3 4 Summary Phycobilisomes (PBS) are light-harvesting supramolecular aggregates of phycobiliproteins attached to the outer surface of the thylakoids of cyanobacteria and red algae. They collect light energy in a region of the visible spectrum where Chi has low absorbance. Cyanobacterial PBS consist of a central core, mainly composed of allophycocyanin, from which rods formed of phycocyanin or phycoerythrocyanin radiate. In the thermophilic cyanobacterium Mastigocladus laminosus changes in the environmental factors like light intensity and light wavelength have been shown to affect the amount of phycoerythrocyanin (PEC) and phycocyanin (PC) in the rod substructure of the PBS while the core substructure seems to be insensitive to changes in the incident light. These results were obtained by isolation of PBS complexes or by electron microscopy of PBS. The core polypeptides ocap, Pap, (xapb, pie.2,1^8.9 and LcM127 play an important role in energy transfer between PBS and the reaction center. Genes encoding all six core polypeptides were cloned by heterologous DNA hybridisation, sequenced and characterised. They are arranged in three transcriptional units. Two of these units, those encoding apcd and apcf, appear to produce monocistronic mrna. The remaining four genes are adjacent on the chromosome and form a 6.4 kb long apceabc gene cluster. apcc and apcf map next to a 3' ORF1 and ORF2 respectively, which are transcribed in the opposite direction to the respective ape-genes. The corresponding amino acid sequences of these two ORF don't show any relevant homology to known protein sequences. It is interesting to note that the order of the genes, with the apca gene occurring 5' to the apcb gene, is opposite to that observed for all cpcba operons and pec(cbe)ba operons. No classical E.coli ribosome binding site was found in front of the apcd coding region, while apcf has a 13 bp long ribosome binding site just in front of the start codon. Comparisons of the typical E.coli promoter consensus sequence with the 5' region of cloned DNA fragments showed possible RNA polymerase binding sites for apca, apcc, apce and apcd. The apcf gene does not exhibit any significant homology to known Computer analysis hase shown that energetically promoter sequences. favourable hairpins, with energy of formation in the range of -55 to -94 kcal/mol, can form at the end of apce, apcb, apcd and apcf. The gene products of apca (486 bp) and apcd (485 bp) predict a 161 amino acid long ocap and a 162 amino acid long ocapb polypeptide respectively. apcb (486 bp) and apcf (513 bp) code for a 161 amino acid long pap ^d a 159 amino acid long pi6.2 polypeptide respectively. Sequence identity between APa subunits is 53% in the amino acid sequence, while it is 57% for the APp subunits. For comparison it is important to mention that ocap and aa?b as well as PA? and p16-2 are less

4 5 conserved than apc and ocpec (62% identity on polypetide sequence) or PPC and ppec (67% identity on polypeptid sequence). The two linker polypeptides encoding genes apcc and apce are located on the apc operon. apcc (205 bp) was found downstream from apcab and encodes a small 68 amino acids long linker polypeptide. apce (3627 bp) is located upstream from apcab. The deduced amino acid sequence predicts a polypeptide of 1134 amino acids with Mr kd. This is in good agreement with the size expected from the SDS-PAGE analysis. The amino acid sequences of the apc gene products are very similar to their counterparts of either cyanobacterial or eucaryotic red algae origin. Indeed there is a 79-94% identity with the ape-genes of Synechococcus sp. PCC 6301, Synechococcus sp. PCC 7002, Calothrix sp. PCC 7601 and Cyanophora paradoxa and a 55-60% identity with the eucaryotic Aglaothamnion neglectum. The large core membrane polypeptide Lcm is a multifunctional polypeptide postulated to act as terminal energy acceptor and as a linker polypeptide that stabilizes the PBS core architecture. Protein sequence analysis of Mastiogocladus laminosus shows a N-terminal biliprotein-like domain followed by four linker-like domains. The N-terminal is 21-35% identical to every phycobilisome subunit (either a or P), but is longer than the typical phycobihproteins. Lcm has a short N-terminal extension of 55 residues and an insertion of 70 amino acids (loop) which starts at residue 150 and separates the two homologous regions. Following the biliprotein-like domain and another spacing region (arml), repeated domains (REP's) appear, each about 120 residues long. Homologies of about 60% (42-53% identity) exist between the different REP's as well as between a REP and the first 160 residues of the N-terminal domains of the rod and rod-core linker polypeptides. Interestingly, REP1 shares the best identity with REP3, and REP2 with REP4. REP's are at variable distances from each other (arm 1-5). No homology could be detected between the arm's. All the Lcm sequences known so far, share a similar polypeptide organisation, the only differences being the number of REP's and arm's. Mastigocladus laminosus possesses four REP's on the cdna as well as on the polypeptide. Both fourth's REP of the two Lcm polypeptides present in the PBS are believed to link two additional rods to the core of PBS. A model consistent with all experimental observations is presented for the core of Mastigocladus laminosus. Based on the comparison of the primary structure of the different Lcm domains it is supposed that the N-terminal biliprotein-like domain will form a hetero dimer with P16-2. The biliprotein-like domain differs slightly in the Cys residue location from the classical subunit. Since Lcm is a terminal emitter of excitation located at the interface which will energy, this biliprotein-like domain is probably between the PBS and the thylakoid membrane. The PBS loop, protrude from the trimeric polypeptide complex, could play PBS anchoring. Because of the closer identity a role in the between REP1 and REP3 we

