98 6. SUMMARY 1. A method was developed to study the interactions between flavor compounds and denatured proteins in various suspending 14... media. Use of Clabeled flavor compounds permits investigation of these interactions at the low concentration levels typical of foods. 2. The interaction between flavor compounds and denatured proteins can be described as weak adsorption of the former on the latter, probably by an equilibriumtype mechanism. Adsorption is independent of temperature, ph and ionic strength of the medium. In the concentration range examined, adsorption is directly pro portional to the amount of protein and increases linearly with increasing ligand concentration. 3. Denatured BSA is an acceptable model for a food protein. Its adsorption behavior was comparable to that for the soy isolate Soyamin 90 in all media studied. 4. Preparation of the denatured protein is not a determining factor in predicting adsorption behavior. The temperature of denaturation, presence of endogenous lipids, moisture content and nature of drying were important only to the extent that they af fected protein hydration. 5. Complexes of native BSA were made with emulsifiers, fatty acids or butter and denatured. They adsorbed ligand to the same extent, if not more, as the corresponding amount of pure denatured protein. 6. Neither suspending denatured protein in media containing emulsifiers, oils or fats, nor coating it with these substances proved to be an effective method for reducing adsorption. 7. Suspending denatured protein in media containing caseinhydrolysate did not affect the amount of ligand adsorbed by the denatured protein. Sodium caseinate, however, slightly decreased adsorption. 8. Denatured protein suspended in O/Wcasein emulsions showed oa. 50 % less adsorption than buffer suspensions of the protein.
" 99 9. Reconstituted nondairy milk powders such as coffee whiteners reduced adsorption to denatured proteins suspended in them by oa. 50 %. 10. Adsorption was also reduced by aa. 50 % when denatured proteins were suspended in milk, milk drink and coffee cream. Skim milk proved less effective, as did buttermilk. 11. It is suggested that the combination of casein and lipid in the suspending media creates surfaces which interact with flavor compounds thus competing with the denatured protein and shifting the equilibrium away from it. These surfaces correspond to the lipoprotein membranes found in buttermilk, which explains why adsorption was slightly decreased in pre parations of reconstituted buttermilk. Lipoprotein membranes are usually present in natural foods. Their absence could explain the difficulty encountered in flavoring synthetic foods.
100 6. ZUSAMMENFASSUNG 1. Es wurde eine Methode entwickelt, die es gestattet, die Wechselwirkungen zwischen Aromastoffen und denaturierten Proteinen 14 in verschiedenen Suspensionen zu studieren. Der Einsatz von C markierten Aromastoffen erlaubt diese Wechselwirkungen bei tiefen Konzentrationen zu untersuchen, wie sie bei Lebensmitteln typisch sind. 2. Die Wechselwirkungen zwischen Aromastoffen und denaturier ten Proteinen konnen als schwacher Adsorptionsvorgang beschrieben werden. Vermutlich handelt es sich um einen Mechanismus in der Art eines Gleichgewichtes. Die Adsorption ist unabhangig von der Temperatur, von ph und der lonenstarke des Mediums. Die Adsorption ist im untersuchten Konzentrationsbereich direkt proportional zur eingesetzten Menge Protein und wachst mit steigender Ligandkonzentration linear an. 3. Denaturiertes BSA ist ein akzeptables Modell fur ein Protein eines Lebensmittels. Sein Adsorptionsverhalten ist in alien unter suchten Medien vergleichbar mit dem aus Soya isolierten Soyamin 90. 4. Die Herstellung von denaturiertem Protein hat keinen entscheidenden Einfluss auf das Adsorptionsverhalten. Die Temperatur des Denaturiervorgangs, die Gegenwart endogener Lipide, Feuchtigkeitsgehalt und die Trocknungsart sind insofern zu beachten als sie die Hydratation des Proteins verandern konnen. 5. Komplexe mit nativem BSA wurden in Gegenwart von Emulgatoren, Fettsauren oder Butter hergestellt und anschliessend denaturiert. Sie adsorbierten Liganden im selben Mass, oder starker, wie die entsprechende Menge des reinen denaturierten Proteins. 6. Weder das Suspendieren denaturierter Proteine in wasserigen Systemen mit Emulgatoren, Oelen oder Fetten noch das Belegen der Proteinoberflache mit diesen Verbindungen erwiesen sich als wirksame Methoden, um die Adsorption herabzusetzen.
101 7. Suspensionen denaturierter Proteine in Caseinhydrolisatlosungen beeinflusste die Adsorption der Liganden an das denaturierte Protein nicht. Hingegen setzte Natriumcaseinat die Adsor ption leicht herab. 8. In Oel/WasserCaseinEmulsionen suspendiertes denaturiertes Protein zeigte eine ca. 50 % geringere Adsorption als gepufferte Proteinsuspensionen. 9. Rekonstituierte kiinstliche Milchpulver, wie "coffee whiteners" reduzieren die Adsorption in Suspensionen denaturierter Proteine um ca. 50 %. 10. Die Adsorption wurde ebenfalls um 50 % herabgesetzt, wenn die denaturierten Proteine in Milch, Milchdrink und Kaffeerahm suspendiert wurden. Magermilch und Buttermilch stellten sich als weniger wirksames Medium heraus. 11. Es wird angenommen, dass die Kombination von Kasein und Lipiden in Suspension Oberflachen erzeugt, Protein um die Aromastoffe konkurrieren. Diese Oberflachen entsprechen den Lipoproteinmembranen wie sie die mit dem denaturierten in Buttermilch gefunden wurden. Das erklart, weshalb die Adsorption wahrend der Versuche mit rekonstituierter Buttermilch leicht herabgesetzt war. Lipo proteinmembranen kommen gewohnlich in natiirlichen Nahrungsmitteln vor. Ihr Fehlen in synthetischen Nahrungsmitteln konnte die Schwierigkeiten bei der Aromatisierung dieser Produkte erklaren.
