V Summary Animal cell cultures are mainly used for the production of compounds needed in the human health industry. Optimization strategies are aiming at high viable cell densities required, for a high volumetric productivity, to decrease the cost prize of the product. Cell death may prevent the attainment of the high viable cell density, while dead cells may lead to the forming of cell debris and the release of intracellular compounds, such as DNA, proteases and underprocessed product, thus interfering with the downstream processing and the final product quality. Therefor cell death poses a main problem in production cultures and was the subject of study. Two forms of cell death may occur: necrosis and apoptosis. Necrotic death, is marked by an increase in the cell volume and a violent breakdown of all cell organelles including membranes, while apoptotic death is an active process regulated from within the cell, marked by DNA fragmentation, chromatin condensation, phosphatidylserine exposure to the surface and finally breaking up of the cell in apoptotic bodies. In vivo these apoptotic bodies are taken up by neigboring cells, while in cell cultures these apoptotic bodies will break up and release their content in the medium, a process called secondary necrosis. Until this process the cell membranes maintain their integrity. Classical methods for the analyses of cell death are based on the breakdown of the membrane integrity, therefor new methods for the measurement of.especially apoptotic, cell death were evaluated. Staining of the cell nucleus with DAPI in order to detect condensated chromatin, as well as the labeling of phosphatidylserine exposure on the cell surface with annexin V-FITC were found to be the methods for the earliest detection of apoptotic cell death. Both events were found to coincide in CHOSSF3 cells, where apoptosis was induced with actinomycin D. Only the increase in cell size was found to proceed both events, however, the cell size can not be used as a detection parameter in a heterogeneous bioreactor system. The exposure of phosphatidylserine observed during all to the cell surface was also phases of mitosis in CHOSSF3 cells. This poses a problem for the use of the annexin V staining method for the measurement of apoptosis.
VI Using the DAPI staining method, apoptotic death could be observed in batch cultivations of hybridoma and CHO cell lines, grown in serum free medium under standard conditions. These cell lines were adapted to serum free media and the apoptotic induction must be due to depletion of media components or the accumulation of waste products rather than the lack of serum components. The effects of the antibiotics, gentamicin and streptomycin, alone and in combination with pluronic were re-evaluated with the emphasis on induction of apoptotic death. In rank of sensitivity hybridoma»chossf3>chossf6, apoptotic death was observed upon addition of the antibiotics, whereby gentamicin was found to be a more potent inducer. Pluronic was found to be an even stronger inducer of apoptosis. Combination of pluronic with gentamicin did not cause an additional effect. Both compounds are interacting with the cell membrane and may induce the same apoptotic pathway, pluronic overruling the effect of gentamicin. In L929 cells contaminated with mycoplasma, the nuclear structure was observed to mimic the apoptotic nuclear structure, this can lead to the detection of artifacts during the study of apoptosis. For the detection of unknown compounds in spent media and of protein patterns of cell lysates, may be related to cell death, a new method was developed using MALDI MS as analytical tool. Using MALDI-MS compatible detergents it was possible to extend the mass range detectable by MALDI MS from currently 16.000 Da to 80.000 Da. These new method have been applied to gene expression experiments to observe the succes of the translation as well as for the monitoring of large scale cultivation of hybridoma cells expressing an antibody of the IgG type. Quantitative measurements of the IgG level during the cultivation compared favourably with those obtained by affinity HPLC. Insulin, light chain of the antibody and a specific protein with 8650 Da that might be linked to cell viability were monitored for the complete cultivation time to allow for the optimization of the production conditions. Cell death can be detected in production cell lines using the DAPI staining method and/or the labeling with annexin V. Future monitoring of cell death coupled with process control as on line processes, may increase the ability to maintain the apoptotic death at a low level.
