4 1. Verwendungszweck RIDASCREEN FAST Folsäure ist ein kompetitiver Enzymimmunoassay zur quantitativen Bestimmung von Folsäure in vitamisierten Lebensmitteln, Futtermitteln und Vitaminprodukten. 2. Allgemeines Folsäure, früher als Vitamin B 9 bezeichnet, gehört zur Gruppe der wasserlöslichen Vitamine und ist ein wichtiger Nahrungsfaktor und Futtermittelzusatz. Folsäure spielt eine Schlüsselrolle für alle Wachstums- und Entwicklungsprozesse im Körper. 3. Testprinzip Die Vertiefungen der Mikrotiterplatte sind mit spezifischen Antikörpern gegen Folsäure beschichtet. Zugegeben werden die Folsäure-Standardlösungen bzw. die Probelösungen und enzymmarkierte Folsäure (Enzymkonjugat). Freie und enzymmarkierte Folsäure konkurrieren um die Folsäure-Antikörperbindungsstellen (kompetitiver Enzymimmunoassay). Nicht gebundene enzymmarkierte Folsäure wird anschließend in einem Waschschritt wieder entfernt. Der Nachweis erfolgt durch Zugabe von Substrat-/Chromogenlösung. Gebundenes Enzymkonjugat wandelt das Chromogen in ein blaues Endprodukt um. Die Zugabe des Stopp- Reagenzes führt zu einem Farbumschlag von blau nach gelb. Die Messung erfolgt photometrisch bei 450 nm; die Extinktion der Lösung ist umgekehrt proportional zur Folsäure-Konzentration in der Probe. 4 RIDASCREEN FAST Folsäure

5 4. Packungsinhalt Mit den Reagenzien einer Packung können 48 Bestimmungen durchgeführt werden (einschließlich Standardbestimmungen). Jeder Testkit enthält: 1 x Mikrotiterplatte mit 48 Kavitäten (6 Streifen à 8 Einzelkavitäten) beschichtet mit Antikörpern gegen Folsäure 6 x Standardlösungen, je 1,3 ml 0 ppb (Nullstandard), 1 ppb, 2 ppb, 5 ppb, 10 ppb, 25 ppb Folsäure in wässriger Lösung, gebrauchsfertig 1 x Konjugat (3 ml)...roter Verschluss Peroxidase-konjugierte Folsäure gebrauchsfertig 1 x Substrat-/Chromogenlösung (10 ml)...brauner Verschluss rötlich gefärbt, enthält Harnstoffperoxid 1 x Stopp-Reagenz (14 ml)... gelber Verschluss enthält 1 N Schwefelsäure 1 x Puffer (50 ml) Probenverdünnungspuffer, gebrauchsfertig 5. Zusätzlich benötigte Reagenzien erforderliches Zubehör 5.1. Geräte: Mikrotiterplatten-Photometer (450 nm) Schlagmühle Zentrifuge + zentrifugierbare Reagenzgläser Schüttler Wasserbad (100 C) Papierfaltenfilter und Trichter Messpipetten variable 20 µl µl und µl Mikropipetten 5.2. Reagenzien: Probenverdünnungspuffer für alle Proben, falls die mitgelieferten 50 ml nicht ausreichen: PBS-Puffer ohne Tween, ph 8,0 (0,55 g NaH 2 PO 4 x H 2 O + 2,85 g Na 2 HPO 4 x 2 H 2 O + 9 g NaCl ad 1000 ml dest. Wasser, ph 8 mit NaOH einstellen) Der Puffer hat eine Haltbarkeit von 1 Woche bei Raumtemperatur bzw. ca. 4-6 Wochen bei 2-8 C. RIDASCREEN FAST Folsäure