5 6 assume that both of them will most probably interact with similar complexes, so that REP1 and REP2 probably interact with trimers of the basal core cylinders and REP3 is making contact with the top cylinder. REP4 links one additional rod cylinder to the PBS. Given the low sequencial identity of the core polypeptides among themselves, as well as compared to the rod polypeptides, a hexameric arrangement is assumed for the AP-trimers, which is significantly different from the one of the PC and PEC hexamers. Thise arrangement allows the rod-core hnker polypeptides and the arm's to penetrate different complexes in the core. and interact with the

6 7 Zusammenfassung Die Phycobilisomen (PBS) sind hochmolekulare Lichtsammlerantennen, die sich auf der Thylakoidmembran von Cyanobakterien und Rotalgen befinden. Sie beliefern das Reaktionszentrum (RC) mit Lichtenergie aus einem Lichtspektrum ( nm), in dem das Chlorophyll des RC nicht mehr absorbieren kann. Die PBS bestehen aus zwei funktionellen Untereinheiten, der Core-Region und der Rod-Region. Der Core besteht hauptsachlich aus Allophycocyanin und spielt eine wichtige Rolle im Energietransport zwischen PBS und RC. An PBS, welche aus dem Cyanobakterium Mastigocladus laminosus gewonnen wurden, konnte durch Isolation von Polypepndkomplexen und Aufnahmen mit dem Elektronenmikroskop gezeigt werden, dass die Menge Phycoerythrocyanin (PEC) und Phycocyanin (PC) in den Rods auf Anderungen im einfallenden Licht (Lichtintensitat oder Lichtqualitat) reagieren, wahrend die Core-Zusammensetzung keine erkennbare Anpassung zeigt. Die Gene fiir die sechs Core-Polypeptide aap, pap aapb, pie.2,1^,8.9 und Lcm127 sind durch heterologe Hybridisierungen kloniert, sequenziert und charakterisiert worden. Diese Core-Gene befinden sich auf drei verschiedenen DNA Fragmenten, wobei vier, apceabc, hintereinander auf dem AP-Operon angeordnet sind und zwei, apcd und apcf, sich auf unabhangigen, nicht benachbarten DNA-Bereichen befinden. Strangabwarts von apcc bzw. apcf wurde jeweils ein ORF identifiziert, dessen abgeleitete Polypeptidsequenz keine relevante Homologie zu bisher bekannten Polypeptiden ergibt. Im Gegensatz zu der Anordnung auf dem PC- bzw. PEC-Operon befindet sich auf dem AP-Operon apc A strangaufwarts von apcb. Bei der Suche nach Ribosomenbindungsstellen vor den Startcodons der klonierten ape-gene wurde keine typische E.coli Ribosomenbindungsstelle vor apcd gefunden, wahrend apcf nur 2 bp vor dem Startcodon eine 13 bp lange purinreiche Sequenz besitzt. Vergleiche der 5' flankierenden Regionen der klonierten ape-gene mit der fiir Promotoren ermittelten Consensus-Sequenz aus E.coli ergaben mogliche Promotorstellen fiir apca, apcc, apcd und apce. Keine zu E.coli homologe Promotorsequenz ist fur apcf erkennbar. Energetisch stabile Terminationsstrukturen, mit einer Faltungsenergie zwichen -55 und -94 kcal/mol sind nach dem Stopcodon von apce, apcb, apcd und apcf vorhanden. Die aus den Gensequenzen abgeleiteten Polypeptidsequenzen wurden auf Identitaten untersucht. Die Gene apca als auch apcb besitzen 485 bp und codieren jeweils ein Polypeptid mit 161 AS-Resten. Die abgeleiteten Polypeptidsequenzen aus apcd (485 bp) und apcf (522 bp) bestehen fur ocapb aus 161 AS-Reste und fiir P16-2 aus 169 AS-Resten. Sequenzvergleiche der AP a-untereinheiten (aapb und aap)

