Product Information Material Safety Data Sheet Technical Support R-Biopharm AG An der neuen Bergstrasse 17, Darmstadt, Germany Phone: +49 (0) Fax: +49 (0) orders@r-biopharm.de Dieses Kit dient der Farbstoffkalibrierung am LightCycler 1.5 und 2.0, um bei duplex (FAM und VIC beim Lightcycler 1.5) und triplex (FAM, VIC und Cy5 beim LightCycler 2.0) real-time PCR-Läufen den Farbstoff- Crosstalk zwischen den Kanälen zu kompensieren. Mit Hilfe von vier Lösungen kann ein Color Compensation File erstellt werden. Damit ist es möglich, Ergebnisse von multiplex real-time PCR-Läufen mit den Farbstoffen FAM, VIC und Cy5 bzw. nur FAM und VIC zu analysieren. Kit-Inhalt und Lagerung Der Kit-Inhalt ist für drei Kalibrierungsläufe ausgelegt. 1x Blank (60 µl) (Code 1) 1x Dye 1 (60 µl) (Code 2) 1x Dye 2 (60 µl) (Code 3) 1x Dye 3 (60 µl) (Code 4) Die Reagenzien sind lichtgeschützt bei 20 C zu lagern. Zusätzliche benötigte Geräte und Materialien Real-time PCR Gerät: LightCycler 1.5 bzw. 2.0 Real-time PCR Verbrauchsmaterialien (LightCycler Kapillaren) Pipetten, Pipettenspitzen mit Filtern Manual Protokoll 1. Ansatz des Farbstoffkalibrierungslaufes am LightCycler 1.5 Für einen Kalibrierungslauf muss je Farbstoff, inklusive dem Farbstoffhintergrund (Blank), eine Reaktion mit je 20 µl in eine LightCycler Kapillare pipettiert werden. Jedoch muss in Kapillare 4 zusätzlich zu 20 µl Dye 3 noch 1 µl Dye 1 pipettiert werden. Vor der Benutzung bitte die Lösungen auftauen, vortexen und zentrifugieren. Lösung Farbstoff Menge pro Reaktion je 20 µl pipettieren in folgende Positionen Blank Hintergrund 20,0 µl 1 Dye 1 FAM 20,0 µl 2 Dye 2 VIC 20,0 µl 3 Dye 3 Cy5 20,0 µl + 1 µl Dye 1 4 Die LightCycler Kapillaren nach dem Verteilen der Lösungen verschließen und zentrifugieren. 2. 2 2. Geräteeinstellungen und starten des Experimentes 2.1 LightCycler 1.5 Click the Samples icon on the left-hand side of the window. Then click the Analysis Type menu, and select Color Compensation. Nach Anmeldung an der Software ist es erforderlich, unter dem Button "Run" das LightCycler-Experiment- Fenster zu öffnen: Define in the Dominant Channel dialog fields the dominant channel for the selected dyes for each position (see Tab.7). Select for the reagent Blank the dominant channel Water. It is not necessary to specify a "Sample Name". Regent Channels Filter Combinations Blank Blank Water Dye1 FAM 530-none Dye2 VIC 560-none Dye3 Cy5 705-none Folgendes Fenster öffnet sich: Place the LightCycler-Carousel with the capillaries in the LightCycler 2.0. Click the Run icon and save the experiment in the proper folder. Evaluation and creation of a Color Compensation File After the experiment ends, click the Analysis button to open the Create New Analysis dialog box. Under Other Methods, select Color Compensation. Then click OK In the Color Compensation window which appears, click the Save CC Object button to save the Color Compensation File in the CCC folder. The created Color Compensation File can now be applied for LightCycler experiments. To apply Color Compensation for a multiplex PCR experiment open the experiment and click the Analysis button. Click the Color Compensation (Off) button, and select the appropriate Color Compensation File. The Color Compensation (Off) button switches to Color Compensation (On) to confirm that color compensation is active. The multiplex real-time PCR experiment can now be analyzed. 