Summary The aim of this study was the biochemical characterization of chondroitin sulfate proteoglycans from the peripheral nervous system and the investigation of their functional implications in cell-toextracellular matrix interactions in view of their possible role in regenerative processes in peripheral nerves. During regeneration of mouse sciatic nerve an upregulation of a chondroitin sulfate epitope, defined by reactivity with monoclonal antibody 473 (Faissner, 1988) against a mouse brain proteoglycan, was observed. To characterize the molecules carrying the epitope, human sciatic nerve was investigated to contain three chondroitin sulfate proteoglycans as a model nerve and shown with molecular weights of 2300 kd, 900 kd and 130 kd. The 900 kd proteoglycan and to a lower extend the 130 kd proteoglycan carried the mab 473 epitope. The epitope itself was found to be located on chondroitin-6- sulfate chains, the glycosaminoglycan part of the molecules. The 900 kd proteoglycan could be identified as a versican-like molecule and the 130 kd proteoglycan as a decorin-like molecule by reactivity with two polyclonal sera against the well characterized chondroitin sulfate proteoglycans versican and decorin from human fibroblasts. In the regenerating sulfate chains of mouse sciatic nerve the number of chondroitin the versican-like proteoglycan was increased, whereas the core protein showed no change in expression. In contrast, the core protein of the decorin-like molecule was upregulated in the regenerating nerve. The two molecules from mouse sciatic nerve were identified as versican- and decorin-like molecules by reactivity with a polyclonal serum against the mab 473 immunoreactive versican- and decorin-like molecules from human sciatic nerve. These results indicated that both chondroitin sulfate proteoglycans may be involved in regenerative processes. Since the mab 473 epitope was found to be localized in Schwann cell basal laminae of adult mouse sciatic nerve, the binding of the versican-like molecule, as the main mab 473 immunoreactive molecule, to typical basal laminae components was investigated. A

3 2 saturable binding of the versican-like molecule to fibronectin, but not to laminin or collagen types I and IV was detected. The already described binding of the decorin-like molecule to fibronectin was confirmed. The possible role of the two chondroitin sulfate proteoglycans in regenerative processes could be elucidated by the use of cell culture test systems. Cell adhesion of several cell lines (fibroblasts, Schwann cells and neuronal cells) and of neonatal dorsal root ganglion neurons and Schwann cells to substrate-coated mixtures of fibronectin with the versican- or decorin-like molecules was reduced with increasing proteoglycan concentrations. In contrast, cell adhesion to substrate-coated mixtures of laminin or collagen types I and IV with the proteoglycans was not affected. Cell adhesion could be restored by enzymatic removal of the chondroitin sulfate from the substrate, indicating that the chondroitin sulfate is the inhibitory component for cell adhesion. Neurite outgrowth of the pheochromocytoma cells PC 12 or neonatal dorsal root ganglion neurons on substrate-coated versican- or decorin-like molecules on substrate-coated mixtures of fibronectin with the versican- or decorin-like molecules was neither positively nor negatively affected in comparison to control substrates. However, at high proteoglycan concentrations a slight reduction of neurite outgrowth was observed. Furthermore, the expression of versican- and decorin-like molecules released in the supernatant of rat transformed Schwann cells was shown to be upregulated, respectively downregulated by the action of the growth factors TGF-6 and NGF. In summary, the versican- and decorin-like molecules, which expression is regulated during regeneration of mouse sciatic nerve probably by action of growth factors, may contribute to regenerative events by influencing cell-to-extracellular matrix-dependent processes, such as cell proliferation or migration rather than by altering growth behaviour of regenerating axons.