7 8 untereinander ergeben eine 53% Identitat in der Polypeptidsequenz, jene der AP p-untereinheiten (P*6-2 und pap) eine 57% Identitat. Interessanterweise zeigen die zwei Core-Polypeptide aap und aapb bzw. pap un(j pi6.2 untereinander eine niedrigere Identitat in der Polypeptidsequenz als die entsprechenden Polypeptide des Rod-Bereiches: acp zu apec (62%) bzw. pcp zu ppec (67%). Die Core-Linkerpolypeptid codierenden Gene apcc (205 bp) und apce (3627 bp) codieren jeweils das endstandige 8.9 kd Polypeptid bestehend aus 68 AS und das 1134 AS lange Core-Membran Linkerpolypeptid. Die Core-Polypeptide aus Mastigocladus laminosus zeigen eine hohe Homologie mit den entsprechenden Polypeptiden von Cyanobakterien und Rotalgen. Diese Identitaten bewegen sich im Bereich 79-94% im Vergleich mit Cyanobakterien wie Synechococcus sp. PCC 6301, Synechococcus sp. PCC 7002, Calothrix sp. PCC 7601 und Cyanophora paradoxa und 55-60% Identitat im Vergleich mit der Rotalge Aglaothamnion neglectum. Das Core-Membran Linkerpolypeptid (Lcm) ist eines der funktionell wichtigsten Polypeptiden im PBS. Neben seiner strukturellen Funktion im Core-Aufbau fixiert es das PBS an die Thylakoidmembran und wirkt als terminale Energietransfer-Einheit zwischen PBS und PSII. Die Sequenzanalyse des Core-Membran Linkerpolypeptides aus Mastigocladus laminosus ergab einen N-terminalen biliprotein-ahnlichen Sequenzbereich, gefolgt von vier Linkerpolypeptid-ahnlichen Regionen (REP's). Die N-terminale biliprotein-ahnliche Region ist 21-35% identisch (43-53% ahnlich) zu den sechs Biliproteinen des PBS. Diese Region ist im Vergleich zu den Biliproteinen um 70 AS-Reste langer. Die zusstzlichen 70 AS-Reste bilden die sogenannte "Loop"-Sequenz, welche die biliprotein-ahnliche Sequenz in zwei ungleiche Teile trennt. Die im Linkerpolypeptid vorhandenen 120 AS langen REP-Regionen sind zu 42-53% identisch (60% ahnlich) sowohl untereinander als auch im Vergleich mit den N-terminalen 160 AS-Reste der Rod- bzw. Rod-Core Linkerpolypeptide. Aufgrund der hohen Homologie zwischen REP1 und REP3 kann angenommen werden, dass diese Sequenzregionen ahnliche oder identische Komplexe im Core verbinden. Die REP's werden von verschieden langen, unter sich nicht homologen Sequenzbereichen, den "Arm's" getrennt. Mastigocladus laminosus ist das bis anhin einzige bekannte Cyanobakterium, welches sowohl in der DNA-Sequenz wie auch in der Polypeptidsequenz vier REP's und demzufolge fiinf Arm's besitzt. Ein Modell, das die experimentellen Daten miteinbezieht und erklart, wird fiir den Core-Aufbau in Mastigocladus laminosus vorgestellt. Anhand der gefundene Sequenzregionen im LcM127-Polypeptid und deren Sekundarstrukturen, wird angenommen, dass der biliprotein-ahnliche N-Terminus, zusammen mit dem P16-2 Polypeptid, ein Monomer bildet. Dadurch, dass Lcm als terminale Energietransfer-Einheit fungiert, befindet sich der chromophortragende N-Terminus sehr wahrscheinlich zwischen der Thylakoidmembran und dem Phycobilisom in der Nahe des PSII. Es

8 9 wird angenommen, dass die "Loop"-Sequenz, welche aus dem trimeren Polypeptidkomplex herausragt, als Anker wirken kann und das PBS durch ionische Wechselwirkungen an die Thylakoidmembran fixiert. Es wird des weiteren angenommen, dass die ionischen AS-Reste der REP's mit den einzelnen Core-Komplexen wechselwirken und diesen eine spezifische Anordnung im Core verleihen. REP1 und REP3 verbinden wegen ihrer konservierten Sequenz mit grosser Wahrscheinlichkeit identische Komplexe. REP 1 und REP 2 befinden sich in den basalen Zylindern, wahrend REP3 im oberen Core-Zylinder anzutreffen ist. REP4 fixiert die zwei zusatzlichen AP-Hexamere an den Core. Die geringe Sequenzhomologie der Core-Polypeptide untereinander und im Vergleich mit den Rod-Polypeptiden lasst darauf schliessen, dass die von den AP-Trimeren gebildeten Hexamere sich deutlich von den Rod- Hexameren unterscheiden. Eine alternative Anordung der Core-Hexamere wird vorgeschlagen, welche in der Lage ist, die im Elektronenmikroskop beobachteten langeren Core-Zylinder zu erklaren. Ausserdem entstehen bei dieser Anordnung "Locher" zwischen den AP-Trimeren, in welche die Rod-Core Linkerpolypeptide eingelagert werden konnen.
