3 Kurzinformation RIDASCREEN FAST Vitamin B12 (Art. Nr. R2103) ist ein kompetitiver Enzymimmunoassay zur quantitativen Bestimmung von Vitamin B12 in Milch, Milchpulver, Lebensmitteln für besondere medizinische Zwecke*, Müsli und Getreideflocken, angereicherten Getreidemehlen, Vitaminpulver, -mischungen, -tabletten und Vitaminsäften. Alle Reagenzien für die Durchführung des Enzymimmunoassays - inkl. Standards - sind im Testkit enthalten. Ein Testkit ist ausreichend für 48 Bestimmungen (einschl. Standardbestimmungen). Zur Auswertung benötigt man ein Mikrotiterplatten-Photometer. In Proben kann die Spezifität aufgrund von Matrixeffekten von den im Puffersystem ermittelten Ergebnissen abweichen. Vor der Analyse von kreuzreaktiven Substanzen muss deren Nachweisgrenze und Wiederfindungsrate in der jeweiligen Matrix durch den Anwender bestimmt werden. Der Test kann nicht zwischen Analyten und kreuzreaktiven Substanzen diskriminieren. 4 Zur Erhöhung der Prüfungsqualität bei ELISA-Verfahren verweisen wir zusätzlich auf unser ELISA-Handbuch in der jeweiligen Fassung. Dieses führt Mindeststandards hinsichtlich der Rahmenbedingungen auf, die bei der Verwendung von Testsystemen der R-Biopharm AG und der Durchführung von ELISA-Analysen mit diesen Testsystemen zu beachten sind. Das Handbuch kann unter der Webseite abgerufen, gedruckt und gespeichert werden. 1. Verwendungszweck RIDASCREEN FAST Vitamin B12 ist ein kompetitiver Enzymimmunoassay zur quantitativen Bestimmung von Vitamin B12 in Milch, Milchpulver, Lebensmitteln für besondere medizinische Zwecke, Müsli und Getreideflocken, angereicherten Getreidemehlen, Vitaminpulver, -mischungen, -tabletten und Vitaminsäften. 2. Allgemeines Vitamin B12 gehört zur Gruppe der wasserlöslichen Vitamine. Es kann vom menschlichen Körper selbst nicht hergestellt werden und muss deshalb über die Nahrung aufgenommen werden. Vitamin B12 wird von unserem Körper nur in geringen Mengen benötigt, dennoch spielt es eine wichtige Rolle für das Nervensystem, hat eine Schutzwirkung für das Herz-Kreislauf-System und spielt eine bedeutende Rolle in der Nukleotid-Biosynthese, der Eisenaufnahme und dem Aminosäurestoffwechsel. Ein Mangel an Vitamin B12 kann zu perniziöser Anämie und neurologischen Symptomen führen. 3. Testprinzip Grundlage ist eine Antigen-Antikörper-Reaktion. Die Kavitäten der Mikrotiterplatte sind mit spezifischen Antikörpern gegen Vitamin B12 beschichtet. Bei Zugabe von Standard bzw. Probe und Konjugat konkurrieren freies und enzymmarkiertes Vitamin B12 um die Vitamin B12-Antikörperbindungsstellen (kompetitiver Enzymimmunoassay). Nicht gebundenes enzymmarkiertes Vitamin B12 wird anschließend in einem Waschschritt wieder entfernt. Der Nachweis erfolgt durch Zugabe von Substrat/Chromogen. Gebundenes Konjugat wandelt das Chromogen in ein blaues Endprodukt um. Die Zugabe der Stopp Lösung führt zu einem Farbumschlag von blau nach gelb. Die Messung erfolgt photometrisch bei 450 nm; die Extinktion der Lösung ist umgekehrt proportional zur Vitamin B12- Konzentration in der Probe. 4 RIDASCREEN FAST Vitamin B

5 4. Packungsinhalt Mit den Reagenzien einer Packung können 48 Bestimmungen durchgeführt werden (einschließlich Standardbestimmungen). 6 5.2. Reagenzien: destilliertes oder demineralisiertes Wasser 6. Vorsichtsmaßnahmen Dieser Test ist nur von geschultem Laborpersonal durchzuführen. Die Gebrauchsanweisung zur Durchführung des Tests ist strikt einzuhalten. Dieser Kit kann gesundheitsgefährdende Substanzen enthalten. Sicherheitshinweise zu den enthaltenen Komponenten entnehmen Sie bitte den Sicherheitsdatenblättern (MSDS) zu diesem Produkt auf unserer Internetseite 7. Reagenzien und ihre Lagerung Die Reagenzien bei 2-8 C lagern. Komponenten des Testkits auf keinen Fall einfrieren. Nicht benötigte Kavitäten zusammen mit dem Trockenmittel im Folienbeutel gut verschlossen aufbewahren und weiterhin bei 2-8 C lagern. Die Standards, das Konjugat und das Substrat/Chromogen sind lichtempfindlich, deshalb direkte Lichteinwirkung vermeiden. Nach Ablauf des Verfallsdatums (siehe Testkit-Außenetikett) kann keine Qualitätsgarantie mehr übernommen werden. Ein Austausch von Einzelreagenzien zwischen Kits verschiedener Chargennummern ist nicht zulässig (Ausnahmen: Probenpuffer, Waschpuffer (Salz), Stopp Lösung). 