2 New techniques developed to edit genomes in a more targeted way than previous GE techniques Recognizes 3 basepairs (nucleotidetriplets) Recognizes single nucleotides (1 basepair) RNA guided editing tool for genome engineering. Can delete larger pieces of DNS

3 POSSIBILITIES OF NEW GENETIC MODIFICATION TECHNIQUES ZINC FINGER, TALEN, CRISPR. All are derived from bacterial gene repair and defense mechanisms against viral intruders Most work via gene disruption and silencing/deleting genes A few (e.g. ZFN3) can at best transfer/ edit (silence/delete) single genes = simple traits as limited possibilities as before

4 RISKS OF NEW GENETIC MODIFICATION TECHNIQUES INTERESTINGLY! All praise site-specificity as superior to random insertion of earlier transgenic technologies. Note: Previously, site-specificity was deemed irrelevanttransgenic technologies claimed superiority over breeding because of known sequence of inserted piece of DNA

5 Postulated advantages: Site - specificity superior to random insertion of known sequence! Increased precision improved control more safety! No recombinant gene transfer necessary more safety No baggage from regulatory and other elements more safety Aim: generate a novel organism not considered GMO to avoid regulations BUT still novel invention to enjoy strict private property rights PATENTS

9 State of disconnect between knowledge groups Applied engineering field of molecular biology still builds on central dogma : - Genetic information is conveyed by DNA sequences only (construction instructions) and - a particular DNA sequence will code for the same and the only gene product - in both the donor and the recipient organism - Plasticity, variability and diversity are problems and road blocks

Die Bindung der Regulatorproteine Tcf7 (rot), Lef1 (violett), Brachyury (grasgrün), Tbx6 (dunkelgrün), Snail1 (türkisblau), Srf (marineblau) an die Promotoren ihrer Zielgene im kaudalen Bereich von Mausembryonen wurde mithilfe der ChIP Methode sichtbar gemacht und nach bioinformatischen Analysen in Form eines Netzwerks dargestellt. Ein Teil der Zielgene wird von mehreren Regulatoren gemeinsam, andere von einzelnen kontrolliert.
