Das PCR-Produkt wird auf einem herkömmlichen Agarosegel ausgewertet. Die ready-to-load Reagenzien erlauben eine direkte Auftragung auf das Gel ohne Zugabe von Gel-Beladungspuffer. Falsch-negative Ergebnisse werden durch eine interne Amplifikationskontrolle minimiert. Der Arbeitsaufwand im Labor reduziert sich damit auf ein Minimum. Mit Ausnahme der Taq DNA Polymerase enthält das Testkit alle benötigten Reagenzien. Nur für Forschungszwecke geeignet // Stand 05/2018 Pathogener Erreger Das Testkit wurde zum Direktnachweis von Mycoplasma gallisepticum aus Trachealtupfern entwickelt. M. gallisepticum ist der Erreger der so genannten CRD (Chronic Respiratory Disease), einer chronischen Krankheit der Atemwege des Huhnes, bei der Trachitis, Luftsackentzündung, Pneumonie und Serositis im Vordergrund stehen. Es handelt sich um eine multifaktoriell bedingte Infektion mit häufiger Beteiligung von z. B. Newcastle Disease, infektiöser Bronchitis und infektiöser Laryngotrachitis. Bei Puten äußert sich die Infektion hauptsächlich als Sinusitis. M. gallisepticum wird aerogen oder durch die Bruteier übertragen. Das BACTOTYPE PCR Amplification Kit Mycoplasma gallisepticum ersetzt sowohl die serologische Diagnostik als auch die kulturelle Anzucht und liefert sensitive und spezifische Ergebnisse bereits nach einem Arbeitstag

Bei vorschriftsmäßiger Lagerung ist das Testkit mindestens 12 Monate haltbar. Wiederholtes Auftauen und Einfrieren sollte vermieden werden. Zusätzlich benötigte Reagenzien - Taq DNA Polymerase BACTOTYPE Testkits werden mit der JumpStart Taq DNA Polymerase (Sigma-Aldrich, Art. Nr. D4184) validiert. Nur bei Verwendung dieser Polymerase garantieren wir eine optimale Funktion der BACTOTYPE Produkte. Für Bestellungen oder weitere Informationen zu der o.g. 6 6 Hinweise Die Einhaltung dieses Protokolls ist unbedingt erforderlich! Um Kontaminationen jeglicher Art auszuschließen, empfehlen wir alle Pipettiervorgänge mit gestopften Pipettenspitzen durchzuführen. DNA Probenvorbereitung, Amplifikation und Elektrophorese sollten in getrennten Räumlichkeiten stattfinden. Beachten Sie die Sicherheitsbestimmungen für das Arbeiten in Laboratorien und tragen Sie Handschuhe. Der PCR Mix und die Positivkontrolle enthalten Natriumazid (NaN 3 ). Natriumazid ist ein Gefahrstoff: Sehr giftig beim Verschlucken, entwickelt bei Berührung mit Säure sehr giftige Gase (R /53, S1/ ). Ebenso ist Ethidiumbromid ein Gefahrstoff. Bitte lesen Sie das entsprechende Sicherheitsdatenblatt und tragen Sie Nitrilhandschuhe. Für BACTOTYPE Komponenten sind die Sicherheitsdatenblätter bei der Biotype Diagnostic GmbH erhältlich. Für Sicherheitsdatenblätter von Reagenzien, die nicht im Testkit enthalten sind, kontaktieren Sie bitte den jeweiligen Hersteller. DNA Isolation Eine erfolgreiche Amplifikation setzt die Verfügbarkeit von hochreiner DNA voraus. Für die DNA Isolierung von Mycoplasma gallisepticum aus Trachealtupfern werden folgende Kitsysteme empfohlen: - NucleoSpin Tissue Kit; Macherey-Nagel, Düren - QIAamp DNA Mini Kit; QIAGEN, Hilden Bitte folgen Sie dem entsprechenden Protokoll des Herstellers.

