3 - host SUMMARY SUMMARY During infection of a host, Salmonella Typhimurium encounters a multitude of different environmentalconditions. S. Typhimuriumharbours a variety of genes and gene Clusters encoded in different regions of their chromosome to survive hostile conditions and escape the host defense mechanisms.one key virulence factor is the type IM secretion system (TTSS). TTSS are used by many Gram-negative pathogens for transporting bacterial toxins (effector proteins) into eukaryotic host cells. Two modes of type IM effector protein transport can be distinguished: transport into the surrounding medium (secretion)and cell contact-induced injection of effectorproteins directly into the host cell cytosol (translocation). Two domains within the N-terminal regions of effector proteins determine the mode of transport. The amino terminal -20 amino acids (N-terminal secretion signal, NSS) mediate secretion. In contrast, translocation generally requires the NSS, the adjacent -100 amino acids (chaperone binding domain, CBD) and binding of the cognate chaperoneto this CBD. TTSS are phylogenetically related to flagellar Systems. Because both Systems are expressed in S. Typhimurium, correct effector protein transport involves at least 2 decisions: transport via the SPI-1 but not the flagellar TTSS (= specificity) and translocation into the host cell instead of secretion into the surrounding media (= transport mode). In this thesis, the question of how specificity is mediated was addressed. For this, the NSS and the CBD of the S. Typhimurium effector protein SopE were altered by alanine-replacement mutagenesis and the secretion and translocation phenotypeof the SopE mutants was investigated. SopE is specifically transported via the SPI-1 TTSS. Secretion and translocation strictly required the cognate chaperone InvB. Alanine replacement of amino acids (and to some extent 44-54) abolished tight InvB binding, abolished translocation into the host cell and led to secretion of SopE via both, the flagellar and the SPI-1 TTSS. In clear contrast to wild type SopE, secretion of SopEAia3o-42 and SopEAia44-54 via the SPI-1 and the flagellar export system did not require InvB. These data reveal a novel function of the CBD: The CBD inhibits secretion of wild type SopE via the flagellar and the SPI-1 TTSS in the absence of the chaperone InvB. Our data provide new insights into mechanisms ensuring specific effector protein transport by TTSS. This work furtherdemonstrated how S. Typhimuriumensures efficient delivery of effectors and swift manipulationof host cell signalling for the beneficial outcome of the bacteria cell interaction. Recently, by using time lapse microscopy, the delivery of the

4 SUMMARY Salmonella type IM effector protein SipA into animal cells was imaged in real time. Yet, nothing is known about the delivery kinetics of other type IM effector proteins. A key question concerning TTSS function and host cell manipulation is if there is a hierarchy in transport of effector proteins and if so, which effector proteins are injected first, which ones later? Comparison of the delivery of the effector proteins SopE and SptP was of special interest since both effectors antagonize each other: SopE activates Rho GTPases while SptP inactivatesthem. To analyse the kinetics of SopE and SptP delivery, depletion of the preformed intrabacterial pool of the effector proteins was analysed using an optimized technique for immunofluorescence staining of intrabacterial proteins in combination with real time imaging of the infection process. Real time imaging revealed that type III transport of SipA and SopE Starts within seconds after docking of the bacteria to the host cell and ends after sec. In contrast, injection of SptP Starts after SipA/SopE have been injected completely, showing that there is indeed a transport hierarchy: effector proteins important for early stages of invasion are injected first, followed by effector proteins required for later stages. As mentioned before, a preformed pool of effector proteins is stored in the bacterial cytoplasm and injected rapidly into the host cell cytosol upon direct contact with the host cell. However, nothing is know about the entry portals Salmonella uses and if any receptors are involved in the early steps of invasion. A number of experiments were conducted to address this issue. The requirementfor the proton-gradient for TTS transport was also studied. In conclusion, this work demonstrates that proper and efficient transport depends on the CBD and that the temporal regulation of effector protein translocation is a prerequisite for swift manipulationof eukaryotic cells by the SPI-1 TTSS.