8. Anzeichen für Reagenzienverfall bläuliche Färbung des rötlichen Substrat/Chromogens vor Zugabe in die Kavitäten Extinktion kleiner 0,6 (E 450 nm 0,6) für den Nullstandard 6 RIDASCREEN FAST Vitamin B

7 9. Probenvorbereitung Die Proben kühl und lichtgeschützt lagern. Die Proben sollten vor der Probenvorbereitung auf C temperiert werden. Zur Qualitätskontrolle wird empfohlen, Kontrollproben mitzuführen. Anmerkung: Die Proben sollten am Tag der Extraktion gemessen werden. Der Probenpuffer ist auch zur Probenvorbereitung von folsäurehaltigen Proben in Verbindung mit dem RIDASCREEN FAST Folsäure (Folic Acid) (R3203) verwendbar Milch, Milchpulver und Lebensmittel für besondere medizinische Zwecke Milch: die homogenisierte Milch kann unverdünnt in den Test eingesetzt werden, eventuell mit Probenpuffer in den Messbereich verdünnen (Verdünnungsfaktor entsprechend neu berechnen!) 50 µl pro Kavität im Test einsetzen Milchpulver, Lebensmittel für besondere medizinische Zwecke: 1 g Probe in ca. 5 ml dest. Wasser suspendieren, mit dest. Wasser auf 10 ml auffüllen (Verdünnungsfaktor = 10) 10 min schütteln / rühren die verdünnte Probe 3 min im Wasserbad bei 100 C erhitzen, schnell abkühlen (Eisbad), eventuell mit Probenpuffer in den Messbereich verdünnen (Verdünnungsfaktor entsprechend neu berechnen!) 50 µl pro Kavität im Test einsetzen 9.2. Müsli und Getreideflocken Eine repräsentative Probe vor dem Extrahieren zermahlen und mischen (Partikelgröße < 250 µm). 1 g der gemahlenen Probe in ein Reagenzgefäß einwiegen, in ca. 5 ml Probenpuffer suspendieren und mit Probenpuffer auf 10 ml auffüllen (Verdünnungsfaktor = 10) 10 min schütteln / rühren ungelöste Bestandteile durch Zentrifugation (alternativ: filtrieren) entfernen RIDASCREEN FAST Vitamin B

8 Überstand abnehmen, falls der Überstand noch trüb sein sollte, filtrieren Überstand oder Filtrat gegebenenfalls mit Probenpuffer in den Messbereich des Tests verdünnen (Verdünnungsfaktor entsprechend neu berechnen!) 50 µl pro Kavität im Test einsetzen 9.3. Angereicherte Getreidemehle Eine repräsentative Probe vor dem Extrahieren mischen, gegebenenfalls zerkleinern (Partikelgröße < 250 µm). 1 g der gemahlenen Probe in ein Reagenzgefäß einwiegen, in ca. 5 ml Probenpuffer suspendieren und mit Probenpuffer auf 10 ml auffüllen (Verdünnungsfaktor = 10) 10 min schütteln / rühren ungelöste Bestandteile durch Zentrifugation (alternativ: filtrieren) entfernen Überstand abnehmen, falls der Überstand noch trüb sein sollte, filtrieren Überstand oder Filtrat gegebenenfalls mit Probenpuffer in den Messbereich des Tests verdünnen (Verdünnungsfaktor entsprechend neu berechnen!) 50 µl pro Kavität im Test einsetzen 9.4. Vitaminpulver, -mischungen und -tabletten Vitaminpulver/-mischung: 1 g Vitaminpulver/-mischung in 5 ml Probenpuffer lösen und mit Probenpuffer auf 10 ml auffüllen (Verdünnungsfaktor = 10) Tabletten: Tabletten mörsern, sorgfältig mischen, davon 1 g in 5 ml Probenpuffer lösen und mit Probenpuffer auf 10 ml auffüllen (Verdünnungsfaktor = 10) Mit diesen Proben wie folgt weiterarbeiten: 10 min schütteln / rühren ungelöste Bestandteile durch Zentrifugation (alternativ: filtrieren) entfernen Überstand oder Filtrat gegebenenfalls mit Probenpuffer in den Messbereich des Tests verdünnen (Verdünnungsfaktor entsprechend neu berechnen!) 50 µl pro Kavität in den Test einsetzen 8 RIDASCREEN FAST Vitamin B

