Arbeitsanleitung peqgold M-MuLV H plus Reverse Transcriptase PRODUKTBESCHREIBUNG peqgold M-MuLV H Plus Reverse Transkriptase (RT) ist eine genetisch modifizierte M-MuLV RT. Sie unterscheidet sich von der Wildtyp M-MuLV RT hinsichtlich der Struktur, katalytischen Eigenschaften und der Temperatur der höchsten Aktivität. Das Enzym besitzt eine RNA- und DNA-abhängige Polymeraseaktivität und eine RNase H- Aktivität, die spezifisch ist für RNA in RNA/DNA-Hybriden und die deutlich geringer ist als die der Reversen Transkriptase des Avian Myeloblastis Virus (AMV) (1,2). Die peqgold M-MuLV H Plus Reverse Transkriptase besitzt die höchste Aktivität bei 42 C (aktiv bis 50 C). Das Enzym kann Erst-Strang cdna bis zu 13 kb synthetisieren. Das Enzym kann zum Einbau modifizierter Nukleotide verwendet werden. Anwendungen Erst-Strang cdna-synthese für RT-PCR und quantitative Real-Time PCR, siehe Protokoll auf Seite 4. cdna-synthese zur Klonierung und Expression. Arbeitsanleitung peqgold M-MuLV H plus Reverse Transcriptase Definition der Unit-Aktivität Eine Unit Enzym baut in 10 min bei 37 C 1 nmol dtmp in ein Polynukleotid ein (auf DE-81). Die Enzymaktivität wurde mit folgendem Ansatz ermittelt: 50 mm Tris-HCl (ph 8.3), 4 mm MgCl 2, 10 mm DTT, 50 mm KCl, 0.5 mm dttp, 0.4 MBq/ml [ 3 H]-dTTP, 0.4 mm polya oligo (dt). Lagerungspuffer Das Enzym wird in einem Lagerungspuffer geliefert: 50 mm Tris-HCl (ph 7.5), 0.1 M NaCl, 1 mm EDTA, 5 mm DTT, 0.1 % (v/v) Triton X 100 and 50 % (v/v) Glycerin. 5x Reaktionspuffer 250 mm Tris-HCl (ph 8.3 at 25 C), 250 mm KCl, 20 mm MgCl 2, 50 mm DTT. Inhibierung und Inaktivierung Inhibitoren: Metall-Chelatbildner, anorganische Phosphate, Pyrophosphate und Polyamine (2). Hitzeinaktivierung bei 70 C für 10 min. Hinweis: peqgold M-MuLV H Plus Reverse Transkriptase weist eine viel schwächere RNase H Aktivität auf als die Reverse Transkriptase des Avian Myeloblasosis Virus (AMV). QUALITÄTSPRÜFUNGEN Endodeoxyribonuklease Assay Bei einer Inkubation von 2.000 Units peqgold M-MuLV H Plus Reverse Transkriptase mit 1 μg puc19 DNA für 4 Stunden bei 37 C, konnte kein Abbau zirkulärer DNA zu zerschnittener DNA nachgewiesen werden. Ribonuklease-Assay Bei einer Inkubation von 2000 Units peqgold M-MuLV H Plus Reverse Transkriptase mit 1 μg [ 3 H]-RNA für 4 Stunden bei 37 C konnte keine RNase Aktivität detektiert werden. Assay mit markierten Oligonukleotiden Bei einer Inkubation von 400 Units peqgold M-MuLV H Plus Reverse Transkriptase für 4 Stunden bei 37 C konnte keine Degradation markierter Oligonukleotide (Einzel- oder Doppelstrang) nachgewiesen werden. Funktioneller Assay 200 Units peqgold M-MuLV H Plus Reverse Transkriptase wurden mit 1.1 kb RNA als Template und Oligo(dT) 18 als Primer inkubiert. Das Reaktionsprodukt wurde anschließend auf einem 1.4 %-igen, basischen Agarosegel analysiert. Nach Färbung mit Ethidiumbromid konnte eine 1.1 kb große cdna-bande als vorherrschend nachgewiesen werden. PEQLAB_v0314_D 3