4 Nach Abschluss der Programmierung ergibt sich folgendes Bild des Experimentes: 2.1 LightCycler 2.0 Open the LightCycler 2.0 software. Click the File menu, and select New to open the Create New Object dialog box. Double-click the LightCyclerExperiment icon to open a new LightCycler experiment file. The following window opens. Die Kapillaren in das LightCyler -Rondell überführen und in das Gerät setzen. Links unter dem Button "Run" den Lauf starten und in dem entsprechenden Ordner abspeichern. Es öffnet sich nun das Fenster zum Programmieren des Layouts. Hier wird nun die Anzahl an Kapillaren bei Maximum Position eingetragen und mit Enter bestätigt. 5 Save the Color Compensation File. The created Color Compensation File can now be applied for LightCycler experiments. Auswertung und Erstellung eines CC-Files Nach Abschluss des Experimentes bietet sich folgendes Bild: To apply Color Compensation for a multiplex PCR experiment click the Select CC Data button to select and import the appropriate Color Compensation File. Select the appropriate channel (e.g. F1 or F2) and click the Quantification button to analyze the experiment. Note: The Color Compensation File is specific for every LightCycler instrument. Hier nun über Select A Program das Segment 3 (TM-Analyse) für die Color Compensation auswählen. Es ist nach Wahl des Segmentes 3 darauf zu achten, dass tatsächlich die grünen Linien den Beginn und das Ende der Schmelzkurve zeigen. 6 Anschließend wird der Color Compensation File abgespeichert. Danach steht dieser File für andere LC-Läufe zur Verfügung. Evaluation and creation of a Color Compensation File After the experiment ends the following window opens. Damit ist die Generierung der Farbstoff-Kalibrierungsdatei abgeschlossen. Zum Anwenden der Farbstoffkalibrierung den jeweiligen duplex Lauf öffnen und unter Select CC Data den entsprechenden CC-File importieren und auswählen. Dann den entsprechenden Kanal (z.b. F1 oder F2) auswählen und durch Drücken des Buttons Quantification den Lauf zur Analyse öffnen. Hinweis: Die Farbstoffkalibrierungsdatei ist spezifisch für das jeweilige Gerät, d.h. bei einem Wechsel des Gerätes (oder auch bei Reparatur der optischen Einheit) ist eine neue Farbstoffkalibrierung nötig. 7 The final screen should look like the screenshot below. 2.2 LightCycler 2.0 Nach Anmeldung an der Software ist es erforderlich, unter dem Button "New " ein neues LightCycler Experiment zu öffnen: Bild 1 Place the LightCycler-Carousel with the capillaries in the LightCycler 1.5. 8 Als erster Schritt sollte das Reaktionsvolumen von 20 µl und eine Seeking-Temperatur von 70 C gewählt werden, sowie die Sample-Anzahl (4 Kapillaren) eingetragen werden. 9 1.1 LightCycler 1.5 Open the LightCycler 1.5 software. Click the Run button to open a new LighCycler Experiment. Für das Programmieren des Layouts des Kalibrierungslaufes, links auf den Button Samples klicken. Hier nun unter "Analysis Type" die Auswahl Color Compensation wählen. In der sich öffnenden Analyse auf den Button Save CC Object..." klicken und diesen Lauf als ein CC Objekt unter dem Ordner CCC abspeichern. 10 Zum Anwenden der Farbstoffkalibrierung den jeweiligen triplex Lauf öffnen und unter Analysis den die Analyse öffnen. Durch Klicken auf den Button Color Compensation (Off) die abgespeicherte Farbstoff- Kalibrierungsdatei auswählen. Durch Wechseln des Buttons Color Compensation (Off) in Color Compensation (On) wird angezeigt, dass eine Farbstoffkalibrierung angewählt ist. Der duplex PCR Lauf kann nun ausgewertet werden. Hinweis: Die Farbstoffkalibrierungsdatei ist spezifisch für das jeweilige Gerät, d.h. bei einem Wechsel des Gerätes (oder auch bei Reparatur der optischen Einheit) ist eine neue Farbstoffkalibrierung nötig.