Vitaminsäfte der homogenisierte Saft kann unverdünnt in den Test eingesetzt werden gegebenenfalls ungelöste Bestandteile durch Zentrifugation (alternativ: filtrieren) entfernen eventuell mit Probenpuffer in den Messbereich verdünnen (Verdünnungsfaktor entsprechend neu berechnen!) 50 µl pro Kavität im Test einsetzen 10. Testdurchführung Testvorbereitungen Alle Reagenzien vor Gebrauch auf Raumtemperatur (20-25 C) bringen. Der Probenpuffer liegt als 2-fach Konzentrat vor. Das Konzentrat 1:2 mit destilliertem Wasser verdünnen (z.b. 50 ml Pufferkonzentrat + 50 ml dest. Wasser). Der verdünnte Probenpuffer hat eine Haltbarkeit von 4-6 Wochen bei 2-8 C. Als Waschpuffer wird ein PBS-Tween-Puffer benötigt, benutzen Sie dazu bitte den beiliegenden Waschpuffer (Salz) (siehe 4.). Zur Herstellung des Puffers den gesamten Inhalt des Beutels in 1 Liter destilliertem Wasser lösen. Der gelöste Waschpuffer ist ca. 4-6 Wochen bei 2-8 C haltbar. Alternativ: Inhalt des Beutels in 100 ml dest. Wasser lösen (10-fach Konzentrat). Die Lösung ist ca Wochen bei Raumtemperatur (20-25 C) haltbar. Um die gebrauchsfertige Lösung herzustellen 1 Teil des 10-fach Konzentrats mit 9 Teilen dest. Wasser mischen Testdurchführung Standards und Proben müssen zeitgleich auf derselben Mikrotiterplatte abgearbeitet werden. Sorgfältiges Waschen ist sehr wichtig. Ein Eintrocknen der Kavitäten zwischen den Arbeitsschritten ist zu vermeiden. 1. So viele Kavitäten in den Halterahmen einsetzen, wie für alle Standards und Proben in Doppelbestimmung benötigt werden. Die Positionen der Standards und der Proben protokollieren. 10 2. Je 50 µl des Standards bzw. der vorbereiteten Proben in die entsprechenden Kavitäten pipettieren; für jeden Standard oder Probe eine neue Pipettenspitze benutzen. 3. Je 50 µl Konjugat in die entsprechenden Kavitäten pipettieren (eventuell einen Mehrfachdispenser oder eine 8-Kanal-Pipette verwenden) und 15 min bei Raumtemperatur (20-25 C) lichtgeschützt inkubieren. 4. Die Kavitäten durch Ausschlagen der Flüssigkeit leeren und die Restflüssigkeit durch kräftiges Ausklopfen (dreimal hintereinander) auf saugfähigen Labortüchern entfernen. Die Kavitäten mit Hilfe einer 8-Kanalpipette mit gebrauchsfertigem Waschpuffer (10.1) waschen (250 µl/kavität), die Kavitäten leeren und die Restflüssigkeit entfernen. Diesen Vorgang noch zweimal wiederholen. 5. Je 100 µl Substrat/Chromogen in jede Kavität pipettieren (eventuell einen Mehrfachdispenser oder eine 8-Kanalpipette verwenden) und 10 min bei Raumtemperatur (20-25 C) im Dunkeln inkubieren. 6. Je 100 µl Stopp Lösung in jede Kavität pipettieren (eventuell einen Mehrfachdispenser oder eine 8-Kanalpipette verwenden). Die Extinktion bei 450 nm innerhalb von 5 min nach Zugabe der Stopp Lösung messen. 11. Auswertung Für die Auswertung ist bei R-Biopharm eine speziell für die RIDASCREEN Enzymimmunoassays entwickelte Software, RIDA SOFT Win / RIDA SOFT Win.net (Art. Nr. Z9996), erhältlich. Diese Software gibt das Ergebnis sowohl in µg/kg bzw. µg/l als auch in µg/100 g bzw. µg/100 ml an. Der Verlauf der Standardkurve kann dem beigefügten Analysenzertifikat entnommen werden. Hinweis für die Berechnung ohne Software: Extinktion Standard (bzw. Probe) Extinktion Nullstandard x 100 = % Extinktion Den Nullstandard somit gleich 100 % setzen und die Extinktionswerte in Prozent angeben. Die errechneten Werte für die Standards in einem Koordinatensystem auf halblogarithmischem Millimeterpapier gegen die Vitamin B12-Konzentration (µg/kg bzw. µg/l) auftragen. 10 RIDASCREEN FAST Vitamin B

