1 Instructions for Use Parainfluenza 1/2/3 IgG ELISA Enzyme immunoassays (microtiter strips) for the qualitative and quantitative determination of IgG antibodies against Parainfluenza virus 1,2 and 3 in human serum and plasma. 2 ENGLISH 1. INTENDED USE Enzyme immunassay for the in-vitro diagnostic qualitative and quantitative determination of IgG antibodies against the parainfluenza viruses 1, 2 and 3 in human serum. 2. SUMMARY AND EXPLANATION The infection with parainfluenza viruses is an air-borne infection and appears endemically worldwide. The seroprevalence of parainfluenza in infants in their first year of life is 50%. Typical for parainfluenza viruses are frequent reinfections, characteristically for parainfluenza 3. Incubation time is 2-6 days. The parainfluenza viruses are a subgroup of the paramyxoviruses. Together with the respiratory syncytial viruses (RS viruses), the pathogens belong to the major viral pathogens of diseases of the respiratory tract, accompanied by severe clinical symptoms. In adults, parainfluenza virus causes a feverish rhinitis and laryngitis. First signs are sudden headaches, pain in muscles and joints, followed by fever of C. If the lower respiratory tract is involved, additionally trachyphonea and dry cough develops as a sign of tracheobronchitis. Parainfluenza 1 causes severe pneumonias in newborns, manifested by high fever, cyanosis, dyspnoea and bloody purulent sputum. Sometimes, meningitis symptoms occur at the same time. Parainfluenza 2 very often causes an acute laryngotracheobronchitis with pseudocroup in infants and children. First signs of the infection are catarrhal symptoms followed by trachyphoena, dry barking cough and inspiratory stridor. Parainfluenza 3 viruses are considered as the major pathogens of pneumonia and bronchiolitis. While types 1, 2 and 3 are distributed worldwide, parainfluenza type 4 appears only in the USA. Infections 1 and 3 occur all the year, while parainfluenza 2 and 4 viruses appear only sporadically. Laboratory diagnosis of parainfluenza viruses is done using haemagglutination-inhibiting test (HIT) complement binding reaction (CF) and neutralisation test (NT). Newer methods are IFA and ELISA, which allow to identify IgG and IgA antibodies in the patient serum. In differential diagnosis, tests for other paramyxoviruses like mumps, shipping fever viruses and simianvirus type 5 have to be performed due to possible cross-reactions. 3. TEST PRINCIPLE Solid phase enzyme-linked immunosorbent assay (ELISA) based on the sandwich principle. The wells are coated with antigen. Specific antibodies of the sample binding to the antigen coated wells are detected by a secondary enzyme conjugated antibody (E-Ab) specific for human IgG. After the substrate reaction the intensity of the color developed is proportional to the amount of IgG-specific antibodies detected. Results of samples can be determined directly using the standard curve. 4. WARNINGS AND PRECAUTIONS 1. For in-vitro diagnostic use only. For professional use only. 2. Before starting the assay, read the instructions completely and carefully. Use the valid version of the package insert provided with the kit. Be sure that everything is understood. For further information (clinical background, test performance, automation protocols, alternative applications, literature, etc.) please refer to the IBL-Homepage. 3. In case of severe damage of the kit package please contact IBL or your supplier in written form, latest one week after receiving the kit. Do not use damaged components in test runs, but keep safe for complaint related issues. 4. Obey lot number and expiry date. Do not mix reagents of different lots. Do not use expired reagents. 5. Follow good laboratory practice and safety guidelines. Wear lab coats, disposable latex gloves and protective glasses where necessary. 6. Reagents of this kit containing hazardous material may cause eye and skin irritations. See MATERIALS SUPPLIED and labels for details. Chemicals and prepared or used reagents have to be treated as hazardous waste according to national biohazard and safety guidelines or regulations. 8. Avoid contact with Stop solution. It may cause skin irritations and burns. 9. Some reagents contain sodium azide (NaN 3 ) as preservatives. In case of contact with eyes or skin, flush immediately with water. NaN 3 may react with lead and copper plumbing to form explosive metal azides. When disposing reagents, flush with a large volume of water to avoid azide build-up. Version 1 / / 5
