3 5 1. Zusammenfassung/Summary 1.1 Zusammenfassung Klebsiella pneumoniae benötigt Natriumionen für das anaerobe Wachstum auf Citrat als einziger Kohlenstoff- und Energiequelle. Citrat wird durch einen Na+-Citrat Symporter in die Zelle transportiert und mit Hilfe der Citrat-Lyase in Oxalacetat und Acetat gespalten. Oxalacetat wird durch eine membrangebundene Na+-Pumpe, die Oxalacetat Decarboxylase, in Pyruvat und CO2 umgewandelt. Durch weitere Reaktionen wird Pyruvat zu Acetat umgesetzt, wobei ein ATP durch Substratkettenphosphorylierung gewonnen wird. Ein Problem der Citratfermentation ist, dass kein NADH gebildet wird, welches für biosynthetische Reaktionen verwendet werden kann. Die Stöchiometrie der Citratfermentation ergibt: aus 1 Citrat entstehen 2 Acetat, 0.5 Formiat und 1.2 CO2. Die Bildung von weniger als 1 Formiat und mehr als 1 CO2 pro Citrat weist darauf hin, dass durch eine Teiloxidation von Formiat zu CO2 Reduktionsäquivalente für die Assimilation von Citrat in den Zellkohlenstoff zur Verfugung gestellt werden. Eine membrangebundene Elektronen Transportkette ist offenbar in die NADH-Synthese solcher Zellen involviert. Die membrangebundene Formiat Dehydrogenase katalysiert die Oxidation von Formiat zu CO2, wobei Ubichinon als Elektonenakzeptor dient. In Gegenwart eines elektrochemischen Na+- Gradienten, kann Chinol seine Elektronen auf NAD+ übertragen. Dabei wird NAD+ zu NADH reduziert (umgekehrter Elektonentransfer). Es konnte gezeigt werden, dass bei einem chemischen Na+-Gradient (ApNa+) von mehr als -100 mv das endogene NADH- Niveau von K. pneumoniae in Gegenwart von Formiat zunahm. Die Bildung von NADH wurde in Gegenwart von Sauerstoff oder durch die Zugabe von Hydroxyquinoline-N-Oxid (HQNO), einem spezifischen Hemmstoff für NADH:Ubichinon Oxidoreduktase, aufgehoben. Inside-out Membranvesikel konnten die Reduktion von NAD+ zu NADH mit Formiat als Elektrondonor katalysieren, wenn man einen elektrochemischen Na+-Gradienten von 120 mv anlegte. Dieser setzt sich aus ApNa+ und Ay von gleicher Grössenordnung zusammen. Sowohl ApNa+ als auch A\ f waren essentiell für die NADH Bildung. Sauerstoff oder HQNO konnten die Reaktion inaktivieren. 4 6 Elektronentransfer in anaeroben K. pneumoniae Zellen ist eine essentielle Anforderrung für die Synthese von Zellmaterial aus Citrat. Vibrio alginolyticus ist ein fakultativ anaerob, marines Bakterium, das Na+ Ionen für sein Wachstum benötigt. Dieser Organismus kann bei alkalischem ph in Gegenwart von Carbonylcyanid m-chlorophenylhydrazon (CCCP), einem Protononophor, wachsen. In diesem Organismus wurde eine Na+-abhägige NADH:Ubichinon Oxidoreduktase (Na+- NQR) als Komponente der Atmungskette gefunden. Wie bereits aus früheren Arbeiten bekannt war, bestand dieses Enzym aus nur drei Untereinheiten und enthielt sowohl FAD als auch FMN als prosthetische Gruppen. Da alle gereinigten IT-abhängigen NADH:Ubichinon Oxidoreduktasen von eukaryotischen und prokaryotischen Organismen aus mehreren Untereinheiten bestehen und nur FMN und mehrere Eisen- Schwefel Cluster als prosthetische Gruppen besitzen, wurden die Untereinheitenkomposition und die prosthetischen Gruppen die Na+-NQR genauer untersucht. In dieser Arbeit wurde ein neues Reinigungsverfahren für die Na+-NQR von V. alginolyticus mit dem Detergenz Lauryldimethylamin N-Oxid (LDAO) etabliert. Vier dominante Banden, sowie mehrere Nebenbanden konnten mittels SDS-PAGE einer gereinigten Preparation sichtbar gemacht werden. Drei der vier dominanten Polypeptide wurden als Genprodukte des nqr Operons, welches für die Na+-NQR kodiert, identifiziert. Das gereinigte Enzym zeigt sowohl NADH Dehydrogenase als auch Chinon Reduktase Aktivität. Die beiden Aktivitäten konnten wirksam durch AgMonen (Kj 0.2 JIM) gehemmt werden. Die Chinon Reduktase Aktivität wurde durch HQNO (Ki 0.4 p.m) gehemmt und durch Nationen (K, 10 mm) aktiviert. Chemische und spektroskopische Analysen haben gezeigt, dass das gereinigte Enzym ein FAD und einen [2Fe-2S] Typ Eisen-Schwefel Cluster als prosthetische Gruppen enthält. Die Funktionen der prosthetischen Gruppen wurden untersucht und ein hypotethisches Modell für den Elektronentransfer vorgeschlagen. Die gereinigte NADH:Ubichinon Oxidoreduktase wurde durch Verdünnung mit Octylglucosid, in Liposomen eingebaut und der Na+-Transport in die entsprechenden

