3 -222- IV. ZUSAMMENFASSUNG Diese Arbeit befasst sich mit dar EPC-Synthese von a-verzweigten, a-heterosubstituierten Carbonylverbindungen. Dabei wurden, ausgehend von den drei wichtigsten Klassen des "Pool of chiral building blocks" (Zucker, Aminosäuren und Hydroxycarbonsauren), über geeignete Derivate chirale Enolate er zeugt und diese mit verschiedenen Elektrophilen, in hoch diastereoselektiven Reaktionen, zur C-C Verknüpfung einge setzt. Diese asymmetrischen Synthesen der o-alkylierten a-heterosubstituierten Carbonsauren wurden ohne zusatzliche chirale Hilfsstoffe durchgeführt. So konnte das Enolat des Acetonides des Weinsäuredimethylesters in ca. 80% Diastereoselektivitat mit Benzyl- resp. AI 1 vlhalogemden alkyliert werden. CO,Me 2" O^-cOjWe "COjMe Durch Acetalisierung von Pivalaldehyd mit a-hydroxy, a-mercaptocarbonsauren oder Prolin wurden Dioxolanone (I), Thioxolanone (II) oder Oxazolmone (III) hergestellt. Trennung der beiden so erhaltenen C-2 Epimeren ergab die Derivate der "chiral pool" Substanzen, die zur Erzeugung der Enolate eingesetzt wurden. Beim Umsetzen mit Elektro philen wurde die neu geschaffene C-C Bindung mit über 95%- lger Diastereoselektivitat trans zur t-butylgruppe gebildet.

4 -223- o-< o-^o >V yo -kx -kx Hh s ü R1 O -*i> *c& iv R1 R1 \y> im Eine Anwendung dieses Prinzipes der "Selbstreproduktion der Chiralität" stellt eine vierstufige EPC-Synthese vom eromon Frontalin in beiden enantiomeren Formen dar. (R)-FRONTALIN Bei der Verzweigung von Aminosäuren konnte das gleiche Prin zip durch den Einsatz von Imidazolidinonen (IV) verwirklicht werden. Es wurde ein Weg gefunden, aus einer optisch aktiven Aminosäure beide Enantiomere der chiralen Imidazolidinon enolate zu erzeugen, was den Zugang zu beiden antipodalen alkylierten Aminosäuren ermöglicht. =*V IV R 1

5 -224- S U M M A R Y The following thesis is concerned with the BPC-synthesis of a-branched, a-hetercsubstituted carboxylic Compounds. Starting with the three most important classes of the "pool of chiral building blocks" (sugars, amino acids and hydroxyacids), via suitable derivatives, enolates were generated and treated with different electrophiles to get C-C conections in very high diastereoselectivities. These asymmetric syntheses of the a-alkylated, a-heterosubstituted carboxylic acids wero done without any additional chiral auxiliary Compounds. In this manner the enolate of the acetonid of tartraric ester could be alkylated with benzyl- or allylhalides in about 80% diastereoselectivity. xl -* xj:* C02Me U C02Me Through acetalisations of pivalaldehyde with a-hydroxy-, a-mercaptocarboxylic acids or proline, dioxolanone (1), thioxolanone (I ) or oxazolinon (III) were synthesized. Separation of two thus-formed C-2 epimers gave the deriva tives of the "chiral pool" substances, which were used to generate the chiral enolates. After treating with electro philes the newly formed C-C bond is trans to the t-butylgrup with more than 95% diastereoselectivity.