3 ENGLISH 10. All reagents of this kit containing human serum or plasma have been tested and were found negative for HIV I/II, HBsAg and HCV. However, a presence of these or other infectious agents cannot be excluded absolutely and therefore reagents should be treated as potential biohazards in use and for disposal. 5. STORAGE AND STABILITY The kit is shipped at ambient temperature and should be stored at 2-8 C. Keep away from heat or direct sun light. The storage and stability of specimen and prepared reagents is stated in the corresponding chapters. The microtiter strips are stable up to the expiry date of the kit in the broken, but tightly closed bag when stored at 2 8 C. 6. SPECIMEN COLLECTION AND STORAGE Serum, Plasma (EDTA, Heparin) The usual precautions for venipuncture should be observed. It is important to preserve the chemical integrity of a blood specimen from the moment it is collected until it is assayed. Do not use grossly hemolytic, icteric or grossly lipemic specimens. Samples appearing turbid should be centrifuged before testing to remove any particulate material. Storage: 2-8 C -20 C Stability: 2 d > 2 d 7. 4 ENGLISH 9. PROCEDURE NOTES 1. Any improper handling of samples or modification of the test procedure may influence the results. The indicated pipetting volumes, incubation times, temperatures and pretreatment steps have to be performed strictly according to the instructions. Use calibrated pipettes and devices only. 2. Once the test has been started, all steps should be completed without interruption. Make sure that required reagents, materials and devices are prepared ready at the appropriate time. Allow all reagents and specimens to reach room temperature (18-25 C) and gently swirl each vial of liquid reagent and sample before use. Mix reagents without foaming. 3. Avoid contamination of reagents, pipettes and wells/tubes. Use new disposable plastic pipette tips for each reagent, standard or specimen. Do not interchange caps. Always cap not used vials. Do not reuse wells/tubes or reagents. 4. Use a pipetting scheme to verify an appropriate plate layout. 5. Incubation time affects results. All wells should be handled in the same order and time sequences. It is recommended to use an 8-channel Micropipettor for pipetting of solutions in all wells. 6. Microplate washing is important. Improperly washed wells will give erroneous results. It is recommended to use a multichannel pipette or an automatic microplate washing system. Do not allow the wells to dry between incubations. Do not scratch coated wells during rinsing and aspiration. Rinse and fill all reagents with care. While rinsing, check that all wells are filled precisely with Wash Buffer, and that there are no residues in the wells. 7. Humidity affects the coated wells/tubes. Do not open the pouch until it reaches room temperature. Unused wells/tubes should be returned immediately to the resealed pouch including the desiccant. 