in Summary Cell proliferation plays a very important role in carcinogenesis and is induced by many nongenotoxic carcinogens. It is of importance to identify mitogenic chemicals specifically hepatomitogens, as the liver is the main target organ for nongenotoxic carcinogens in long-term rodent carcinogenicity studies. The question arose as to whether the mitogenic activity of a chemical can already be detected in the premitotic phase. Two systems were used: the decondensation of hepatocyte nuclei detected by quinacrine dihydrochloride (QDH) fluorescence measurements and the expression of two immediate-early genes. Isolated rat hepatocytes in culture were exposed to the endogenous mitogen epidermal growth factor (EGF), the differentationinducing compound retinoic acid and the three nongenotoxic carcinogens cyproterone acetate, phenobarbital and thioacetamide. After 6 h, 24 h or 48 h the QDH fluorescence of nuclei of the three hepatocyte subpopulations (2N, 4N and 8N ploidy groups) was determined by means of image analysis. Dose-response relationships were established at physiological conditions (periportal and perivenous oxygen tensions). A 6-hour treatment with EGF resulted in a significant decrease in nuclear QDH fluorescence (GO-Gl transition), whereas after exposure to retinoic acid an increase in QDH fluorescence was observed. The nongenotoxic rodent liver carcinogens, cyproterone acetate, phenobarbital and thioacetamide all accomplished a significant, dose-dependent decrease in QDH fluorescence of the hepatocyte nuclei after 6 h. At this time-point the data are the most reliable, since the QDH fluorescence is increasing again towards the S phase. Cyproterone acetate-treated hepatocytes showed the largest decreases in QDH fluorescence after all three exposure intervals, especially under periportal oxygen conditions. Cyproterone acetate induced primarily a cell cycle entry (GO-Gl transition) and not a progression into the S phase. The effects of phenobarbital were not so dramatic and showed no difference with respect to the two culture conditions. The highest decreases in QDH fluorescence after treatment with phenobarbital were found after 6 h, implying a property of phenobarbital to induce a rapid entry into the cell cycle. Thioacetamide resulted in a maximal decrease in QDH fluorescence after 48 h, indicating a slow entrance into the cell cycle. clearly dose- and oxygen tension-dependent. IV return. When the cells are already in the S phase after longer treatment (24 or 48 h) with potent mitogens, smaller decreases in QDH fluorescence are observed. The second approach for the detection of mitogenic chemicals in the premitotic phase of the cell cycle is based on the transient increased expression of a set of genes, the immediate-early genes, within minutes after a growth stimulus (G0-G1 transition). The two nuclear proto-oncogenes c-/os and c-myc belong to this set of genes and 1 to 3 h, respectively. and are induced after 15 to 40 min RNA from the cultured, treated hepatocytes was isolated and hybridized on Northern blots with c-myc or c-fos probes. The induction of c-/os (after 40 min 10-fold) and c-myc (after 3 h 3-fold) observed after treatment with cyproterone acetate correlated well with the strong decrease in QDH fluorescence demonstrated by these hepatocytes. However, after exposure to phenobarbital only a slight increase in c-/os and c-myc mrna was observed. These discrepancies with the QDH analysis were further investigated by in situ hybridization with c-myc. A smaller proportion of hepatocytes expressed c-myc in high levels (>80 grains/cell) after phenobarbital treatment than after cyproterone acetate treatment. In contrast to the Northern blot analysis the mean labelling with c-myc in these experiments was also 3-fold increased after phenobarbital cyproterone acetate. treatment as was the case with The close correlation between the cytological marker, QDH fluorescence, and the expression of c-myc and c-/os appears to make the measurement of both parameters a potential efficient screening system for premitotic events.