7 7 1.2 Protokoll Vorbereitung der Reagenzien: Vor Verwendung der Reagenzien empfehlen wir, alle Röhrchen zu vortexen und ca. 5 s bei 3000 U/min zu zentrifugieren. Die blaue Farbe in der Magnesiumchlorid-Lösung ist durch den Gel-Beladungspuffer bedingt. Das PCR-Ergebnis wird hierdurch nicht beeinflusst. Vermeiden Sie zu intensives Zentrifugieren der Magnesiumchlorid- Lösung, das zu einer Aufkonzentrierung des Gel-Beladungspuffers im unteren Bereich des Röhrchens führen kann. A) PCR Protokoll bei einer Probenanzahl ungleich der Verpackungseinheit (Multiplikator) Sollte die Probenanzahl kleiner als die der Verpackungseinheit des Testkits sein, so pipettieren Sie wie folgt in ein PCR-Gefäß (siehe auch Pipettierschema Tabelle 1): 17.0 µl PCR Mix µl Magnesiumchlorid µl Nuklease-freies Wasser + 0,4 µl Taq DNA Polymerase (hier: JumpStart 2.5 U/ µl) - Pipettieren Sie von diesem Master-Mix 22.5 µl in ein PCR-Gefäß und fügen 2.5 µl Ihrer DNA- Probe bzw. der Kontrolle hinzu. - Hinweis: Das Reaktionsvolumen sollte immer in Abhängigkeit der verwendeten Taq Polymerase mit Nukleasefreiem Wasser auf 25 µl aufgefüllt werden. B) PCR Protokoll bei einer Probenanzahl entsprechend der Verpackungseinheit Sollte Ihre Probenanzahl der Verpackungseinheit des Testkist entsprechen, empfehlen wir die direkte Zugabe von Magnesiumchlorid, Nuklease-freiem Wasser und Taq DNA Polymerase zum PCR Mix. Für 10 Proben (entsprechend der Verpackungseinheit von 10 Reaktionen) PCR Mix µl Magnesiumchlorid µl Nuklease-freies Wasser µl Taq DNA Polymerase (hier: JumpStart 2.5 U/µL)

8 8 Für 50 Proben (entsprechend der Verpackungseinheit von 50 Reaktionen) PCR Mix µl Magnesiumchlorid + 33 µl Nuklease-freies Wasser + 22 µl Taq DNA Polymerase (hier: JumpStart 2.5 U/µL) - Pipettieren Sie von diesem Master-Mix 22.5 µl in ein PCR-Gefäß und fügen 2.5 µl Ihrer DNA- Probe bzw. der Kontrolle hinzu. Positivkontrolle Im Testkit ist eine gebrauchsfertige Positivkontrolle (rote Verschlusskappe) enthalten, die neben den DNA Proben stets mitgeführt werden sollte. Hierbei handelt es sich um nicht-infektiöse DNA mit erregerspezifischen Sequenzen. Negativkontrolle Neben den DNA Proben sollte stets eine Negativkontrolle (z. B. Nuklease-freies Wasser) mitgeführt werden. Die Menge der einzusetzenden DNA richtet sich nach ihrer Konzentration und kann zwischen 1-3 μl variiert werden. Die Menge an Nuklease-freiem Wasser ist entsprechend zu korrigieren, so dass das Gesamtvolumen des PCR-Ansatzes immer 25 μl beträgt.

9 9 PCR Amplifikationsparameter Wir empfehlen unbedingt die Verwendung der Sigma JumpStart Taq DNA Polymerase. Temperatur Zeit 94 C 3 min Hot Start * 94 C 30 s 60 C 30 s (RAMPING 1 C/s ** ) 35 Zyklen 72 C 30 s 72 C 5 min 10 C Bis zum Ende * Bei der Verwendung von hot start -Polymerasen sollten die vom Hersteller vorgegebenen Aktivierungszeiten eingehalten werden (für JumpStart Taq DNA Polymerase: 3 min). ** Dieser Wert ist bei den Thermocyclern gerätespezifisch einzustellen. Eine Kontrolle und ggf. Korrektur wird empfohlen Elektrophorese Die Analyse der PCR-Produkte erfolgt mit einem 2%igen Agarosegel und Anfärbung mit Ethidiumbromid ( µg/ml) in TAE-Puffer. 10 µl des PCR-Ansatzes werden pro Bahn aufgetragen. Die Zugabe von Gel-Beladungspuffer entfällt, da dieser bereits im Master-Mix enthalten ist. Zum Größenvergleich wird ein Längenstandard im niedermolekularen Bereich empfohlen (100 bp DNA Leiter). Die Elektrophorese sollte gerätespezifisch durchgeführt werden.