10. PRE-TEST SETUP INSTRUCTIONS Preparation of Components Dilute/ Component dissolve 60 ml Wash Buffer ad 600 ml Diluent Relation bidist. TEST PROCEDURE Pipette 100 µl of each Standard and diluted sample into the respective wells of the Microtiter Plate. In the qualitative test only Standard B is used. Cover plate with adhesive foil. Incubate 60 min at C. Remove adhesive foil. Discard incubation solution. Wash plate 3 x with 300 µl of diluted Wash Buffer. Remove excess solution by tapping the inverted plate on a paper towel. Pipette 100 µl of Enzyme Conjugate into each well. Cover plate with new adhesive foil. Incubate 30 min at C. Remove adhesive foil. Discard incubation solution. Wash plate 3 x with 300 µl of diluted Wash Buffer. Remove excess solution by tapping the inverted plate on a paper towel. For adding of Substrate and Stop Solution use, if available, an 8-channel Micropipettor. Pipetting should be carried out in the same time intervals for Substrate and Stop Solution. Use positive displacement and avoid formation of air bubbles. Pipette 100 µl of TMB Substrate Solution into each well. Incubate 20 min at C in the dark (without adhesive foil). Stop the substrate reaction by adding 100 µl of TMB Stop Solution into each well. Briefly mix contents by gently shaking the plate. Color changes from blue to yellow. Version 1 / / 5
5 ENGLISH Measure optical density with a photometer at 450 nm (Reference-wavelength: nm) within 60 min after pipetting of the Stop Solution. 12. QUALITY CONTROL The test results are only valid if the test has been performed following the instructions. Moreover the user must strictly adhere to the rules of GLP (Good Laboratory Practice) or other applicable standards/laws. All standards must be found within the acceptable ranges as stated on the QC Certificate. If the criteria are not met, the run is not valid and should be repeated. Each laboratory should use known samples as further controls. In case of any deviation the following technical issues should be proven: Expiration dates of (prepared) reagents, storage conditions, pipettes, devices, incubation conditions and washing methods. It is recommended to participate at appropriate quality assessment trials. 13. CALCULATION OF RESULTS The obtained OD of the standards (y-axis, linear) are plotted against their concentration (x-axis, logarithmic) either on semi-logarithmic graph paper or using an automated method. A good fit is provided with cubic spline, 4 Parameter Logisitcs or Logit-Log. For the calculation of the standard curve, apply each signal of the standards (one obvious outlier of duplicates might be omitted and the more plausible single value might be used). The concentration of the samples can be read from the standard curve. The initial dilution has been taken into consideration when reading the results from the graph. Results of samples of higher predilution have to be multiplied with the dilution factor. Samples showing concentrations above the highest standard have to be diluted as described in PRE-TEST SETUP INSTRUCTIONS and reassayed. Typical Calibration Curve OD 450 nm (Example. They have to be correlated to other 8-12 equivocal clinical observations and diagnostic tests. > 12 positive 15. LIMITATIONS OF THE PROCEDURE Specimen collection has a significant effect on the test results. See SPECIMEN COLLECTION AND STORAGE for details. For cross-reactivities, see PERFORMANCE. Azide and thimerosal at concentrations > 0.1 % interfere in this assay and may lead to false results. 7 DEUTSCH 1. ZWECKBESTIMMUNG Enzymimmunoassay zur qualitativen und quantitativen Bestimmung in humanem Serum und Plasma. 2. KLINISCHE BEDEUTUNG Die Infektion mit Parainfluenza-Viren erfolgt durch Tröpfcheninfektion von Mensch zu Mensch. Als Virusreservoir können hierbei verschiedene Tierarten dienen. Parainfluenza-Viren sind endemisch weltweit verbreitet. Die Durchseuchung mit Parainfluenza erfolgt bei etwa 50% der Kinder im 1. Lebensjahr. Parainfluenza-Viren zeichnen sich durch häufige Reinfektionen aus. Dies gilt vor allem für Parainfluenza-3- Viren. Die Inkubationszeit beträgt 2-6 Tage. Die Parainfluenza-Viren sind Untergruppen der Paramyxoviren und haben eine Größe von etwa nm. Sie sind empfindlich gegen Äther, agglutinieren Menschen- oder Hühnererythrozyten und besitzen ein Rezeptor zerstörendes Enzym, wie es auch bei den Influenza-Viren bekannt ist. Sie lassen sich gut in Affen- Zellkulturen oder menschlichen Epithelzellkulturen