V Zusammenfassung Zellproliferation spielt eine grosse Rolle in der Kanzerogenese und wird von vielen nicht-genotoxischen Karzinogenen induziert. Daher ist es von grosser Bedeutung, ein Testsystem zu entwickeln, das solche Substanzen erfassen kann. Speziell die Lebermitogene sind von grosser Bedeutung, da die Leber das Hauptzielorgan fur nicht-genotoxische Karzinogene in Langzeit-Kanzerogenese Studien ist. Der Frage, ob die mitogene Aktivitat einer Substanz schon in der pramitotischen Phase nachgewiesen werden kann, wurde nachgegangen. Dazu wurden zwei Methoden benutzt: Dekondensierung von Hepatozytenkernen, gemessen mit dem Fluoreszenzfarbstoff Quinacrine Dihydrochloride (QDH), und Expression von zwei "immediate-early" Genen. Isolierte Rattenhepatozyten wurden in vitro mit dem endogenen Mitogen epidermal growth factor (EGF), der differenzierungsinduzierenden Substanz Vitamin A Saure und den drei nichtgenotoxischen Leberkarzinogenen, Phenobarbital, Cyproteron Acetat und Thioacetamid behandelt. Nach 6, 24 und 48 Stunden wurde mit Hilfe der Bildanalyse die QDH-Fluoreszenz der Kerne fur jede der drei Ploidie- Gruppen (2N, 4N und 8N) bestimmt. Dosis-Wirkung Beziehungen wurden unter physiologischen Bedingungen (periportalen und perivenosen Sauerstoff-Partialdruck) bestimmt. Behandlung mit EGF bewirkte nach 6 Stunden eine signifikante Abnahme der QDH-Fluoreszenz in den Kernen (G0-G1 Ubergang), wahrend nach Behandlung mit Vitamin A Saure eine Zunahme der Fluoreszenz beobachtet wurde. Behandlung mit jedem der nicht-genotoxischen Leberkarzinogenen verursachte eine dosis-abhangige Abnahme der QDH-Fluoreszenz nach 6 Stunden. Die Ergebnisse nach 6 Stunden Behandlung sind am zuverlassigsten, da die QDH-Fluoreszenz ab der spaten Gl-Phase wieder zunimmt. Kerne der Cyproteron Acetat-behandelten Hepatozyten zeigten die grossten Abnahmen der QDH-Fluoreszenz und zwar bei alien drei Behandlungszeiten. Die Effekte waren noch deutlicher wenn die Hepatozyten unter periportalem Sauerstoff-Partialdruck kultiviert wurden. Cyproteron Acetat verursachte in den Hepatozyten in erster Linie einen Eintritt in den Zellzyklus (G0-G1 Ubergang) und weniger eine Progression in die S-Phase. Die Effekte von Phenobarbital waren weniger ausgepragt. Kein Unterschied zwischen den beiden Sauerstoff Bedingungen konnte festgestellt werden.
VI Die grossten Abnahmen der QDH-Fluoreszenz wurden nach 6 Stunden Behandlung Zellzyklus lmphziert mit Phenobarbital beobachtet was einen raschen Eintntt in den Thioacetamid hingegen zeigte die grossten Effekte erst nach emer Behandlungsdauer von 48 Stunden (langsamer Eintntt in die Gl-Phase) Die QDH-Fluoreszenz war eindeutig Dosis- und Sauerstoff-Partialdruckabhangig Die pramitotische Phase, detektiert anhand der Abnahme der QDH- Fluoreszenz nach 6 Stunden Behandlungszeit, reprasentiert Zellzyklus, die vor dem "Point of no return" hegt eine Phase un Befindet sich eine grossere Anzahl Zellen durch eine langere Behandlung (24 oder 48 Stunden) mit einem potenten Mitogen schon in der spaten Gl-Phase, resultiert das in einer genngeren Abnahme der Fluoreszenz Die zweite Methode, den Eintntt in den Zellzyklus in der pramitohschen Phase zu erfassen, basiert auf dem Nachweis der Expression von "immediate-early" Genen Diese Gruppe von Genen, zu der auch die nuklearen Proto-Onkogene c-fos und c-myc gehoren, werden innerhalb von Minuten nach einem Wachstumsstimulus (G0-G1 Ubergang) induziert c-fos Transknpte werden nach 15-40 Minuten, c-myc Transkripte Stunden stark vermehrt vorgefunden nach 1-3 RNA von kultivierten, behandelten Hepatozyten wurde isoliert und auf Northern Blots mit c-fos oder c-myc Sonden hybridisiert Nach Behandlung mit Cyproteron Acetat konnte eine Induktion von c-fos (nach 40 Minuten 10-fach) und c-myc (nach 3 Stunden 3-fach) nachgewiesen werden, die sehr gut mit der beobachteten starken Abnahme der QDH-Fluoreszenz korreherte Nach Behandlung mit Phenobarbital hingegen wurde nur eine geringfugige Zunahme der c-myc und c-fos Transkripte detektiert Dieser Widerspruch zu den Resultaten der QDH-Analyse wurde mit Hilfe der in situ Hybndisierung mit einer c-myc Sonde untersucht Nach Phenobarbital-Behandlung expnmierte ein kleinerer Prozentsatz der Hepatozyten c-myc in sehr hohen Mengen (>80 grains/zelle) als nach Behandlung nut Cyproteron Acetat Im Gegensatz zu den Resultaten der Northern Blot Analyse war der mittlere Markierungsgrad der Hepatozyten nach Phenobarbital Behandlung wie bei Cyproteron Acetat dreifach erhoht Durch die eindeutige Korrelation der zytologischen Marker QDH- Fluoreszenz mit der Expression von c-fos und c-myc konnte die Messung dieser beiden Parameter zu einer effizienten Screerungsmethode fur pramitotische Ereignisse werden
