5 Institut Virion\Serion GmbH Serion Immundiagnostica GmbH Progress by Experience and Innovation 1978 saw the founding of Institut Virion\Serion GmbH in the city of Würzburg in southern Germany. From the outset the company has specialized in the manufacture of in vitro diagnostic products for the infectious disease serology and has, due to continual growth, twice expanded into new premises over the past 35 years. This family run business with its roots in Würzburg has remained in the region and currently provides employment and career opportunities for around 125 employees. Our product range offers a selection of analysis methodologies to aid the diagnosis of viral, bacterial, parasitic and fungal diseases. The complement fixation test (CFT) range alone offers in excess of 50 antigens plus associated reagents and controls for the detection of pathogen specific antibodies. SERION ELISA classic products allow for the differentiation of antibody classes for disease state and immune status determinations. In contrast, SERION ELISA antigen products allow for direct pathogen detection. Immunomat TM and Gemini are capable of automatically processing SERION ELISA classic and SERION ELISA antigen immunoassays. All products are manufactured exclusively in Würzburg, Germany, by Institut Virion\Serion GmbH in compliance with national and international quality assurance standards related to in vitro medical devices such as the European Directive IVDD 98/79/EC. Zu diesem Zweck werden Antikörper nachgewiesen, die von den Zellen des Immunsystems als Antwort auf die Gegenwart einer fremden Substanz, dem Antigen, gebildet werden und zur Abwehr des Krankheitserregers dienen. Serological investigations are important in the diagnosis of infectious diseases. The immune system produces antibodies as a vital component of the response to infection and the presence of foreign bodies or antigens. The detection of these antibodies is utilized in the diagnostic process. SERION ELISA classic SERION ELISA classic sind qualitative und quantitative Immunoassays für den Nachweis von humanen Antikörpern in Serum, Plasma und ggf. Liquor cerebrospinalis gegen spezifische Antigene von Bakterien, Viren, Pilzen oder Parasiten zur Unterstützung bei der Diagnose von Infektionserkrankungen. SERION ELISA classic tests are qualitative and quantitative immunoassays for the detection of human antibodies directed against specific antigens of bacteria, viruses, fungi and parasites in serum, plasma or, when indicated, cerebrospinal fluid, for the diagnosis of infectious diseases. Testprinzip Der ELISA (Enzyme Linked Immunosorbent Assay) ist ein immunologisches Nachweisverfahren, das sich insbesondere in der Infektionsserologie zur Erfassung von Antikörpern bewährt hat. Die Reaktion basiert auf der spezifischen Interaktion von Antikörpern mit der nachzuweisenden Substanz. Zu diesem Zweck werden die Teststreifen der Miktotiterplatten der SERION ELISA classic mit spezifischen Antigenen von Infektionserregern beschichtet, an die in der Patientenprobe vorhandene Antikörper binden. Ein weiterer Antikörper, der zuvor mit dem Enzym Alkalische Phosphatase markiert wurde, detektiert den so gebildeten Immunkomplex. Dieses Enzym katalysiert eine Reaktion, in deren Verlauf das farblose Substrat p-nitrophenoylphosphat in das farbige Produkt p-nitrophenol umgewandelt wird. Die Signalstärke des Reaktionsprodukts wird photometrisch erfasst und ist proportional zur Menge der nachzuweisenden Substanz in der Probe

9 Test Principle The ELISA (Enzyme Linked Immunosorbent Assay) is an immunoassay, which is particularily suited to the determination of antibodies in the field of infectious serology. The reaction is based on the specific interaction of antibodies with their corresponding antigen. Therefore, the test strips of the SERION ELISA classic microtiter plate are coated with specific antigens of the pathogen of interest. If antibodies in the patient s serum sample are present, they bind to the fixed antigen. A secondary antibody, which has been conjugated with the enzyme alkaline phosphatase, detects the immune complex. The colorless substrate p-nitrophenylphosphate is then converted into the colored product p-nitrophenol. The signal intensity of the reaction product is measured photometrically and proportional to the concentration of the analyte in the sample. An indicator system consisting of antibody coated erythrocytes (haemolytic system) is finally added to the system after a suitable time lapse for the complement fixation to complete. Lysis of the indicator system is dependent upon the presence or absence of immune complexes formed between the sample and specific antigen used. No lysis indicates the presence of immune complexes while lysis indicates non. Mild centrifugation of the system sediments any surviving erythrocytes, allowing the test to be evaluated visually. A button of red cells indicates a positive result whereas a reddish clear solution is negative