vermehren, dagegen schlecht im bebrüteten Hühnerei. Man unterscheidet die Parainfuenzaviren 1, 2, 3 und 4. Die Erreger zählen mit dem Respiratory Syncytial Virus zu den viralen Haupterregern der Respirationstrakt- Erkrankungen bei Säuglingen, Kindern und älteren Menschen. Die Primärinfektion erfolgt meist bereits im Säuglingsalter als hochfebriler Infekt der Atemwege, der mit einer schweren klinischen Symptomatik einhergeht. bei Erwachsenen ruft das Parainfluenza-Virus eine fieberhafte Rhinitis und Laryngitis hervor. Erste Anzeichen sind plötzliche Kopf-, Glieder und Muskelschmerzen, gefolgt von Körpertemperaturen zwischen C. Bei einer Beteiligung der unteren Luftwege treten zusätzlich Heiserkeit und trockener Husten als Zeichen einer Tracheobronchitis auf. Parainfluenza 1 führt bei Neugeborenen zu schweren Pneumonien, die durch hohes Fieber, Zyanose, Dyspnoe und blutrig-eitriges Sputum gekennzeichnet sind. Gelegentlich treten gleichzeitig meningitische Symptome auf. Parainfluenza 2 ruft bei Säuglingen und Kindern sehr oft eine Laryngotracheo-Bronchitis mit Pseudokrupp hervor. Die Erkrankung beginnt mit katarrhalischen Erscheinungen, gefolgt von Heiserkeit, trockenem bellendem Husten und inspiratorischem Stridor. Parainfluenza 3-Viren zählen zu den wichtigsten Erregern einer Pneumonie und Bronchiolitis. Während die Typen 1, 2 und 3 weltweit verbreitet sind, tritt Parainfluenza Typ 4 nur in den USA auf. Die Infektionen 1 und 3 beobachtet man das ganze Jahr über, während Parainfluenza 2-Viren nur sporadisch verbreitet sind. Die Labordiagnose von Parainfluenzaviren kann nur durch den HHT, die KBR und den NT erfolgen. Neuere Methoden sind die IFA und der ELISA, mit deren Hilfe es gelingt, die Antikörper im Patientenserum als IgG oder IgA zu identifizieren. Zur Differentialdiagnose müssen wegen möglicher Kreuzreaktionen Untersuchungen auf andere Paramyxoviren wie Mumps, Shipping fever Viren und Simianvirus Typ 5 durchgeführt werden. 3. TESTPRINZIP Sandwich-Enzymimmunoassay (ELISA). Die Wells der Mikrotiterstreifen sind mit Antigen beschichtet. Die spezifischen Antikörper der Probe binden an die Antigen-beschichteten Wells und werden von einem zweiten enzymmarkierten Antikörper (E-Ab), der gegen humanes IgG gerichtet ist, detektiert. Die Intensität der gebildeten Farbe nach der Substratreaktion ist proportional zur IgG-Konzentration. Die Ergebnisse der Proben können direkt anhand der Standardkurve bestimmt werden. 4. WARNHINWEISE UND VORSICHTSMASSNAHMEN 1. Nur zum In-vitro-Gebrauch. Nur für den Gebrauch durch Fachpersonal. 2. Vor der Testdurchführung sollte die Arbeitsanleitung vollständig und sorgfältig gelesen werden und verstanden worden sein. Die gültige Version aus dem Kit verwenden. Weitere Informationen (klinischer Hintergrund, Testcharakteristika, Automatisierung, alternative Anwendungen, Literatur, etc.) finden Sie auf der IBL-Homepage. 3. Im Falle einer erheblichen Beschädigung der Testpackung ist IBL bzw. der jeweilige Lieferant innerhalb einer Woche nach Empfang der Ware schriftlich zu benachrichtigen. Beschädigte Komponenten dürfen nicht zur Testdurchführung verwendet werden, sondern sollten solange aufbewahrt werden, bis der Transportschaden endgültig geregelt ist. 4. Chargen-Nummer und Verfallsdatum beachten. Es dürfen keine Reagenzien aus unterschiedlichen Chargen in einem Test verwendet werden. Verfallene Reagenzien dürfen nicht verwendet werden. 5. Gute Laborpraxis und Sicherheitsrichtlinien beachten. Je nach Bedarf sollten Laborkittel, Einmal- Latexhandschuhe und Schutzbrillen getragen werden. Version 1 / / 5
