It is particularly important in semen samples with less than 40% of the forward moving (motile) sperms. Principle The determination of the vitality of sperm cells is judged by the integrity of the sperm membrane. The dye exclusion method is based on the fact that dead sperms with damaged plasma membrane absorb certain dyes. Storage and stability 2-8 C 24 months from date of manufacture. Content Reagent 1 20 ml Reagent 2 30 ml Required utensils Gloves Immersion oil Microscope Native ejaculate or washed sperm (20-50 µl) Slides Paper towels Pipettes and tips ( µl) Test tubes (1.5 or 2 ml) Test tube holder Procedure This test should begin immediately after liquefaction of the semen sample, preferably after 30 minutes and not later than 60 minutes to avoid negative influences: 1. Pipette µl sperm ejaculate in a test tube. 2. Add 2 drops of reagent 1, mix (avoid foaming) and incubate at room temperature for 30 seconds. 3. Add 3 drops of reagent 2 and mix again. 4. Transfer 10 µl of the mixture to a slide, grease the mixture with a cover slip and let air dry. 5. Evaluate the sperms with immersion oil at 1000x magnification Evaluation Vital sperms appear colorless, transparent or light pink; dead or not viable cells are stained red. Sometimes in sperms within the colored neck region and not colored head and flagellum are observable- This caused by damage to the membrane and these sperm are classified as vital. Count 200 cells and distinguish between vital sperms from dead sperms. The total number of membrane-intact sperms is of biological significance. The value is determined by multiplying the total number of sperms in the ejaculate by the percentage of membrane-intact cells. Total number of vital sperms = Total number of sperms x percentage vital sperms Example Total number of sperms in ejaculate: 20 Million Percentage of vital sperms 55% or 0.55, respectively Total number of vital sperms = 20 Million x 0.55 = 11 Million Vital sperms are not necessarily motile. 3 sperms are living or dead cells. Test results should be made in connection with the evaluation of the motility of the same semen sample. Vital but immotile sperms may have structural defects in the flagellum. A high number of immotile and dead sperms (necrozoospermia) may indicate a dysfunction of the epididymis. The lowest reference value for the vitality of the sperms is 58% (WHO 2010). Safety information / precautions All semen samples should be considered potentially infectious. Handle with all samples like HIV or hepatitis infected material. When working with samples and reagents wear always protective clothing (gloves, gowns, eye / face protection). Reagent 1 is containing eosin Y and reagent 2 is containing nigrosine. Both substances are not classified as toxic References 1. ZR Anwendungen Gebrauchsanweisung BITTE SORGFÄLTIG LESEN (Spermien Vitalitäts Test) in vitro Diagnostik Anwendung Der dient zur Motilitäts- und Vitalitätsprüfung von Spermien. Er ist besonders wichtig bei Samenproben mit weniger als 40% sich vorwärts bewegenden (motilen) Spermien. Nur für den professionellen Gebrauch Durchführung Der Test sollte unmittelbar nach Verflüssigung der Samenprobe beginnen, möglichst nach 30 Minuten und nicht später als 60 Minuten. Damit werden negative Einflüsse vermieden. Prinzip der Methode Die Bestimmung der Spermienvitalität wird durch die Integrität der Spermienmembran beurteilt. Die Farbaus-schlussmethode basiert auf der Tatsache, dass tote Spermien mit geschädigter Plasmamembran bestimmte Farbstoffe aufnehmen. Lagerung und Haltbarkeit Inhalt 2-8 C 24 Monate ab Herstellungsdatum Reagenz 1 20 ml Reagenz 2 30 ml Benötigte Utensilien Handschuhe Immersionsöl Mikroskop mit 1000x Vergrößerung Nativejakulat oder gewaschene Spermien (20-50 µl) Objektträger Papiertücher Pipetten und Spitzen ( µl) Reagenzgefäß (1,5 oder 2 ml) Reagenzgefäßhalter µl Ejakulat in ein Reagenzgefäß pipettieren Tropfen Reagenz 1 dazugeben, mischen (Schaumbildung vermeiden) und 30 Sekunden bei Raumtemperatur inkubieren Tropfen Reagenz 2 hinzugeben und erneut mischen µl der Mischung mit einem Deckglas dünn auf einen Objektträger ausstreichen und an der Luft trocknen lassen. 5. Den Objektträger anschließend bei 1000x Vergrößerung mit Öl mikroskopieren. Auswertung Vitale Spermien erscheinen farblos, durchsichtig oder hellrosa; tote bzw. nicht vitale Zellen sind rot gefärbt. Gelegentlich findet man Spermien mit gefärbter Nackenregion und ungefärbtem Kopf und Flagellum, verursacht durch Schädigung der Membran. Diese Spermien werden als vital eingestuft. Zählen Sie 200 Zellen aus und differenzieren Sie die Lebendspermien von den toten Zellen. Die Gesamtzahl membranintakter Spermien im Ejakulat ist von biologischer Bedeutung. Der Wert wird ermittelt durch die Multiplikation der Gesamtzahl aller Spermien im Ejakulat mit dem Prozentsatz der membranintakten Zellen. 5 Beispiel Gesamtzahl an Spermien im Ejakulat: 20 Millionen Prozentsatz an Vitalspermien: 55% bzw. 0,55 Gesamtzahl an Vitalspermien = 20 Millionen x 0,55 = 11 Millionen Vitale Spermien sind nicht unbedingt motil. Daher ist es von klinischer Bedeutung, ob es sich bei immotilen Spermien um lebende oder tote Zellen handelt. Die Testergebnisse sollten in Zusammenhang mit der Bewertung der Motilität derselben Samenprobe erfolgen. Vitale aber immotile Spermien haben möglicherweise strukturelle Defekte im Flagellum. Eine hohe Anzahl an immotilen und toten Spermien (Nekrozoospermie) deutet möglicherweise auf eine Dysfunktion des Epididymis (Nebenhoden) hin. Fertility and Sterility 49: Sicherheitshinweise / Vorsichtsmaßnahmen (Bitte zusätzlich Sicherheitsdatenblätter hinzuziehen) Alle Samenproben sollten als potentiell infektiös betrachtet werden. Behandeln Sie alle Proben so, als ob sie HIV oder Hepatitis übertragen könnten. Tragen Sie immer Sicherheitskleidung, wenn Sie mit Proben und Reagenzien arbeiten (Handschuhe, Kittel, Augen-Gesichtsschutz). Das Reagenz 1 enthält Eosin Y und das Reagenz 2 enthält Nigrosin.
