert (Scheme 3). xiii

Zuletzt wurde der Auswendungsbereich der aza-michael Addition von Tetrahydrodiazocine (44a) ausgeweitet. Verschiedene aktivierte Alkine und Alkene wurden als Michael Akzeptoren verwendet, jedoch waren sehr wenige Verbindungen davon reaktiv. Ohne aktivierende Substituenten war der Michael Akzeptor unreaktiv. Die Reaktionsprodukte könnten weiter für die Synthese verschiedener Grundgerüste modifiziert werden (Scheme 4). xiv