11 Um die in den Proben enthaltene tatsächliche Vitamin B12-Konzentration zu erhalten, muss die aus der Standardkurve abgelesene Konzentration noch mit dem entsprechenden Verdünnungsfaktor multipliziert werden. Diese Angaben entsprechen dem heutigen Stand unserer Kenntnisse und sollen über unsere Produkte und deren Anwendungsmöglichkeiten informieren. Sie haben somit nicht die Bedeutung, bestimmte Eigenschaften der Produkte oder deren Eignung für einen konkreten Einsatzzweck zuzusichern. R-Biopharm übernimmt keine Gewährleistung, außer für die standardisierte Qualität der Reagenzien. Defekte Produkte werden ersetzt. No. R2103) is a competitive enzyme immunoassay for the quantitative determination of Vitamin B12 in milk, milk powders, food for special medical purpose*, grain and cereals, fortified flour, vitamin powders, -mixtures, -tablets, and vitamin juices. All reagents required for the enzyme immunoassay - including standards - are contained in the test kit. The test kit is sufficient for 48 determinations (including standards). A microtiter plate spectrophotometer is required for quantification. In samples, the specificity may deviate from those determined in the buffer system due to matrix effects. Prior to the analysis of cross-reactive substances, the user has to determine the Limit of Detection and the recovery for the substance in the respective sample matrix. The test cannot discriminate between analytes and cross-reactive substances. Intended use RIDASCREEN FAST Vitamin B12 is a competitive enzyme immunoassay for the quantitative determination of Vitamin B12 in milk, milk powders, food for special medical purpose, grain and cereals, fortified flour, vitamin powders, -mixtures, -tablets, and vitamin juices. 2. General Vitamin B12 belongs to group of the water soluble vitamins. It is not produced by the human body and must be included in the diet. Vitamin B12 is needed only in small concentrations, however it plays an important role for the nervous system, has protective effects for the heart and plays an important role in nucleotide biosynthesis, iron intake, and metabolism of amino acids. A lack of vitamin B12 can lead to anemia and neurological deficits. 3. Test principle The basis of the test is the antigen-antibody reaction. The microtiter plate is coated with specific antibodies against Vitamin B12. Vitamin B12 standards, respectively sample and enzyme labeled Vitamin B12 (conjugate) are added. Free and enzyme labeled Vitamin B12 compete for the antibody binding sites (competitive enzyme immunoassay). Any unbound conjugate is then removed in a washing step. Substrate/chromogen is added to the wells, bound conjugate converts the chromogen into a blue product. The addition of the stop solution leads to a color change from blue to yellow. The measurement is made photometrically at 450 nm. The resulting absorbance values are inversely proportional to the Vitamin B12 concentration of the sample. RIDASCREEN FAST Vitamin B

15 6. Warnings and precautions for the users This test should only be carried out by trained laboratory employees. The instruction for use must be strictly followed. This kit may contain hazardous substances. For hazard notes on the contained substances please refer to the appropriate material safety data sheets (MSDS) for this product, available online at 7. Storage instructions Store the kit at 2-8 C (36-46 F). Do not freeze any test kit components. Return any unused microwells to their original foil bag, reseal them together with the desiccant bag provided and further store at 2-8 C (36-46 F). The standards, the conjugate and the substrate/chromogen are light sensitive, therefore, avoid exposure to direct light. No quality guarantee is accepted after the expiration date on the kit label. Do not interchange individual reagents between kits of different lot numbers. (exception: sample buffer, wash buffer (salt), stop solution) 8. Indication of instability or deterioration of reagents any bluish coloration of the red stained substrate/chromogen prior to test