6 Natrium-Phosphatpuffer für die Probenaufarbeitung von angereichertem Getreidemehl: 200 mm Natrium-Phosphatpuffer, ph 7,0 (21,6 g Na 2 HPO 4 x 2 H 2 O + 10,8 g NaH 2 PO 4 x H 2 O in 1000 ml dest. Wasser lösen und ph 7,0 +/- 0,1 einstellen) Der Puffer hat eine Haltbarkeit von 4 Wochen bei Raumtemperatur. 20 mm Natrium-Phosphatpuffer, ph 7,0 (den 200 mm Natrium-Phosphatpuffer, ph 7,0, 1:10 (1+9) mit dest. Wasser verdünnen) Der Puffer hat eine Haltbarkeit von 1 Woche bei Raumtemperatur bzw. ca. 4-6 Wochen bei 2-8 C. 6. Vorsichtsmaßnahmen Dieser Test ist nur von geschultem Laborpersonal durchzuführen. Die Gebrauchsanweisung zur Durchführung des Tests ist strikt einzuhalten. Das Stopp-Reagenz enthält 1 N Schwefelsäure (R36/38, S2-26). 7. Reagenzien und ihre Lagerung Die Reagenzien bei 2-8 C lagern. Komponenten des Testkits auf keinen Fall einfrieren. Nicht benötigte Kavitäten zusammen mit dem Trockenmittel im Folienbeutel gut verschlossen aufbewahren und weiterhin bei 2-8 C lagern. Die Substrat-/Chromogenlösung ist lichtempfindlich, deshalb direkte Lichteinwirkung vermeiden. Nach Ablauf des Verfallsdatums (siehe Testkit-Außenetikett unter Expiration) kann keine Qualitätsgarantie mehr übernommen werden. Ein Austausch von Einzelreagenzien zwischen Kits verschiedener Chargennummern ist nicht zulässig. 8. Probenvorbereitung Die Proben kühl und lichtgeschützt lagern Milch und Milchpulver Milch: zur Entfettung zentrifugieren: 3500 g / 10 min / 10 C die obere Fettschicht mittels einer Pasteurpipette über Vakuum absaugen die entfettete Milch mit Probenverdünnungspuffer 1:10 (1+9) verdünnen 50 µl pro Kavität im Test einsetzen Milchpulver: 10 g Milchpulver in ca. 50 ml dest. Wasser lösen, mit dest. Wasser auf 100 ml auffüllen und 10 min schütteln / rühren 10 ml Probe erhitzen: 3 min / 100 C Probe auf Raumtemperatur (20-25 C) unter Eis abkühlen, dabei mischen zur Entfettung zentrifugieren: 3500 g / 10 min / 10 C die obere Fettschicht mittels einer Pasteurpipette über Vakuum absaugen die entfettete Milch mit Probenverdünnungspuffer 1:10 (1+9) verdünnen 50 µl pro Kavität im Test einsetzen Anmerkung: Zur Qualitätskontrolle wird empfohlen Kontrollproben (z.b. eine nicht vitaminisierte Milchpulverprobe) mitzuführen, um eventuell falsch positive Ergebnisse bei Milchpulverproben zu erkennen Getreide und Müsli Eine repräsentative Probe vor dem Extrahieren zerkleinern und mischen. 5 g der gemahlenen Probe in ein verschraubbares Reagenzglas einwiegen, in ca. 50 ml dest. Wasser lösen, mit dest. Wasser auf 100 ml auffüllen bis zum vollständigen Lösen der Probe schütteln (manuell oder mittels Schüttler) Probe erhitzen: 3 min / 100 C Probe auf Raumtemperatur (20-25 C) unter Eis abkühlen, dabei mischen zentrifugieren: 4000 g / 10 min / 15 C (alternativ: filtrieren) Überstand abnehmen, falls der Überstand noch trüb sein sollte, über einen Papierfilter filtrieren RIDASCREEN FAST Folsäure

8 Überstand oder Filtrat 1:5 (1+4) mit Probenverdünnungspuffer verdünnen (z.b. 0,1 ml Überstand oder Filtrat + 0,4 ml Probenverdünnungspuffer) 50 µl pro Kavität im Test einsetzen 9.3. Angereicherte Getreidemehle Eine repräsentative Probe vor dem Extrahieren zerkleinern und mischen. 1 g der homogenen Mehlprobe in ein verschraubbares 50 ml Reaktionsgefäß, z.b. Polypropylenröhrchen, einwiegen, 40 ml 20 mm Natrium-Phosphatpuffer zugeben und gut mischen das Reaktionsgefäß im kochenden Wasserbad 30 min erhitzen (3-5 mal schütteln) Probe auf Raumtemperatur (20-25 C) unter Eis abkühlen zentrifugieren: 3000 g / 10 min / Raumtemperatur Überstand 1:10 (1 + 9) oder 1:20 (1 + 19) mit Probenverdünnungspuffer verdünnen 50 µl im Test einsetzen 9.4. Vitaminpulver, -premixes, -tabletten, -kapseln und -bonbons Bei der Probenvorbereitung wird mehr Probenverdünnungspuffer benötigt, als im Test mitgeliefert wird. Deshalb vor Beginn eine ausreichende Menge Probenverdünnungspuffer herstellen (siehe 5.2.). 1 g Vitaminpulver /-premix in 100 ml Probenverdünnnungspuffer lösen Tabletten zermörsern und 1g in 100 ml Probenverdünnnungspuffer lösen Kapseln: 1 Kapsel wenn möglich aufschneiden und den Inhalt in 100 ml Probenverdünnnungspuffer lösen Bonbons: 1 Bonbon in 100 ml Probenverdünnnungspuffer lösen Mit allen Proben wie folgt weiterarbeiten: jeweilige Probelösung erhitzen: 3 min / 100 C (dabei löst sich die Probe vollständig) Probe auf Raumtemperatur (20-25 C) unter Eis abkühlen, dabei mischen zentrifugieren: 4000 g / 10 min / 15 C (alternativ: filtrieren) Überstand abnehmen, falls noch Schwebteilchen vorhanden sind, Überstand über einen Papierfilter filtrieren Überstand oder Filtrat 1:10 (1+9) mit Probenverdünnungspuffer verdünnen (z.b. 0,1 ml Überstand oder Filtrat + 0,9 ml Probenverdünnungspuffer) 50 µl pro Kavität im Test einsetzen 8 RIDASCREEN FAST Folsäure