Arbeitsanleitung peqgold M-MuLV H plus Reverse Transcriptase PROTOKOLL ZUR ERST-STRANG CDNA-SYNTHESE Das folgende Protokoll wurde für die Erst-Strang cdna-synthese in einer Zwei-Schritt RT-PCR optimiert. Mischen und zentrifugieren Sie kurz alle verwendeten Komponenten nach dem Auftauen und halten Sie diese auf Eis. 1. Fügen Sie folgende Komponenten in der angegebenen Reihenfolge in ein steriles, nukleasefreies Reaktionsgefäß: Template RNA Primer Total RNA oder poly(a) RNA oder spezifische RNA Oligo(dT) 18 oder Random Hexamer oder genspezifisch 0.1 ng - 5 μg 10 pg - 500 ng 0.01 pg - 0.5 μg 0.5 μg (100 pmol) 0.2 μg (100 pmol) 15-20 pmol DEPC-behandeltes Wasser auf 12.5 μl auffüllen 2. Optional: Falls Ihr RNA-Template sehr GC-reich ist oder Sekundärstrukturen enthält, mischen Sie es vorsichtig, zentrifugieren Sie kurz und inkubieren Sie die Probe für 5 min bei 65 C. Kühlen Sie die Probe aus Eis ab, kurz zentrifugieren und auf Eis halten. 3. Fügen Sie die folgenden Komponenten in der angegebenen Reihenfolge zu: 5x Reaktionspuffer RNase-Inhibitor dntp Mix, jeweils 10 mm 4 μl 20 u (Details nach Angaben des Herstellers) 2 μl (1 mm finale Konzentration) peqgold M-MuLV H Plus Reverse Transkriptase 1 μl (200 u) Endvolumen 20 μl Vorsichtig mischen und kurz zentrifugieren. 4. Bei Verwendung von oligo(dt) 18 oder genspezifischen Primern, den Ansatz für 60 min bei 42 C inkubieren. Bei Verwendung von Hexamer-Primern den Ansatz erst 10 min bei 25 C, anschließend für 60 min bei 42 C inkubieren. Die Reaktionstemperatur kann bis auf 45 C erhöht werden, wenn GC-reiche RNA-Templates umgeschrieben werden sollen. 5. Die Reaktion wird durch Hitzeinaktivierung für 10 min bei 70 C gestoppt. Das Enzym sollte nicht vor der Analyse langer cdna hitzeinaktiviert werden, um eine Spaltung zu vermeiden. Hinweis: Das Produkt der Reversen Tranksription kann direkt in einer PCR eingesetzt oder bei - 20 C gelagert werden. Für eine PCR mit einem Reaktionsvolumen von 50 μl sollten 2 μl Reaktionsansatz eingesetzt werden. PEQLAB_v0314_D 4

Instruction Manual peqgold M-MuLV H plus Reverse Transcriptase DESCRIPTION peqgold M-MuLV H Plus Reverse Transcriptase (RT) is a genetically modified M-MuLV RT. It differs from wildtype M-MuLV RT by its structure, catalytic properties and in the optimum activity temperature. The enzyme possesses RNA-dependent and DNA-dependent polymerase activity and a RNase H activity specific to RNA in RNA-DNA hybrids which is significantly lower than that of Avian Myeloblastis Virus (AMV) reverse transcriptase (1, 2). peqgold M-MuLV H Plus Reverse Transcriptase activity is optimal at 42 C (active up to 50 C). The enzyme is capable of first strand cdna synthesis up to 13 kb. The enzyme incorporates modified nucleotides. Instruction Manual peqgold M-MuLV H plus Reverse Transcriptase Definition of Activity Unit One unit of the enzyme incorporates 1 nmol of dtmp into a polynucleotide fraction (adsorbed on DE-81) in 10 min at 37 C. Enzyme activity is assayed in the following mixture: 50 mm Tris-HCl (ph 8.3), 4 mm MgCl 2, 10 mm DTT, 50 mm KCl, 0.5 mm dttp, 0.4 MBq/ml [ 3 H]-dTTP, 0.4 mm polya oligo (dt). Storage Buffer The enzyme is supplied in: 50 mm Tris-HCl (ph 7.5), 0.1 M NaCl, 1 mm EDTA, 5 mm DTT, 0.1 % (v/v) Triton X 100 and 50 % (v/v) glycerol. 