implementation an absorption smaller than 0.6 (A 450 nm < 0.6) for the zero standard 9. Preparation of samples The samples should be stored in a cool place, protected from light. The samples should be brought to room temperature before analysis (20-25 C / F). For quality control it is recommended to analyse control samples. Remark: The samples should be measured at the day of extraction. The sample buffer can also be used for the sample preparation of folic acid containing samples in combination with the RIDASCREEN FAST Folsäure (Folic Acid) (R3203). RIDASCREEN FAST Vitamin B

16 9.1. Milk, milk powder and food for special medical purpose Milk: homogenized milk can be directly applied in the test. Test implementation Test preparation Bring all reagents to room temperature (20-25 C / F) before use. The sample buffer is provided as a 2-fold concentrate. The concentrate is diluted 1:2 with distilled water (e.g. 50 ml sample buffer concentrate + 50 ml distilled water). The diluted sample buffer expires after approx. 4-6 weeks at 2-8 C (36-46 F). A PBS-Tween buffer is required as wash buffer, please use the wash buffer salt (pouch) contained in the kit. Dissolve the total content of the pouch in one liter of RIDASCREEN FAST Vitamin B

18 distilled water. The ready to use wash buffer expires after approx. 4-6 weeks at 2-8 C (36 46 F). Alternatively: Dissolve the content of the pouch in 100 ml of distilled water to obtain a 10-fold concentrated wash buffer. This solution expires after approx weeks, stored at room temperature (20-25 C / F). Use one part of this concentrate and dissolve with 9 parts of distilled water to obtain the ready to use washing buffer Test procedure Standards and samples must be tested on one microtiter plate at one time. Carefully follow the recommended washing procedure. Do not allow microwells to dry between working steps. 1. Insert a sufficient number of wells into the microwell holder for all standards and samples to be run in duplicate. Record standard and sample positions. 2. Add 50 µl of each standard or prepared sample to the wells in duplicate; use a new pipette tip for each standard or sample. 3. Add 50 µl of conjugate to each well (multichannel pipette or 8 channel pipette) and incubate for 15 min at room temperature (20-25 C / F) in the dark. 4. Pour the liquid out of the wells and tap the microwell holder upside down vigorously (three times in a row) against absorbent paper to ensure complete removal of liquid from the wells. Using a 8 channel pipette, fill the wells with ready to use wash buffer (250 µl/well) (10.1). Empty the wells again and remove all remaining liquid. Repeat the washing step two more times. 5. Add 100 µl of the substrate/chromogen to each well (multichannel pipette or 8 channel pipette); and incubate for 10 min at room temperature (20-25 C / F) in the dark. 6. Add 100 µl of stop solution to each well (multichannel pipette or 8 channel pipette); and measure the absorbance at 450 nm within 5 min. 11. Results A special software, the RIDA SOFT Win / RIDA SOFT Win.net (Art. No. Z9996) is available for evaluation of the RIDASCREEN enzyme immunoassays. The results are given in µg/kg respectively µg/l and µg/100 g respectively µg/100 ml. 19 Remark for the calculation without software: absorbance standard (or sample) absorbance zero standard x 100 = % absorbance The zero standard is thus made equal to 100 % and the absorbance values are quoted in percentages. The values calculated for the standards are entered in a system of coordinates on semi-logarithmic graph paper against the vitamin B12 concentration (µg/kg or µg/l). In order to obtain the Vitamin B12 concentration actually contained in a sample, the concentration read from the standard curve must be further multiplied by the corresponding dilution factor.