9 9.5. Vitaminsäfte Bei der Probenvorbereitung wird mehr Probenverdünnungspuffer benötigt, als im Test mitgeliefert wird. Deshalb vor Beginn eine ausreichende Menge Probenverdünnungspuffer herstellen (siehe 5.2.). 1 ml Saft 1:100 (1+99) mit Probenverdünnungspuffer verdünnen zentrifugieren: 4000 g / 10 min / 15 C (alternativ: filtrieren) 50 µl Überstand oder Filtrat pro Kavität im Test einsetzen Anmerkung: Hoch konzentrierte Proben müssen weiter verdünnt werden. 10. Testdurchführung Testvorbereitungen Alle Reagenzien vor Gebrauch auf Raumtemperatur (20-25 C) bringen Testdurchführung Sorgfältiges Waschen ist sehr wichtig. Ein Eintrocknen der Kavitäten zwischen den Arbeitsschritten vermeiden. 1. So viele Kavitäten in den Halterahmen einsetzen, wie für alle Standards und Proben benötigt werden. Die Positionen der Standards und der Proben protokollieren. 2. Je 50 µl der Standardlösung bzw. der vorbereiteten Proben in die entsprechenden Kavitäten pipettieren; für jeden Standard oder Probe neue Pipettenspitzen benutzen. 3. Je 50 µl Enzymkonjugat-Lösung (roter Verschluss) in die entsprechenden Kavitäten pipettieren, vorsichtig manuell mischen und 15 min (+/-1) bei Raumtemperatur (20-25 C) inkubieren. 4. Die Kavitäten durch Ausschlagen der Flüssigkeit leeren und die Restflüssigkeit durch kräftiges Ausklopfen (dreimal hintereinander) auf saugfähigen Labortüchern entfernen. Die Kavitäten mit Hilfe einer Multikanal-Pipette mit dest. Wasser waschen (250 µl/kavität), die Kavitäten leeren und die Restflüssigkeit entfernen. Diesen Waschvorgang noch zweimal wiederholen. 10 5. Je 100 µl Substrat-/Chromogenlösung (brauner Verschluss) in jede Kavität pipettieren. Vorsichtig manuell mischen und 10 min (+/- 0,5) bei Raumtemperatur (20-25 C) im Dunkeln inkubieren. 6. Je 100 µl Stopp-Reagenz (gelber Verschluss) in jede Kavität pipettieren und vorsichtig manuell mischen. Die Extinktion bei 450 nm innerhalb von 10 min nach Zugabe des Stopp-Reagenzes messen. 11. Auswertung Für die Auswertung ist bei R-Biopharm eine speziell für die RIDASCREEN Enzymimmunoassays entwickelte Software, die RIDA SOFT Win (Art. Nr. Z9999), erhältlich. Der Verlauf der Standardkurve kann dem beigefügten Analysenzertifikat entnommen werden. Hinweis für die Berechnung ohne Software: Extinktion Standard (bzw. Probe) Extinktion Nullstandard x 100 = % Extinktion Den Nullstandard somit gleich 100 % setzen und die Extinktionswerte in Prozent angeben. Die errechneten Werte für die Standards in einem Koordinatensystem auf halblogarithmischem Millimeterpapier gegen die Folsäure-Konzentration [µg/kg] bzw. [µg/l] auftragen. Um die in den Proben enthaltene tatsächliche Folsäure-Konzentration zu erhalten, muss die aus der Eichkurve abgelesene Konzentration noch mit dem entsprechenden Verdünnungsfaktor multipliziert werden. Beim Arbeiten nach den angegebenen Vorschriften gelten die in der nachfolgenden Tabelle 1 aufgeführten Verdünnungsfaktoren. Im Falle von Vitaminkapseln bezieht sich die Berechnung auf den gesamten Kapselinhalt und hängt somit vom individuellen Volumen einer jeden Kapsel ab. 10 RIDASCREEN FAST Folsäure