8 DEUTSCH 6. Reagenzien dieses Kits, die Gefahrstoffe enthalten, können Reizungen der Augen und der Haut hervorrufen. Siehe Angaben in KOMPONENTEN DES KITS und auf den Etiketten. Sicherheitsdatenblätter für dieses Produkt sind auf der IBL-Homepage zum Download verfügbar oder auf Anfrage direkt von IBL erhältlich. 7. Chemikalien und vorbereitete oder gebrauchte Reagenzien sind unter Beachtung der jeweiligen nationalen Bestimmungen als Gefahrstoffabfall zu entsorgen. 8. Kontakt mit Stopplösung vermeiden. Kann Hautreizungen und Verätzungen hervorrufen. 9. Einige Reagenzien enthalten Natriumazid (NaN 3 ) als Konservierungsmittel. Bei Kontakt mit den Augen oder der Haut gründlich mit Wasser abspülen. NaN 3 kann mit Blei und Kupfer explosive Azide bilden. Bei Entsorgung über das Abwasser gut spülen, um Azidanreicherung zu vermeiden. 10. Alle Reagenzien dieses Kits, die humanes Serum oder Plasma enthalten, ergaben bei der Prüfung auf HCV, HBsAg bzw. Antikörper gegen HIV I/II-Virus ein negatives Ergebnis. Trotzdem kann das Vorhandensein solcher infektiöser Erreger nicht mit absoluter Sicherheit ausgeschlossen werden. Die Reagenzien sollten deshalb wie potentiell infektiöses Material behandelt werden. 5. LAGERUNG UND HALTBARKEIT Der Kit wird bei Umgebungstemperatur angeliefert und sollte bei 2-8 C gelagert werden. Vor Hitze und direkter Sonneneinstrahlung schützen. Hinweise zur Lagerung und Haltbarkeit der Proben und vorbereiteten Reagenzien sind den entsprechenden Kapiteln zu entnehmen. Die Mikrotiterplatte ist auch nach dem Öffnen der Verpackung bis zum Verfallsdatum des Kits haltbar, wenn der Beutel sorgfältig wieder verschlossen und bei 2-8 C gelagert wird. 6. PROBENGEWINNUNG UND -AUFBEWAHRUNG Serum, Plasma (EDTA, Heparin) Die üblichen Vorsichtsmaßnahmen bei der Blutabnahme sind einzuhalten. Die chemische Integrität der Blutproben muss vom Zeitpunkt der Blutabnahme bis zur Testdurchführung erhalten bleiben. Keine hämolytischen, ikterischen oder lipämischen Proben verwenden. Getrübte Proben sollten vor der Testdurchführung zentrifugiert werden, um Partikel zu entfernen. Lagerung: 2-8 C -20 C Haltbarkeit: 2 d > 2 d 7. Fehler bei der Handhabung der Proben oder Abweichungen von der beschriebenen Testdurchführung können die Ergebnisse verfälschen. Die angegebenen Pipettiervolumina, Inkubationszeiten, Temperaturen und Vorbereitungsschritte sind unbedingt gemäß Arbeitsanleitung einzuhalten. Nur kalibrierte Pipetten und Geräte verwenden. 2. Sobald mit der Testdurchführung begonnen wird, sollten alle Arbeitsschritte ohne Unterbrechung durchgeführt werden. Es ist sicherzustellen, dass alle benötigten Reagenzien, Geräte und Hilfsmittel zur rechten Zeit zur Verfügung stehen. Alle Reagenzien und Proben müssen auf Raumtemperatur (18-25 C) gebracht und vor Gebrauch vorsichtig ohne Schaumbildung gemischt werden. 3. Kontaminationen der Reagenzien, Pipetten und Wells/Röhrchen sind zu vermeiden. Neue Einmal- Pipettenspitzen für jedes Reagenz, jeden Standard und jede Probe verwenden. Die Deckel der Fläschchen nicht vertauschen. Nicht benötigte Fläschchen immer verschlossen halten. Wells/Röhrchen oder Reagenzien dürfen nicht wiederverwendet werden. 4. Es sollte ein Pipettierschema verwendet werden um die Identifikation der Standards und Proben auf der Platte sicherzustellen. 5. Die Inkubationszeiten beeinflussen die Ergebnisse. Bei jedem Pipettierschritt sollten alle Wells in der gleichen Reihenfolge und im gleichen Zeittakt behandelt werden. Die Verwendung einer 8-Kanal- Mikropipette zum Pipettieren in alle Wells wird empfohlen. 6. Die korrekte Durchführung der Waschschritte ist entscheidend. Ungenügend gewaschene Wells ergeben falsche Ergebnisse. Die Verwendung einer Multikanalpipette oder eines automatischen Waschsystems für Mikrotiterplatten wird empfohlen. Zwischen den Inkubationen die Wells nicht austrocknen lassen. Beim Waschen und Ausschütteln dürfen die beschichteten Wells nicht beschädigt werden. Alle Reagenzien müssen daher mit Vorsicht pipettiert werden. Beim Waschvorgang ist es wichtig, dass alle Wells vollständig und gleichmäßig mit Waschpuffer gefüllt werden und nach dem Ausschütteln kein Rückstand an Flüssigkeit zurückbleibt. 7. Feuchtigkeit beeinflusst die beschichteten Wells/Röhrchen. Verpackung nicht öffnen bevor Raumtemperatur erreicht ist. Nicht benötigte Wells/Röhrchen sofort in den wiederverschließbaren Beutel mit Trockenmittel zurückgeben. 