3 Abstract 3 Abstract Type 2 diabetes is characterized by hyperglycemia that is caused by the inability of the pancreatic β cells to meet the increased insulin demands that are associated with insulin resistance. β cell failure originates from both, a gradual deterioration of insulin secretory capacity and a decline in β cell mass. Transmembrane protein 27 (Tmem27), a β cell specific cell surface protein, has previously been shown to promote β cell proliferation, a process that seemed to be shut off through ectodomain cleavage by a cell surface protease. Thus it was postulated that if this protease could be inhibited, Tmem27 concentrations and thereby β cell expansion could be augmented and type 2 diabetes reversed or delayed in patients with a remaining population of β cells. The identification of Beta site amyloid precursor protein cleaving enzyme 2 (Bace2) via a small interfering ribonucleic acid (sirna) screen as the primary Tmem27 protease in MIN6, a murine pancreatic β cell line, is described in Section 6. Downregulation of Bace2 led to decreased levels of shed Tmem27 in the cell supernatant and increased full length Tmem27 in the cell, while overexpression of Bace2 led to elevated Tmem27 cleavage products and full length protein destabilization. These effects were not elicited by Bace1, the close homologue of Bace2. The sites of co localization and the molecular interaction between Bace2 and Tmem27 are unravelled in Section 7. Contrary to Bace1, which displays a broad expression pattern, Bace2 mrna and protein were mainly found in peripheral organs with neuroendocrine cell populations and were most enriched in the pancreatic β cell, while absent from the kidney, the other major organ of Tmem27 expression. Bace2 localized to non insulin granules near the β cell surface, as did Tmem27. Since dimeric Tmem27 was resistant to Bace2 cleavage, Bace2 is likely to interact with and cleave Tmem27 in its monomeric form. Section 8 elucidates the effect of genetically inactivated Bace2 on glucose homeostasis and pancreatic β cell mass in mice. Mice expressing only a catalytically inert form of Bace2, Bace2ΔE6, which lacks exon 6 that encodes the catalytically active aspartyl residue D303, displayed ameliorated glucose homeostasis that correlated with elevated full 12
3 Abstract length Tmem27 in the islet, increased serum insulin levels and augmented β cell proliferation and mass. The characterization of the Bace2 selective chemical inhibitor Compound J that targets β cell Bace2 in vivo is presented in Section 9. The improvement in glucose tolerance and acceleration of β cell replication upon treatment with this compound in C57BLKS db/db and C57Bl6 ob/ob mice, two mouse models of obesity dependent insulin resistance and diabetes, are evidenced in Section 10. Overall, the experiments here described identify Bace2 as a potential drug target for improving pancreatic β cell function and mass in diabetic settings. 13
4 Zusammenfassung 4 Zusammenfassung Typ 2 Diabetes ist von einer Hyperglykämie gekennzeichnet, die sich in der Insuffizienz der pankreatischen β Zelle, im Zuge der Insulinresistenz genügend Insulin zu sekretieren, begründet. Ursprung dieses Defizites ist sowohl der graduelle Verlust der sekretorischen Funktion der β Zelle, als auch das Abnehmen an β Zellmasse. Eine vorhergehende Studie hat gezeigt, dass Transmembranprotein 27 (Tmem27), ein in β Zellen angereichertes Transmembranprotein, β Zellproliferation stimulieren und somit diesem Rückgang potentiell entgegenwirken kann. Da Tmem27 außerdem von einer Zelloberflächenprotease geschnitten und inaktivert zu werden schien, lag die Hypothese nahe, dass die Inhibierung dieser Protease zu erhöhten Tmem27 Konzentrationen, demzufolge vermehrter β Zellmasse und somit zur Verbesserung oder Verzögerung von Typ 2 Diabetes führen könnte. Mittels einer systematischen Herunterregulierung von Proteasen durch kleine interferierenden Ribosenukleinsäuren (RNS) wurde Beta site amyloid precursor protein cleaving enzyme 2 (Bace2) als Tmem27 Protease in der MIN6 Zelle, einer murinen β Zelllinie, identifiziert (Sektion 6). Die Abwesenheit von Bace2 hatte eine Verminderung von sezeniertem Tmem27 im Zellüberstand und eine Anreicherung von intaktem Tmem27 im Zellysat zur Folge, während die Überexpression von Bace2 zur Destabilisierung von Tmem27 führte. Diese Effekte konnten von Bace1, dem homologen Protein von Bace2, nicht erzielt werden. Die Orte der Ko existenz sowie die molekulare Interaktion zwischen Bace2 und Tmem27 werden in Sektion 7 ermittelt. Im Gegensatz zu Bace1, das weit verbreitet vorzufinden ist, wurden Bace2 mrns und Protein hauptsächlich in Organen mit einer neuroendokrinen Zellpopulation, insbesondere in den β Zellen der pankreatischen Insel, detektiert, während es für Expression von Bace2 in der Niere, dem anderen Organ mit signifikanter Tmem27 Expression, keine Anzeichen gab. Ähnlich wie Tmem27 war Bace2 in plasmamembran nahen Vesikeln angereichert, die allerdings kein Insulin enthielten. Nachdem sich Dimere von Tmem27 resistent gegen proteolytische Degradation durch Bace2 verhielten, ist das Tmem27 Monomer wahrscheinliches Interaktionsmolekül und Substrat von Bace2. 14
4 Zusammenfassung In Sektion 8 wird der Effekt genetischer Inaktivierung von Bace2 auf Blutglukoseregulierung und pankreatische β Zellmasse in Mäusen untersucht. Mäuse, die nur eine katalytisch inaktive Form von Bace2 besitzen Bace2ΔE6, dem das Exon 6 und somit der katalytisch kritische Aspartylrest D303 fehlt wiesen eine verbesserte Glukosehomeostase auf, die sowohl mit Stabilisierung von Tmem27 in der pankreatischen Insel, als auch mit gesteigerter Seruminsulinkonzentration sowie β Zellproliferation und masse korrelierte. Die Charakterisierung des Bace2 selektiven chemischen Inhibitors Compound J, der Bace2 in β Zellen in vivo inaktiviert, ist in Sektion 9 erörtert, während die erhöhte Glukosetoleranz und β Zellteilung und masse in mit diesem Wirkstoff behandelten C57BLKS db/db und C57Bl6 ob/ob Mäusen, zwei Mausmodellen für fettleibigkeitsbedingte Insulinresistenz und Diabetes, in Sektion 10 erläutert sind. Insgesamt identifizieren die hier beschriebenen Experimente Bace2 als ein neues potentielles Target zur Steigerung von β Zellfunktion und masse im Kontext des Diabetes. 15