5 ZUSAMMENFASSUNG ZUSAMMENFASSUNG Während der Infektion einer Wirtszelle ist Salmonella Typhimuriumverschiedenen Umwelteinflüssen ausgesetzt. Mit Hilfe einer Vielzahl von Virulenzfaktoren überleben die Bakterien die feindseligen" Bedingungen im Wirt und entkommen dessen Abwehrmechanismen. Ein wichtiger Virulenzfaktor ist das auf der Salmonella Pathogenitäts-Insel 1" (SPI-1) kodierte Typ III Sekretionssystem (TTSS). Diese Transportsysteme kommen in vielen Gram-negativen pathogenenbakterien vor und werden verwendet, um bakterielle Effektorproteine in die Wirtszelle zu transportieren. Es gibt zwei Arten von Transport: Transport in das umgebende Medium (Sekretion) oder Transport in das Wirtszellzytosol (Translokation). Zwei Domänen am N- terminalen Ende der Effektorproteine entscheiden zwischen den beiden Transportarten: Die ersten 20 Aminosäuren (N-terminales Sekretionssignal, NSS) werden für die Sekretion benötigt, während für die Translokation sowohl das NSS, als auch die anschliessenden 100 Aminosäuren (Chaperon-Bindedomäne,CBD) und die Bindung des Chaperons and die CBD benötigt werden. Zwei verwandte TTSS das Virulenz-assoziierteTTSS und das werden parallel in S. Typhimuriumexprimiert, Flagellensystem. Daher beinhaltet der Transport von Effektorproteinen zwei Entscheidungen: Transport über das SPI-1 aber nicht das Flagellen-TTSS (= Spezifität) und Translokation in das Wirtszellzytosol anstelle der Sekretion in das umgebende Medium (Art des Transports). In dieser Arbeit wurde der Mechanismus, der für die Spezifität des Transports verantwortlich ist, untersucht. Konservierte Bereiche des NSS als auch der CBD von SopE wurden durch Alanin ersetzt und die Sekretion sowie die Translokation der Mutanten näher untersucht. Der EffektorSopE von S. Typhimurium wird spezifisch über das SPI-1 TTSS transportiert. Die Translokation von SopE ist abhängig vom Chaperon InvB. Das Austauschen der Aminosäuren (und zu einem geringerengrad 44-54) mit Alanin verhinderte die Bindung von InvB an SopE, die Translokation in die Wirtszelle und führte zur Sekretion des Effektors über beide Transportsysteme, dem SPI-1 und Flagellen- TTSS. Im Gegensatz zum Wildtyp SopE war die Sekretion von SopEAia3o-42 und SopEAia über beide TTSS unabhängig von InvB. Die hier präsentierten Daten zeigen zum ersten Mal eine neue Funktion der CBD: Die CBD verhindert die Sekretion von Wildtyp SopE über das SPI-1 und Flagellen TTSS in der Abwesenheit vom Chaperon. In der vorliegenden Arbeit werden zum ersten Mal neue Ideen vorgestellt, wie die Bakterien den spezifischen Transport von Effektorproteinen über

6 ZUSAMMENFASSUNG das richtige TTSS sichern. Desweiteren wird gezeigt, wie die Bakterien es schaffen Effektorproteine effizient in die Wirtszelle zu transportieren und damit eine schnelle Manipulationder Wirtszell-Signalkaskade gewährleisten. Mit Hilfe von time-lapse" Mikroskopie konnte der Transport des Effektorproteins SipA in eukaryotische Wirtszellen in Echtzeit verfolgt werden. Leider ist über die Transportkinetik anderer SPI-1 Effektorproteine nichts bekannt. Die Frage ob es vielleicht eine Hierarchie zwischen verschiedenen Effektorproteinen gibt, war hier von besonderem Interesse. Der Vergleich zwischen den beiden SPI-1 Effektorproteinen SopE und SptP war besonders wichig, da beide Proteine eine entgegengesetzte Wirkung auf die Wirstzelle haben: SopE aktiviert eukaryotische Rho GTPasen, um die bakterielle Invasion zu gewährleisten, während SptP sie wieder deaktiviert. Effektorproteine werden bereits vor dem Kontakt mit der Wirtszelle synthetisiert. Um die Kinetik des Transports von SopE und SptP zu untersuchen, wurde die Depletion dieser vorgefertigten Proteine aus dem bakteriellen Zytosol mit Hilfe einer optimierten Immunofluoreszenzfärbung in Kombination mit Echtzeit-Aufnahmen verfolgt. Es wurde gezeigt, dass die Translokation der Effektorproteine SopE und SipA innerhalb weniger Sekunden nachdem die Bakterie an die Wirstzelle gebunden hat, startet und nach Sekunden die gesamte Menge an Effektorproteinen vollständig transportiert wurde. Im Gegensatz dazu beginnt die Translokation von SptP erst wenn SipA oder SopE fast vollständig in die Wirtszelle injiziert wurden. Dies zeigt zum ersten Mal, dass tatsächlich eine Hierarchie zwischen verschiedenen Effektorproteinen existiert und Effektorproteine, die wichtig für den Beginn der Infektion sind, zuerst transloziert werden, gefolgt von Effektorproteinen die wichtig für spätere Etappen der Invasion sind. Wie bereits erwähnt, wird die vorgefertigtemenge an Effektorproteinen sehr schnell und sehr effizient in die Wirtszelle transportiert, sobald die Bakterie Kontakt mit der Wirtszelle hat. Es ist jedoch nicht bekannt, welche Wirtszellfaktorenwichtig für den ersten Kontakt von S. Typhimuriumsind. Um mögliche Wirtszellfaktoren zu identifizieren, wurden verschiedene zellbiologische Experimente durchgeführt. Die Abhängigkeit des TTSS vom Protonengradienten wurde ebenfalls näher untersucht. Die Ergebnisse dieser Arbeit verdeutlichen, dass eine effiziente Wirtszell-Manipulation durch die schnelle Translokation von Effektoren über das richtige TTSS und die zeitliche Regulationder Translokation erreicht wird. 10