10 10 Auswertung und Interpretation der Ergebnisse Die Größe der Amplifikate wird durch Vergleich mit dem DNA-Längenstandard bestimmt. Eine Fotodokumentation ist für Archivierungszwecke empfehlenswert. Schützen Sie Augen und Haut vor UV-Bestrahlung! Positives Ergebnis: Ist der getestete P. multocida Stamm ein Toxinbildner ( D-Stamm ), wird neben der erregerspezifischen Bande von 415 bp bevorzugt die Bande für den Toxinnachweis (331 bp) amplifiziert. Bildet der Stamm kein Toxin, fehlt die Bande für den Toxinnachweis (331 bp) vollständig und es ist nur die erregerspezifische Bande (415 bp) sichtbar. Bei niedriger Erregerkonzentration kann es vorkommen, dass die 838 bp Bande ebenfalls sichtbar ist. Der Test ist in diesen Fällen dennoch gültig. Negatives Ergebnis: Ist nur die Bande von 838 bp (Amplifikationskontrolle) sichtbar, ist das Resultat negativ. Ungültiges Ergebnis: Sollte keine der oben genannten Banden erkennbar sein, ist der Test nicht auswertbar und muss wiederholt werden. 11 11 2 Instructions for Use 2.1 Product description Technology The product group BACTOTYPE PCR Amplification Kit comprises optimised systems for the identification of bacterial pathogens via polymerase chain reaction (PCR): Highest sensitivity and specificity: All components are carefully developed and perfectly optimised Easy and comfortable work: The reagents are ready-to-use and ready-to-load Reliable results: The test kit contains an internal amplification control The primer pair of the test kit is specific for two gene fragments of Pasteurella multocida. The PCR products are analysed on a conventional agarose gel. The ready-to-load reagents allow direct loading of the samples on the gel without addition of gel loading buffer. False negative results are minimized by using an internal amplification control. Manual work is reduced to a minimum. With exception of Taq DNA Polymerase, the test kit consists of all reagents necessary for the detection of the bacterial germs. Pathogenic Germ For research use only // 05/2018 Pasteurella multocida is part of the physiological bacterial flora of the upper respiratory tract of pigs. As secondary pathogen invading lungs injured by other bacteria or viruses P. multocida induces pneumonia. Subsets of P. multocida isolates are critical to induce the so called progressive atrophic rhinitis. The isolates synthesize a toxin encoded by the chromosomal toxa gene that serves as essential virulence factor. Non-toxigenic P. multocida isolates do not cause atrophic rhinitis. Culture, species identification, and toxin testing of P. multocida are timeconsuming and costly. 13 13 Product Warranty All contents of the BACTOTYPE PCR Amplification Kit are subjected to extensive quality procedures by Biotype Diagnostic GmbH. In case of any problem, please report immediately. Notes Please, carry out precisely the instructions of the protocol! In order to avoid any kind of contamination we recommend pipetting with filter tips. DNA preparation, amplification, and electrophoresis should be carried out in separate rooms. Take note of safety regulations for working in laboratories, and wear gloves. The PCR Mix and the Positive Control contain sodium azide (NaN 3 ). Sodium azide is hazardous: Very toxic if swallowed, develops toxic gases when it gets in contact with acids (R /53, S1/ ). Ethidium bromide is hazardous, too. Please refer to the Material Safety Data Sheet (MSDS) and wear nitrile gloves. MSDS of all BACTOTYPE components are available from Labor Diagnostic Leipzig. For MSDS of additional reagents to be needed, please contact the corresponding manufactures. DNA Isolation Solely, ultrapure DNA implies the successful amplification. For DNA isolation of Pasteurella multocida from tissue samples or swabs the following test systems are recommended: - NucleoSpin Tissue Kit; Macherey-Nagel, Düren - QIAamp DNA Mini Kit; QIAGEN, Hilden Please perform isolation according to the manufacturer s information.