A microtiter plate spectrophotometer is required for quantification. 13 1. Intended use RIDASCREEN FAST Folsäure (Folic acid) is a competitive enzyme immunoassay for the quantitative analysis of folic acid in fortified food, feed and vitamin products. 2. General Folic acid, also known as vitamin B 9, belongs to the water soluble vitamins. It is an important factor in food and also an important feed additive. Folic acid plays a key role in all growth and development processes in the body. 3. Test principle The microtiter plate is coated with specific antibodies against folic acid. Folic acid standards, respectively sample solution and enzyme labeled folic acid (enzyme conjugate) are added. Free and enzyme labeled folic acid compete for the antibody binding sites (competitive enzyme immunoassay). Any unbound enzyme conjugate is then removed in a washing step. Substrate/chromogen is added to the wells, bound enzyme conjugate converts the chromogen into a blue product. The addition of the stop solution leads to a color change from blue to yellow. The measurement is made photometrically at 450 nm. The resulting absorbance values are inversely proportional to the folic acid concentration of the sample. RIDASCREEN FAST Folsäure

14 4. Reagents provided Each kit contains sufficient materials for 48 measurements (including standard analyses). Warnings and precautions for the users This test should only be carried out by trained laboratory employees. The instruction for use must be strictly followed. The stop solution contains 1 N sulfuric acid (R36/38, S2-26). 7. Storage instructions Store the kit at 2-8 C (35-46 F). Do not freeze any test kit components. Return any unused microwells to their original foil bag, reseal them together with the desiccant provided and further store at 2-8 C (35-46 F). The substrate/chromogen solution is light sensitive, therefore, avoid exposure to direct light. No quality guarantee is accepted after the expiration date on the kit label. Do not interchange individual reagents between kits of different lot numbers. 8. Test implementation Test preparation Bring all reagents to room temperature (20-25 C / F) before use Test procedure Carefully follow the recommended washing procedure. Do not allow microwells to dry between working steps. 1. Insert a sufficient number of wells into the microwell holder for all standards and samples to be run. Record standard and sample positions. 2. Add 50 µl of each standard solution or prepared sample to separate wells; use a new pipette tip for each standard or sample. 3. Add 50 µl of enzyme conjugate solution (red cap) to each well. Mix gently by shaking the plate manually and incubate 15 min (+/-1) at room temperature (20-25 C / F). 4. Pour the liquid out of the wells and tap the microwell holder upside down vigorously (three times in a row) against absorbent paper to ensure complete removal of liquid from the wells. Using a multichannel pipette, fill the wells with distilled or deionized water (250 µl/well). Empty the wells again and remove all remaining liquid. Repeat the washing step two more times. 5. Add 100 µl of the substrate/chromogen solution (brown cap) to each well. Mix gently by shaking the plate manually and incubate for 10 min (+/- 0.5) at room temperature (20-25 C / F) in the dark. 6. Add 100 µl of stop solution (yellow cap) to each well. Mix gently by shaking the plate manually and measure the absorbance at 450 nm within 10 min. 18 RIDASCREEN FAST Folsäure

19 11. Results A special software, the RIDA SOFT Win (Art. No. Z9999), is available for evaluation of the RIDASCREEN enzyme immunoassays. The course of the standard curve is shown in the Quality Assurance Certificate enclosed in the test kit. Remark for the calculation without software: absorbance standard (or sample) absorbance zero standard x 100 = % absorbance The zero standard is thus made equal to 100 % and the absorbance values are entages. The values calculated for the standards are entered in a system of coordinates on semilogarithmic graph paper against the folic acid concentration [µg/kg] or [µg/l]. In order to obtain the folic acid concentration actually contained in a sample, the concentration read from the standard curve must be further multiplied by the corresponding dilution factor. When working in accordance with the regulation stated, the dilution factors can be read out of Table 1.
