Beschreibung: Pancreatic carcinoma (PC) is one of the leading causes of cancer-related deaths. The 5-year survival rate of patients is estimated at 3%-5%. Poor survival can be attributed to aggressive tumor growth and a high incidence of metastasis. Potential treatment options include radiotherapy and chemoradiotherapy. Essential steps during metastasis are cell migration and invasion. Both processes include a variety of signalling processes typically resulting in the regulation of integrins, matrix-degrading enzymes, and cell-cell adhesion molecules. Furthermore epithelial-mesenchymal transition (EMT) drives the early phase of cancer metastasis. EMT induces changes in the shape and motility of epithelial cells. Once transforming into mesenchymal phenotype, cancer cells lose their cell-cell contacts and become mobile and invasive in order to spread into nearby tissues and subsequently distant organs. Recently, mirnas, which are fine tuners of gene expression, have been reported to be major components in the regulation of the invasive and metastasizing phenotype of cancer cells. So far several in vivo and in vitro studies found that ionizing radiation might influence migration and invasion of tumor cells. However, results are controversial and the underlying mechanisms are poorly understood. Task 1: Radiation triggered cell movement Three pancreatic cancer cell lines differing in radiosensitivity will be irradiated with clinically relevant doses (0; 2; 4; 8 Gy). Potential radiation effects on cell migration and invasion will be measured either by the classical wound healing assay (migration) or by using a transwell chamber assay (invasion). Task 2: Biochemical characterization of radiation effects on migration To gain insights into radiation-triggered pathways, key processes of cell movement will be analyzed on the molecular level: - Matrixmetalloproteinase activity (zymography, protein quantification) - Signalling pathway activation (Akt/PI3K, NFkappaB, p38 activation; fluorescence microscopy) - Measurement of EMT transition (quantification of epithelial versus mesenchymal marker proteins on RNA and protein level, e.g. N- E- cadherin, vimentin) Task 3: Manipulation of radiation-triggered migration and invasion micrornas with validated impact on migration (e.g. mir-21, mir-10b, mir-31) will be overexpressed or blocked (transfection of mirna mimics or antagomirs) and their interrelationship with radiation in migration and invasion will be measured. Christine Preibisch 1 Projekt Projekt-ID: Thema: How do mild cognitive deficits arise in patients with high grade carotid artery stenosis: a multiparametric functional MRI study Beschreibung: Though asymptomatic in terms of stroke, patients with high-grade carotid artery stenosis (CAS) often suffer from mild cognitive deficits, especially attention impairments. In these patients, perfusion and cerebrovascular reactivity are typically reduced in the hemisphere ipsilateral to the stenosis, which is reversible by treatment such as endovascular stenting or surgical endarterectomy. To date, however, it is unknown whether and how cognitive deficits of CAS are linked with such asymmetric physiological impairments. Individual differences in basic attentional mechanisms including attentional asymmetries across visual hemifields are associated with the synchronicity of slowly ongoing brain activity, which is called intrinsic functional connectivity (FC). FC can be measured by correlated blood oxygenation level dependent (BOLD) fluctuations of resting-state functional magnetic resonance imaging (fmri). We hypothesize that asymmetries in FC mediate the link of asymmetric perfusion and neurovascular coupling with attention deficits in CAS. In order to explore the influence of altered perfusion and neurovascular coupling on BOLD-based FC and cognition, it is necessary to acquire relevant information about vascular reactivity, perfusion and oxygenation as well as cognitive performance. This can be achieved within a single assessment session comprising novel, advanced physiological MRI and psychophysical examinations of basic attentional mechanisms. Our current multi-parametric MRI protocol comprises measurements of BOLD-FC (restingstate-fmri), cerebrovascular reactivity (breathhold fmri), perfusion (non-invasive arterial spin labeling and contrast agent based MRI), brain oxygenation, structural MRI (diffusion tensor imaging), lesion detection and morphometry. These measurements are performed in patients with CAS before and after surgery to enable causal analysis of the link between physiology, BOLD-FC and cognitive changes. In detail, the major aim of this study is a comprehensive investigation of attentional and physiological impairments in patients with high-grade CAS. Specifically, we plan to explore to which extent reductions in BOLD-FC in patients with CAS