VII Zusammenfassung Tierische Zellkulturen werden, neben in vitro Gentherapie und in vitro Zelltherapie, hauptsachlich fur die Produktion von Pharmaka verwendet. Optimierungsstrategien wollen hohe Zelldichten erzielen, urn eine hohe volumetrische Produktivitat zu erreichen, und urn die Produktionskosten zu senken. Zelltod ist in zweierlei Hinsicht unerwunscht: zum einen verhindert er hohe lebende Zelldichten; zum anderen fuhrt er zu Zelldebris und zur Freisetzung intrazellularer Komponenten wie DNA, Proteasen und nicht ganz prozessierte Produkte, die beim Downstream-Processing storen und/oder die Produktqualitat vermindern. Zelltod ist demnach ein Hauptproblem bei der Produktion und ist Thema dieser Arbeit. Die beiden Formen von Zelltod sind Nekrose und Apoptose. Wahrend beim passiven ProzeR der Nekrose das Zellvolumen zunimmt und Zellorganellen, einschlieblich Membranen, zerstort werden, ist die Apoptose ein aktiver ProzeR, bei dem die DNA fragmentiert, das Chromatin kondensiert wird, Phosphatidylserin auf der Oberflache erscheint und letzten Endes die Zelle in sogenannte "Apoptotic Bodies" zerfallt. Im folgenden der Nekrose ahnlichen ProzeB zerfallen wiederum diese "Apoptotic Bodies" und ihr Inhalt wird freigesetzt. Bis dahin bleibt die Membranintegritat aber erhalten. Klassische Methoden zur Analyse von Zelltod basieren auf dem Zusammenbruch der Membranintegritat. Deshalb werden hier neue Methoden zur Ermittlung des Zelltodes, vornehmlich der Apoptose, erarbeitet. In der vorliegende arbeit sind die Farbung des Zellkerns mit DAPI zur Detektion kondensierten Chromatins sowie die Markierung von Phosphatidylserin auf der Zelloberflache mit Annexin V-FITC als Methoden zur fruhsten Detektion gefunden worden. Beide Ereignisse kommen gleichzeitig in CHOSSF3 Zellen vor, die mit Actinomycin D zur Apoptose induziert wurden. Allein das Zunehmen der Zellgrosse geht diesen beiden Ereignissen voraus; sie kann aber nicht als Bestimmungsparameter in einen Bioreaktorsystem herangezogen werden. Bei der Analyse von Apoptose verursacht die, bei CHOSSF3 beobachtete, Translokation von Phosphatidylserin wahrend der
VIII ganzen Mitose, ein Problem bezuglich Aussagekraft der Annexin V Farbemethode. Mittels DAPI Farbung konnte die Apoptose in Hybridoma und CHO Zellinien gegen Ende von Batchkulturen nachgewiesen werden. Beide Zellinien waren auf serumfreie Medien adaptiert und wurden unter Standardbedingungen kultiviert. Die Apoptose tritt hier eher als Folge von limitierende Medienkomponenten und/oder der Anhaufung von Abfallprodukten vom Fehlen der Serumfaktoren. auf denn In Hinblick auf die Induktion von Apoptose wurde die Wirkung der Antibiotika Gentamicin und Streptomycin wurden je allein und in Kombination mit Pluronic F68 untersucht. Nach der Beifugung des Agens wurde die Apoptose gemessen, wobei Pluronic sich als der starkste Induktor erwies, gefolgt von Gentamicin. Beziiglich sensitivitat nimmt bei den untersuchten Zelllinien Hybridoma die erste Stelle ein, gefolgt von SSF3 und SSF6. Die Kombination von Pluronic und Gentamicin verursacht keinen additiven Effekt. Beide Komponenten interagieren mit Zellmembranen und induzieren wohl eine Apoptose uber denselben Mechanismus. Fur die Analysen von unbekannten Komponenten in konditionierten Medien und von Proteinstrukturen lysierter Zellen, die mit dem Zelltod in Zusammenhang stehen durften, ist basierend auf MALDI MS eine neue Methode entwickelt worden. Dank dieser Methode ist eine 8kDa schwere Komponente gefunden worden, die mit dem Zelltod zusammenhangen durfte. Zur Bestimmung des Zelltodes bei Produktionszellinien kommen nur die DAPI Farbungsmethode und/oder die Markierung mit Annexin V in frage. Die zukunftige Entwicklung dieser Analysemethode in Richtung einer on-line Oberwachungsmethode wird die Moglichkeit bringen den apoptotischen Zelltod auf einem tiefen Niveau zu halten.