10. TESTVORBEREITUNGEN Vorbereitung der Komponenten Verd./ rekonst. Komponente 60 ml Waschpuffer ad 600 ml Diluent bidest. Wasser 1:10 Bemerkungen Auf 37 C erwärmen, um Kristalle aufzulösen. 10 DEUTSCH 11. TESTDURCHFÜHRUNG Je 100 µl von jedem Standard und jeder verdünnten Probe in die entsprechenden Wells der Mikrotiterplatte pipettieren. Im qualitativen Assay wird nur Standard B verwendet. Platte mit Haftklebefolie abdecken. 60 min bei C inkubieren. Folie entfernen. Inkubationslösung verwerfen. Platte 3 x mit je 300 µl verdünntem Waschpuffer waschen. Restliche Flüssigkeit auf Papiertüchern ausklopfen. 100 µl Enzymkonjugat in jedes Well pipettieren. Platte mit neuer Folie abdecken. 30 min bei C inkubieren. Folie entfernen. Inkubationslösung verwerfen. Platte 3 x mit je 300 µl verdünntem Waschpuffer waschen. Restliche Flüssigkeit auf Papiertüchern ausklopfen. Substrat- und Stopplösung möglichst mit einer 8-Kanal-Pipette pipettieren und Substrat- und Stopplösung in denselben Zeitintervallen zugeben. Mit positivem Vorhub pipettieren, um die Bildung von Luftbläschen zu vermeiden. 100 µl TMB Substratlösung in jedes Well pipettieren. 20 min bei RT (18-25 C) im Dunkeln inkubieren. (Ohne Haftklebefolie.) Die Substratreaktion durch Zugabe von 100 µl TMB Stopplösung in jedes Well stoppen. Platte kurz schütteln. Die Farbe wechselt von blau nach gelb. Die optische Dichte mit einem Photometer innerhalb von 60 min nach Zugabe der Stopplösung bei 450 nm messen (Referenzwellenlänge: nm). 12. QUALITÄTSKONTROLLE Die Testergebnisse sind nur gültig, wenn der Rest gemäß der vorliegenden Arbeitsanleitung abgearbeitet wurde. Ferner muss der Anwender die GLP- Regeln (Good Laboratory Practice) und andere einschlägige Normen und Gesetze beachten. Alle Standards des Kits müssen innerhalb der Akzeptanzbereiche, die auf dem QC-Zertifikat angegeben sind, gefunden werden. Wenn die Kriterien nicht erfüllt sind, sind die Ergebnisse ungültig und der Test sollte wiederholt werden. Jedes Labor sollte darüber hinaus eigene bekannte Proben als weitere Kontrollen mitführen. Es wird empfohlen, an den einschlägigen Ringversuchen teilzunehmen. Bei Abweichungen sind die folgenden Fehlermöglichkeiten zu überprüfen: Haltbarkeit der (vorbereiteten) Reagenzien, Lagerungsbedingungen, Pipetten, Geräte und Hilfsmittel, Inkubationsbedingungen und Waschmethoden. 13. TESTAUSWERTUNG Die erhaltenen OD der Standards (y-achse, linear) gegen deren Konzentration (x-achse, logarithmisch) auftragen, entweder auf semi-logarithmischem Papier oder durch ein entsprechendes Computerprogramm. Bei Verwendung eines Computerprogramms werden die Cubic-Spline-Methode, 4-Parameter-Analyse (linlog) oder Logit-Log-Berechnung empfohlen. Zur Berechnung der Standardkurve sollten alle Werte der Standards verwendet werden (bei Doppelwerten kann ein offensichtlicher Ausreißerwert eliminiert und stattdessen der plausiblere Einzelwert verwendet werden). Die Konzentrationen der Proben können von der Standardkurve abgelesen werden. Die standardmäßig durchzuführende Verdünnung ist in dem oben beschriebenen Auswerteverfahren bereits berücksichtigt. Wenn Proben anders oder weiter verdünnt wurden, müssen die Ergebnisse mit dem entsprechenden Verdünnungsfaktor multipliziert werden. Proben, die oberhalb des höchsten Standards gemessen werden, müssen wie in TESTVORBEREITUNGEN beschrieben verdünnt und erneut analysiert werden. Typische Standardkurve OD 450 nm (Beispiel. NORMWERTE In einer eigenen Studie zeigten offensichtlich gesunde Probanden die folgenden Ergebnisse: Interpretation Ig Isotyp n positiv grenzwertig negativ IgG % 3.8 % 0 % 16. GRENZEN DES VERFAHRENS Die korrekte Durchführung der Probengewinnung ist entscheidend für die Testergebnisse. Näheres siehe PROBENGEWINNUNG UND -LAGERUNG. Angaben zu Kreuzreaktivitäten sind im Kapitel TESTCHARAKTERISTIKA zu finden. Azid und Thimerosal in Konzentrationen > 0.1 % stören im Assay und führen evtl. zu falschen Ergebnissen.