5 7 Proteoliposomen untersucht. Unter geeigneten Bedingungen katalysierten diese Proteoliposomen die Na+-Aufnähme in Gegenwart von NADH und Ubichinon-1. Die Stöchiometrie des Na+-Einstromes lag bei 0.5 Na+ pro NADH. Der Transport von Na+- Ionen wurde durch HQNO oder Monensin gehemmt, und durch CCCP und Valinomycin stimuliert. Diese Daten beweisen, dass die aus V. alginolyticus isolierte Na*-NQR ein primäre Na+ Pumpe ist. Ein indirekter Na+-Transportmechanisrnus durch eine IT-abhängige NADH:Ubichinon Oxidoreduktase in Kombination mit einem Na7H+ Antiporter ist dadurch ausgeschlossen.

6 8 1.2 Summary Klebsiella pneumoniae needs sodium ions when grown under anaerobic condition with citrate as sole carbon and energy source. Citrate is transported into the cell by a Na+-citrate symporter and is then cleaved by citrate lyase to oxaloacetate and acetate. Oxaloacetate is degraded to pyruvate and CO2 by the membrane-bound Na+-pump oxaloacetate decarboxylase. Through further reactions, pyruvate is converted to acetate forming one ATP by Substrate level phosphorylation. A problem of the citrate fermentation pathway is that no NADH is formed which could be used for biosynthetic reactions. The stoichiometry of citrate fermentation is 1 citrate fermented to 2 acetate, 0.5 formale and 1.2 C02. The formation of less than 1 formate and more that 1 CO2 per citrate can be accounted for by the oxidation part of the formate to C02 in order to provide reducing equivalent for the assimilation of citrate into cell carbon. A membrane-bound electron transport chain is apparently involved in NADH synthesis by these cells. Membrane-bound formate dehydrogenase catalyses the oxidation of formate to C02 with ubiquinone as electron acceptor. In the presence of an electrochemical Na+ gradient (Ap:Na+), the quinol can further deliver its electrons to NAD+ and reduce it to NADH (reversed electron transfer). It was found that the endogenous NADH level of K. pneumoniae increased in the presence of formate and in response to a chemical Na+ gradient (ApNa+) of more than -100 mv. NADH formation was completely abolished in the presence of oxygen or after addition of hydroxyquinoline-n-oxide, a specific inhibitor for NADH:ubiquinone oxidoreductase. Inverted membrane vesicles of K. pneumoniae catalysed the reduction of NAD* to NADH with formate as electron donor after application of an electrochemical Na+ gradient of about 120 mv consisting of ApNa+ and AH-* of about the same magnitude. Both ApNa+ and t^v were essential for NADH formation and oxygen or hydroxyquinoline-n-oxide inactivated the reaction. The NADH formation by reversed A(iNa+-coupled electron transfer in anaerobic K. pneumoniae cells is an essential requirement for the synthesis of cell material from the highly oxidised citrate.

7 9 Vibrio alginolyticus is a falcutative anaerobic marine bacterium that requires Na* ions for growth. Remarkably, this organism can grow at alkaline ph in the presence of the proton ionophore carbonyl Cyanide m-chlorophenylhydrazone (CCCP). A Na*- dependent NADH:ubiquinone oxidoreductase (Na*-NQR) was found as a respiratory chain component. According to previous work, the enzyme is composed of three subunits containing both FMN and FAD as prosthetic groups. Since all purified H*- translocating NADH:ubiquinone oxidoreductases from eukaryotic and prokaryotic organisms are composed of much more subunits and contain FMN and several ironsulfur Clusters as prosthetic groups, the subunit compositum and the prosthetic groups present in this enzyme have been re-examined. In this work, a new purification procedura for the Na*-NQR of V. alginolyticus with the detergent lauryldimethylamine N-oxide (LDAO) was established. Four dominant bands and a number of minor bands were visible on SDS-PAGE of the purified preparation. Three of the four dominant Polypeptides have been identified as gene products of the nqr Operon which encodes Na+-NQR. The purified enzyme showed both NADH dehydrogenase and quinone reductase activities. The two activities were effectively inhibited by Ag+ ions (K, 0.2 im). The quinone reductase activity was inhibited by HQNO (K, 0.4 JIM) and was activated by Na* ions (K 10 mm). Chemical and spectroscopic analyses have revealed that the purified enzyme contained a FAD and a [2Fe-2S] type iron-sulfur Cluster as the prosthetic groups. Functions of the prosthetic groups have been studied and an hypothetical model of electron transfer has been proposed. Purified NADH:ubiquinone oxidoreductase was incorporated into liposomes by detergent dilution with octyl glucoside and Na+ transport into the derived proteoliposomes was investigated. Under proper conditions, the proteoliposomes catalysed Na* uptake in the presence of NADH and ubiquinone-1 with an apparent stoichiometry of 0.5 Na+ per NADH. The transport of Na* ions was inhibited by HQNO or monensin and stimulated by CCCP or valinomycin. These data provided firm proof that the Na*-NQR isolated from V. alginolyticus is a primary Na* pump. An indirect Na*

8 10 transport mechanism by a H^translocating NADH:ubiquinone oxidoreductase in combination with a Na+/H* antiporter has therefore been excluded.