might be caused by physiological impairments and mediate the link to attentional deficits. In order to explore the extent to which these impairments are reversible after treatment, longitudinal MRI and attentional assessment are planned in patients with high-grade CAS before and after treatment. Machbarkeit: The doctoral candidate will be able to address relevant research questions regarding the interplay between physiology and cognition based on existing data and software. After amendment of the existing ethics approval, the candidate will also be involved in patient recruitment and data acquisition. The experience and results obtained within this project greatly benefited the development of the oxygenation sensitive method, which is currently employed in the multi-parametric MRI study in patients with high-grade CAS, being funded by the Dr.-Ing. Heiko Lickert 1 Projekt Projekt-ID: Thema: Evaluation of beta cells in a humanized mouse transplantation model Beschreibung: For insulin-dependent type-1-diabetes meanwhile there are advanced options for therapy, such as artificial pancreas and adaptive insulin pumps. However, no curative approach (beta-cell replacement, bio-artificial pancreas) for routine application in humans is available yet. A number of research projects aim at in vitro generation of functional beta-cells. At the IDR we succeded in the generation of beta-like cells with various genetic make-ups. The overarching goal of this research proposal is to establish a "humanized"model of beta cell replacement therapy to investigate source materials, such as in vitro differentiated human beta-cell or islet preparations with various gene expression profiles and maturation stages model regarding their functionality, i.e. the regulation of blood glucose levels in vivo. The results of this project shall be relevant to support the choice of treatment regimen for type 1 diabetes mellitus in human subjects. The work plan will cover the following steps: - Establish transplantation assay ("humanized"mouse model) - Examine survival, functionality, and safety of transplants - Extrapolate findings to support development of clinical study protocols in human Machbarkeit: The regulatory authorization for the mouse transplantation model will be in place. 13 Promotionsprogramm Translationale Medizin Antragsteller: Ellen Renner 1 Projekt Projekt-ID: Thema: Funktionsanalyse in ips-zellen von STAT3-HIES Patienten Beschreibung: STAT3 Hyper-IgE Syndrom (STAT3-HIES) ist ein seltener angeborener Immundefekt, verursacht durch heterozygote Mutationen im STAT3 Gen. Das mutierte STAT3-Protein führt hierbei zu einem dominant negativen Effekt auf die STAT3 Funktion, welche eine zentrale Rolle in der Immunregulation insbesondere für Zelldifferenzierung und Entzündungsregulation hat. Da eine hämatopoetische Stammzelltransplantation für STAT3-HIES nicht in gleichem Maße erfolgversprechend ist wie bei anderen angeborenen Immundefekten, haben wir uns zum Ziel gemacht die STAT3-HIES Patienten mittels Genreparatur zu heilen. Um diesem hochgesteckten Ziel näher zu kommen fokussiert sich diese Doktorarbeit darauf die Differenzierung von naiven T-Zellen zu TH17-Zellen in HIES-Patienten wiederherzustellen. Denn es ist bekannt, dass die gestörte STAT3-Funktion beim STAT3-HIES zu einer stark reduzierten Anzahl von TH17-Zellen führt. Die Hypothese dieses Projektes ist, dass ips-zellen (ipscs) von STAT3-HIES-Patienten im Gegensatz zu Wildtyp-iPSCs nicht oder nur ungenügend zu TH17-Zellen differenziert werden können. Nach erfolgreicher Genreparatur sollte es hingegen möglich sein ipscs von STAT3-HIES-Patienten zu funktionsfähigen TH17-Zellen zu differenzieren. Ziel dieser Arbeit ist somit die Differenzierung von ipscs zu TH17-Zellen zunächst mit Wildtyp-Zellen zu etablieren, die TH-Zelldifferenzierung aus Wildtyp- und STAT3-HIES-iPSCs zu vergleichen und schließlich die Genreparatur von STAT3-HIES-iPSCs funktionell mit Hilfe der TH-Zelldifferenzierung zu untersuchen. Angewandte Methodik: immunologische Funktionsanalyse wie Durchflusszytometrie, Westernblotting und real-time-pcr, Zelldifferenzierung durch Zytokinstimulation, ips-zellkultivierung, Sequenzierung Aufgaben des Doktoranden: Um den Erfolg einer STAT3-Genreparatur funktionell untersuchen zu können, sollen im Rahmen dieses Projektes aus ipscs naive CD4+ T-Zellen und daraus dann verschiedene T-Helfer-Subtypen (v.a. TH17-Zellen) differenziert werden. Hierzu sollen zunächst Wildtyp-iPSCs und später unveränderte und reparierte ipscs von STAT3-HIES-Patienten verwendet werden. Anspruch: Experimentelle Doktorarbeit mit dem Ziel funktionelle Zellen nach Genreparatur nachzuweisen auf dem Weg zur Etablierung eines gentherapeutischen Ansatzes für STAT3-HIES. Ein kumulativer Abschluss mit dem Verfassen von Erst- und Koautorenpaper ist somit im vorgegebenen Zeitraum gegeben. Machbarkeit: Die ips-zellkultivierung erfolgt gemeinsam mit der ipsc Core Facility des HMGU München, so dass direkt mit der Etablierung der TH-Zelldifferenzierung aus ipscs begonnen werden kann. Dr. Rainer Haseneder 1 Projekt Projekt-ID: Thema: Optogenetische Untersuchungen zum Einfluss der Anästhetika Dexmedetomidin und Sevofluran auf das thalamokortikale Netzwerk Beschreibung: Hintergrund: Der Mechanismus, wie Anästhetika zum Bewusstseinsverlust führen, ist nach wie vor unklar. Der Thalamus nimmt eine zentrale Funktion bei der Weitergabe sensorischer Informationen zum Kortex ein. Er wurde als SSchalterßwischen Bewusstsein und Bewusstseinsverlust bezeichnet (Alkire & Miller 2005). Eine Modulation dieser Region durch Anästhetika könnte ursächlich für deren hypnotische Wirkung sein. Nachdem sensorisch evozierte Potentiale im Kortex auch unter Anästhesie weiterhin auslösbar sind, erscheint jedoch eine komplette Blockade des Informationsflusses durch den Thalamus unwahrscheinlich. Die neuronale Aktivität des Thalamus wird hauptsächlich durch kortikale Feedback-Fasern gesteuert (Vahle-Hinz et al. 2007). Durch eine Hemmung derer könnte eine sekundäre Inaktivierung des Thalamus erklärt werden. Anästhetika beeinflussen jedoch auch die intrakortikale Signalverarbeitung (Jordan et al. 2013). Letztlich ist die Sensitivität der im thalamokortikalen Netzwerk kommunizierenden Hirnareale gegenüber Anästhetika nach wie vor unklar und soll im beschrieben Projekt untersucht werden. Fragestellung: Die Wirkung von Dexmedetomidin und Sevofluran auf das thalamokortikale Netzwerk soll detailliert untersucht werden: 1. Wie beeinflussen o.g. Anästhetika die thalamokortikale Transmission? 2. Wie beeinflussen o.g. Anästhetika das kortikothalamische Feedback? 3. Wie beeinflussen o.g. Anästhetika intrakortikale Transmission? 4. Zeigen die bei untersuchten Parameter unterschiedliche Sensitivität gegenüber den Substanzen? Angewandte Methodik: Mittels stereotaktischer Injektion wird ein viraler Vektor in den Thalamus/Kortex der Maus eingebracht. Dieser führt zur Expression des lichtsensitiven Kanals ChR-2. Anschließend wird in vitalen Hirnschnitten mittels Patch-Clamp-Technik der Einfluss o.g. Anästhetika auf die durch optogenetische Stimulation hervorgerufene synaptische Transmission im thalamokortikalen Netzwerk untersucht. Aufgaben des Doktoranden: Stereotaktische Virusinjektion, fluoreszenzmikroskopische Evaluation der Expression, elektrophysiologische Messungen, Auswertung der Ergebnisse sowie Publikation. Anspruch: Neben allgemeinem wissenschaftlichem Interesse erwarten vom Doktoranden Interesse an anästhesiologischneurophysiologischen Fragestellungen, manuelles Geschick, Verantwortungs-bewusstsein, Zuverlässigkeit und die Fähigkeit zum selbständigen Arbeiten sowie zur Arbeit im Team in einer etablierten Laborumgebung. The intracellular signaling pathways emanating from these innate immune sensors drive acute inflammatory responses that are critical for host defense. We have previously identified key immune pathways that are activated after nucleic acid recognition upon viral infection and beta-glucan sensing in the cell wall of fungal pathogens. These signaling pathways are controlled by a protein complex composed of CARD9 and BCL10 that mediates inflammatory cytokine production. However, the precise regulation of these mechanisms is unclear. Question. Here the molecular regulatory mechanisms that control CARD9/BCL10 signaling during immune responses will be explored. The anticipated results will yield basic insights into innate immune signaling with implications for host defense and immune pathology. Methods. Large innate immune signaling complexes are often negatively regulated through protein degradation via a process called selective autophagy. Independent studies have reported an inhibitory role for the autophagy regulator Rubicon on CARD9-mediated innate immune signaling. We produced a series of knock-out cell lines with mutations in specific autophagy regulators including Rubicon and Beclin-1. Activation of CARD9 signaling resulted indeed in enhanced cytokine responses in these cells. The student will use these already established autophagy-defective cells to precisely analyze the function of individual autophagy regulators in the negative control of CARD9 signaling. This project will employ the use of fluorescence microscopy, molecular biology, biochemical and immunological assays, that are all established in our laboratory Tasks of the student. The student will investigate the formation and degradation of CARD9/BCL10- containing protein complexes in the different autophagy-defective cell lines by fluorescence microscopy. The results will be corroborated with biochemical and immunological analyses. The student will learn to design and execute conclusive experiments and analyze and represent experimental data.