14 Protocol Preparation of the reagents: Before using the reagents of the test kit the first time, mix thoroughly and centrifuge the reagents for 5 sec at 3000 rpm. The blue colour in the magnesium chloride solution arises from the gel loading buffer and does not affect the PCR reaction. Please, avoid elongated centrifugation steps of the magnesium chloride solution. The gel loading buffer could be concentrated onto the bottom of the tube. A) PCR Protocol for a Number of Samples different from the Packaging Unit (Multiplicator) If there are less samples than the packaging unit, pipette as followed into a PCR reaction tube (also see pipetting scheme table 1): 17.0 µl PCR Mix µl Magnesium Chloride µl Nuclease-free Water µl Taq DNA Polymerase (here: JumpStart 2.5 U/µL) - Pipet 22.5 µl of the master mix into a PCR reaction tube and add 2.5 µl of your DNA sample or control. - Attention: Depending on the used Taq Polymerase, complete with Nuclease-free Water to a total volume of 25 µl per reaction. B) PCR protocol for an Amount of Samples corresponding to Packaging Unit If the number of samples corresponds to the packaging unit, we recommend the addition of Magnesium Chloride, Nuclease-free Water, and Taq DNA Polymerase directly to the PCR Mix. For 10 samples (corresponding to the content of 10 reactions) PCR Mix µl Magnesium Chloride µl Nuclease-free Water µl Taq DNA Polymerase (here: JumpStart 2.5 U/µL)

15 15 For 50 samples (corresponding to the content of 50 reactions) PCR Mix µl Magnesium Chloride + 33 µl Nuclease-free Water + 22 µl Taq DNA Polymerase (here: JumpStart 2.5 U/µL) - Pipet 22.5 µl of the master mix into a PCR reaction tube and add 2.5 µl of your DNA sample or control. Positive Control The test kit is provided with a ready-to-use Positive Control (red cap) of non-infectious DNA with germ-specific sequences. We strongly recommend performing a positive control for each run. Negative Control It is strongly recommended to perform a negative control (e. g. Nuclease-free Water) for each run. The volume of the DNA to be employed depends on its concentration and may be varied from 1 to 3 μl. Adjust the volume with Nuclease-free Water to total reaction volume of 25 μl.

16 16 PCR Amplification Parameters We strongly recommend the use of Sigma JumpStart Taq DNA Polymerase. Temperature Time 94 C 3 min Hot Start * 94 C 30 s 60 C RAMPING 1 C/s ** 30 s 35 Cycles 72 C 30 s 72 C 5 min 10 C hold * For hot start Polymerase, please follow the activation time conditions specified by the manufacturer (for JumpStart Taq DNA Polymerase: 3 minutes). ** The value is adjusted for most of the thermocyclers. Cyclers that exceed the ramping rate should be monitored and corrected as required. Electrophoresis PCR fragments are analysed on a 2% agarose gel and staining is done with ethidium bromide ( µg/ml) in TAE buffer. Put 10 µl of the PCR product per lane onto the gel (gel loading buffer already included). In order to compare the molecular size of the amplification products a size standard (100 bp DNA ladder) is recommended. Run the electrophoresis as described in the manufacturer s handbook.

17 17 Analysis and Interpretation of the Results Size designation of the PCR fragments is done by comparison with the DNA size standard on a transilluminator. Photographic documentation is recommended. Protect eyes and skin against UV-radiation! Positive result:p. multocida strains encoding toxa show a toxin-specific band at 331 bp in addition to the germ-specific band at 415 bp. If the strain does not encode ToxA, only the germ-specific band at 415 bp can be detected. The 838 bp (amplification control) band may be weak or may not appear at all due to low concentrations of bacterial DNA, whereas the germspecific band is clearly visible at all. In this case the test is valid. The result is positive. Negative result: If the 838 bp band (amplification control) appears only, germ detection is considered to be negative. Invalid result: If none of the bands mentioned above are detectable, the assay is invalid and has to be repeated.