Zusammenfassung Das Ziel dieser Arbeit war die biochemische Charakterisierung von Chondroitinsulfatproteoglykanen des peripheren Nervensystems und die Untersuchung ihrer funktionellen Eigenschaften bei Interaktionen von Zellen mit der Extrazelluldrmatrix unter besonderer Berucksichtigung ihrer mcglichen Rolle bei regenerativen VorgSngen in peripheren Nerven. wahrend der Regeneration des Ischiasnerven der Maus konnte eine erhohte Expression eines Chondroitinsulfatepitopes beobachtet werden, das durch den monoklonalen AntikOrper 473 (Faissner, 1988) gegen ein Proteoglykan aus Maushirn definiert wird. Urn die, das Epitop tragenden, Molekule zu identifizieren, wurde humaner Ischiasnerv als Modellnerv benutzt. Aus diesem Gewebe konnten drei Chondroitinsulfatproteoglykane mit Molekulargewichten von 2300 kd, 900 kd und 130 kd isoliert werden. Das 900 kd Proteoglykan und in geringerem AusmaB das 130 kd Proteoglycan tragen das 473-Epitop. Das Epitop konnte auf Chondroitin-6-sulfat-ketten, den Glykosaminoglykanen der Proteoglykane, lokalisiert werden. Mit Hilfe zweier polyklonaler Seren gegen bereits bekannte Proteoglykane konnte das 900 kd Proteoglykan als ein Versican-flhnliches und das 130 kd Proteoglykan als ein Decorin-dhnliches Molekul identifiziert werden. Im regenerierenden Ischiasnerven der Maus konnte eine Zunahme des Chondroitinsulfat eines der beiden Proteoglykane festgestellt werden, wohingegen das Kernprotein keine Anderung der Expression zeigte. Dieses Molekul konnte mit Hilfe eines polyklonalen Serums gegen die 473 immunreaktiven Versican- und Decorin-ahnlichen Molekule aus humanem Ischiasnerven als Versican-Shnliches Molekul identifiziert werden. Ebenso konnte eine erhohte Expression des Kernproteins des Decorin-ahnlichen Molekuls gezeigt werden. Diese Ergebnisse deuten darauf hin, daft beide Proteoglykane an VorgSngen wfihrend der Regeneration beteiligt sein konnen. Da das 473 immunoreaktive Chondroitinsulfatepitop in den

5 4 Basallaminae SchwamVscher Zellen im peripheren Nerven lokalisiert werden konnte, wurden die Bindung des Versicandhnlichem MolekQIs, als 473 immunoreaktive Hauptkomponente, an typische Basallaminaemolekule untersucht. Es konnte eine sattigbare Bindung des Versican-Shnlichen Molekuls an Fibronektin, aber nicht an Laminin, Kollagen Typ I oder IV gezeigt werden. Die schon bekannte Bindung des Decorin-Shnlichen Molekuls an Fibronektin konnte bestatigt werden. Die mogliche Rolle der beiden Proteoglykane bei der Regeneration peripherer Nerven konnte mit Hilfe von Zellkultur-Testsystemen aufgekiart werden. Die ZelladhSsion verschiedener Zellinien (Fibroblasten, Schwann'sche Zellen und neuronale Zellen) und von Neuronen oder von Schwann*schen Zellen aus neonatalen Hinterwurzelganglien auf substratgebundene Mischungen von Fibronektin mit dem Versican-ahnlichen oder Decorin-ahnlichen Molekul wurde mit steigenden Proteoglykankonzentrationen reduziert. Der inhibitorische Effekt konnte jedoch nicht auf substratgebundenen Mischungen von Laminin oder Kollagen Typ I oder IV mit diesen Proteoglykanen beobachtet werden. Durch enzymatische Entfernung des Chondroitinsulfat konnte der Effekt wieder ruckgsngig gemacht werden. Dies zeigte, daft das Chondroitinsulfat die wesentliche inhibitorische Komponente fur die zelladhasion ist. Das Neuritenwachstum von Pheochromocytomazellen PC 12 oder von neonatalen Neuronen aus Hinterwurzelganglien auf substratengebundenen Versican- oder Decorin-Shnlichen Molekulen oder auf substratgebundenen Mischungen von Fibronektin mit den Versican- und Decorin-dhnlichen Molekulen wurde weder positiv noch negativ beeinfluftt, im Vergleich zu Kontrollsubstraten. Bei hohen Proteoglykankonzentrationen konnte jedoch ein reduziertes Neuritenauswachstum beobachtet werden. Weiterhin konnte gezeigt werden, daft die Expression der Versicanund Decorin-ahnlichen Molekule in Kulturuberst&nden von transformierten Schwann Zellen der Ratte durch die Wachstumsfaktoren TGF-ft und NGF hoch- beziehungsweise

6 5 herunterreguliert wird. Zusammenfassend betrachtet scheinen die Versican- und Decorin- Shnlichen Molekule, deren Expression wdhrend der Regeneration von Ischiasnerven der Maus moglicherweise durch Wachstumsfaktoren reguliert wird, eher an Zell-Extrazellul&rmatrix vermittelten Prozessen wie Zellproliferation und -wanderung als am Neuritenwachstum wdhrend der Regeneration beteiligt zu sein.