5x Reaction Buffer 250 mm Tris-HCl (ph 8.3 at 25 C), 250 mm KCl, 20 mm MgCl 2, 50 mm DTT. Inhibition and Inactivation Inhibitors: metal chelators, inorganic phosphate, pyrophosphate and polyamines (2) Inactivated by heating at 70 C for 10 min. Note peqgold M-MuLV H Plus Reverse Transcriptase has much lower RNase H activity than Avian Myeloblastosis Virus (AMV) reverse transcriptase. QUALITY CONTROL ASSAY DATA Endodeoxyribonuclease Assay No conversion of covalently closed circular DNA to nicked DNA was detected after incubation of 2000 units of peqgold M-MuLV H Plus Reverse Transcriptase with 1 μg of puc19 DNA for 4 hours at 37 C. Ribonuclease Assay No contaminating RNase activity was detected after incubation of 2.000 units of peqgold M-MuLV H Plus Reverse Transcriptase with 1 μg of [ 3 H]-RNA for 4 hours at 37 C. Labeled Oligonucleotide (LO) Assay No degradation of single-stranded and double-stranded labeled oligonucleotide was observed after incubation with 400 units of peqgold M-MuLV H Plus Reverse Transcriptase for 4 hours at 37 C. Functional Assay 200 units of peqgold M-MuLV H Plus Reverse Transcriptase were incubated with 1.1 kb RNA as a template and oligo (dt) 18 as a primer. The reaction product was analyzed on 1.4 % alkaline agarose gel. After ethidium bromide staining a 1.1 kb cdna band was visualized as a predominant band. PEQLAB_v0314_E 9
Instruction Manual peqgold M-MuLV H plus Reverse Transcriptase PROTOCOL FOR FIRST STRAND CDNA SYNTHESIS The following protocol is optimized to generate first-strand cdna for use in two-step RT-PCR. Mix and briefly centrifuge all components after thawing, keep on ice. 1. Add into sterile, nuclease-free tube on ice in the indicated order: Template RNA Primer DEPC-treated water total RNA or poly(a) RNA or specific RNA Oligo(dT) 18 or Random hexamer or gene-specific primer 0.1 ng-5 μg 10 pg-500 ng 0.01 pg-0.5 μg 0.5 μg (100 pmol) 0.2 μg (100 pmol) 15-20 pmol to 12.5 μl 2. Optional: If RNA template is GC rich or is known to contain secondary structures, mix gently, centrifuge briefly and incubate at 65 C for 5 min, chill on ice, briefly centrifuge and place on ice. 3. Add the following components in the indicated order: 5x Reaction Buffer RNase Inhibitor dntp Mix, 10 mm each 4 μl 20 u (see manufacturer's manual for details) 2 μl (1 mm final concentration) peqgold M-MuLV H Plus Reverse Transcriptase 1 μl (200 u) Total volume 20 μl Mix gently and centrifuge briefly. 4. If oligo(dt) 18 primer or gene-specific primer is used, incubate 60 min at 42 C. If random hexamer primer is used, incubate 10 min at 25 C followed by 60 min at 42 C. For transcription of GC rich RNA reaction temperature can be increased to 45 C. 5. Terminate the reaction by heating at 70 C for 10 min. Do not heat-inactivate enzyme prior to analysis of long cdna to avoid cleavage. Note The reverse transcription reaction product can be directly used in PCR or stored at 20 C.
